- چکیده: هدف: نظر به این که تغذیه بر تمامی فعل و انفعالات بدن، به ویژه بر ژنوم و نیز بیان ژنها تاثیر گذار است و همه موجودات همواره در حال تغذیه هستند و بدون تغذیه حیات امکانپذیر نیست. لذا هدف این پژوهش بررسی اجمالی نقش تغذیه در بیان ژن، همانندسازیDNA ، جلوگیری از آسیب DNA و ترمیمDNA بود. مواد و روشها: در این مطالعه با استفاده از کلید واژههای سرطان، ریبونوکلئاز، مهارکننده ریبونوکلئازی و اسید ریبونوکلئیک، در پایگاههای اطلاعاتی معتبر Scopus، SID، IranDoc، PubMed، Google Scholar، Web of Science و IranMedex جستجو انجام شد. به منظور انتخاب مستندات مورد استفاده، تمام مقالاتی که به زبان غیرانگلیسی و فارسی چاپ شده بودند، مقالات تکراری، مقالاتی که امکان دستیابی به متن کامل مقاله وجود نداشت و همچنین مقالاتی که به صورت چکیده ارائه شده بودند حذف گردیدند. نهایتا موارد منتخب به منظور تنظیم مقاله حاضر به طور کامل مطالعه و خلاصهسازی شدند. نتایج: تنظیم بیان ژن برای حیات و سلامت ضروری است و به عوامل درون زا و رژیم غذایی و سایر عوامل محیطی که در سطوح مختلف و با مکانیسمهای متعدد عمل می کنند حساس است. پیوندهای مهم و نزدیکی بین رژیم غذایی/تغذیه، وضعیت متابولیک، مسیرهای سیگنالدهی و تنظیم ژن وجود دارد. فناوریهای پساژنومی شامل هیبریداسیون ریزآرایه و توالییابی نسل بعدی امروزه امکان تجزیه و تحلیل با بازدهی و خروجی بالا، در سطح گسترده ژنوم، رونویسی و پویایی آن (حضور و کمیتRNA ها، الگوهای بیانRNA ) را فراهم می کند. علاوه بر این، تغییرات ناشی از رژیم غذایی یا عوامل دیگر در اشغال محل اتصال فاکتورهای نسحهبرداری، ویژگیهای کروماتین، الگوهای جهانی تباهی رونویسی و پروفایل ترجمه نیز امروزه می تواند در سطح ژنوم مورد مطالعه قرار گیرد. شواهد زیادی وجود دارد که تعداد زیادی از ریز مغذیها، از جمله ویتامینها و مواد معدنی به عنوان کوفاکتورهای آنزیمها یا به عنوان بخشی از ساختار پروتئینها (متالوآنزیمها) در ترمیمDNA ، جلوگیری از آسیب اکسیداتیو به DNA و همچنین نگهداری و متیلاسیون DNA مورد نیاز هستند. نقش ریز مغذیها در حفظ ثبات ژنوم به طور گستردهای بررسی شده است. بسیاری از ریز مغذیهای در فرآیندهای مختلف پایداری ژنوم درگیر هستند. همچنین مکانیسمهای مختلفی وجود دارند که به وسیله آنها کمبود ریز مغذی میتواند باعث آسیب بهDNA ، تسریع پیر شدن و ناپایداری کروموزومی شود. نتیجه گیری: ادغام نتایج حاصل از این فناوریهای متنوع در نهایت به درک جامع و گسترده ای از سیستم کنترل بیان ژن و روابط رژیم غذایی-ژن-سلامت در علم تغذیه مدرن کمک میکند.
- چکیده انگلیسی: Objective Since nutrition affects all the interactions of the body, especially the genome as well as the expression of genes, and all organisms are always feeding, and without feeding life is not possible. Therefore, the aim of this study was to review the role of nutrition in gene expression, DNA replication, prevention of DNA damage and DNA repair. Materials and methods In this study, the keywords such as cancer, ribonuclease, ribonuclease inhibitor and RNA were used to search in databases including Scopus, SID, IranDoc, PubMed, Google Scholar, Web of Science and IranMedex. In order to select the documents used, all articles published in non-English and Persian languages, duplicate articles, articles that could not be accessed to the full text, as well as articles that were presented as abstracts were removed. Finally, the selected cases were thoroughly studied and summarized in order to prepare the current review. Results Regulation of gene expression is essential to life and health and is sensitive to endogenous and dietary and other environmental factors that exert their action at multiple levels and by multiple mechanisms. There are important and intimate links between diet/nutrition, metabolic status, signaling pathways, and gene regulation. Postgenomic technologies including microarray hybridization and massive next-generation sequencing (RNA-Seq) enable nowadays highthroughput analysis, at genome-wide level, of the transcriptome and its dynamics (presence and quantity of RNAs, RNA expression patterns). Further, changes induced by dietary or other factors in TF binding site occupancy, chromatin features, global patterns of transcript decay, and translational profiling can also be studied nowadays at a genome-wide level. There is overwhelming evidence that a large number of micronutrients (vitamins and minerals) are required as cofactors for enzymes or as part of the structure of proteins (metalloenzymes) involved in DNA repair, prevention of oxidative damage to DNA, as well as maintenance and methylation of DNA. The role of micronutrients in the maintenance of genome stability has been extensively reviewed. Some of micronutrients involved in various genome stability processes. There are various mechanisms by which micronutrient deficiency could cause DNA damage, accelerate senescence and chromosomal instability. Conclusion Integration of results from these varied technologies will ultimately allow a comprehensive, system-wide understanding of gene expression control and of diet-gene-health relationships in modern nutrition science.

- چکیده: هدف: بز کرکی راینی یکی از مهمترین نژادهای بز در ایران است. این حیوانات هم برای تولید گوشت و هم برای تولید کرک نگهداری میشوند. یکی از اقدامات اساسی در حیوانات اهلی مطالعه ژنها و پروتئینهای مرتبط با صفات اقتصادی و مطالعه آنها در سطح سلولی یا کروموزومی است. یکی از ژنهای مهم لپتین است. لپتین به وسیله بافت چربی سفید تولید میشود و نقش مهمی در تنظیم مصرف غذا، تعادل انرژی، باروری و فعالیتهای ایمنی بازی میکند. هدف این پژوهش مطالعه بیان ژن لپتین در بافتهای چربی، کبد، کلیه، شش و قلب در بز کرکی راینی با استفاده از تکنیک Real Time PCR بود.
مواد و روشها: از بافتهای قلب، شش، کلیه، کبد و بافت چربی (از هر بافت 3 تکرار) از 6 بز کرکی راینی نمونه برداری انجام شد. از این بافتها RNA کل استخراج شد. RNA استخراج شده در دمای منفی80 درجه سانتیگراد نگهداری شد. کیفیت و کمیت RNA بررسی و سنتز cDNAانجام شد. واکنش Real Time PCR برای ژن لپتین و بتااکتین صورت گرفت. محصولات PCR روی ژل آگارز 5/1 درصد الکتروفورز شد. منحنیهای ذوب و سطوح مختلف بیان در بافتهای ذکر شده، مورد تجزیه و تحلیل قرار گرفت.
نتایج: نتایج منحنیهای Real Time PCR و مشاهده نتایج الکتروفورز محصولات PCR روی ژل آگارز نشان داد نشان داد که این ژن در تمامی بافتهای بررسی شده بیان شده است و بیشترین سطح بیان در بافت چربی (5/4 برابر) و کبد (7/3 برابر) و کمترین سطح بیان در بافتهای قلب (4/1 برابر) مشاهده شد.
نتیجهگیری: این نتایج میتواند نشان دهنده این امر باشد که لپتین در متابولیسم چربی نقش ویژهای دارد. مطالعات بیشتری باید انجام شود تا نقش لپتین در فیزیولوژی ساخت و متابولیسم چربی و مواد دیگر مشخص شود. این امر کمک خواهد کرد تا مکانیسمهایی برای شناخت اثر فاکتورهای تغذیهای و چربیهای بدن در فرآیندهای مختلف درک شوند.- چکیده انگلیسی: Objective
Raini Cashmere goat is one of the most important goat breeds in Iran. These animals are breed both for meat production and cashmere production. One of the basic measures in the domestic animals is the study of genes and proteins associated with economic traits and their study at cellular or chromosomal levels. One of these important genes is the leptin. Leptin is produced by white adipose tissue and plays an important role in the regulation of feed intake, energy balance, fertility and immune functions. The aim of this research was to study leptin gene expression in adipose tissue, liver, kidney, lung and heart of Raini Cashmere goat using Real Time PCR technique.
Materials and methods
Tissue sampling from heart, lung, liver, kidney and adipose tissue (3 replicates from each tissue) of 6 animals was performed and RNA was extracted. Extracted RNA were immediately stored at -80°C.The Quality and quantity of RNA were evaluated and cDNA was synthesized and Real Time PCR was performed. PCR Products were electrophoresed on 1.5% agarose gel. Melting curves from Real Time PCR were examined and were evaluated different levels of expression in the studied different tissues.
Results
The results of Real Time PCR curves and observation of electrophoresis of PCR products on agarose gel showed that the leptin gene was expressed in all the tested tissues and the highest level of expression was observed in adipose tissue (4.5) and liver (3.7) and the lowest level was detected in heart (1.4).
Conclusions
These results may show that leptin plays a particular role in fat metabolism. Further studies are needed to clarify role of leptin in the physiology of fat metabolism and other materials. This would help us to better understand the mechanisms for the known effect of nutritional factors and body fatness on various functions.- انتشار مقاله: 24-12-1399

- چکیده: هدف: برآوردها نشان میدهد که در دنیا تقریبا سی میلیون راس بز و در ایران حدودا پنج میلیون راس بز کرکی در حال نگهداری و پرورش است. یکی از با اهمیتترین این نژادها بز کرکی راینی است. هورمونهای استروئیدی از طریق تنظیم رونویسی ژنهای هدف وظایف فیزیولوژیکی خود را انجام میدهند. به طوریکه گیرندههای این هورمونها از طریق شبکههای رونویسی در پوکی استخوان، التهاب و سرطان نقش ایفا میکنند. گیرنده استروژن (ER) یک فاکتور رونویسی است که واسطه عملکرد هورمون استروژن در بسیاری از فرآیندهای فیزیولوژیکی و آسیب شناختی است. دو گیرنده استروژن وجود دارد، ESR1 و ESR2که در اندامهای تولید مثلی واقع شده اند. میزان تولید مثل، به ویژه باروری، یکی از مهمترین صفات اقتصادی در تولید حیوانات است و توسط عوامل ژنتیکی و محیطی تنظیم میشود. لذا، هدف این پژوهش بررسی پروفایل بیانی ژن ESR2 در بافتهای مختلف بز کرکی راینی با استفاده از Real Time PCR بود.
مواد و روشها: نمونه گیری از بافتهای بیضه، تخمدان، رحم، پستان، مغز، کلیه و قلب یک راس بز نر و یک راس بز ماده در کشتارگاه انجام شد (از هر بافت 3 تکرار). از بافتهای مورد نظر استخراج RNA کل انجام و کیفیت و کمیت آن ارزیابی شد. سپس cDNA ساخته و Real Time PCR اجرا شد. همچنین از ژن خانه دار GAPDH به عنوان کنترل استفاده شد. منحنیهای ذوب بررسی و تجزیه و تحلیل دادههای حاصل از Real Time PCR انجام شد.
نتایج: نتایج منحنیهای Real Time PCR و مشاهده نتایج الکتروفورز محصولات PCR روی ژل آگارز 2 درصد نشان داد که ژن ESR2 در بافتهای بیضه، تخمدان، رحم، پستان، مغز، کلیه و قلب بیان شده است. بیشترین میزان بیان در تخمدان و سپس مغز و رحم مشاهده شد و کمترین سطح بیان مربوط به بافتهای کلیه، قلب و بیضه بود.
نتیجهگیری: با توجه به نتایج پژوهش حاضر و نتایج سایر محققین میتوان نتیجه گرفت که ESR2 نقش مهمی در باروری مادهها دارد، در پژوهش حاضر مشخص شد ESR2در بافت تخمدان نسبت به بافتهای دیگر بسیار بیشتر بیان میشود. لذا، ESR2 به احتمال زیاد برای باروری مادهها بسیار ضروری هستند و نتایج این پژوهش را برای پژوهشهای آینده جهت توصیف نقش ژن ESR2 به عنوان یک ژن کاندیدا برای باروری بهتر و فیزیولوژی طبیعی در حیوانات اهلی، به ویژه بز فراهم کرده است.- چکیده انگلیسی: Objective
Estimates show that in the world about 30 million goats and in Iran about five million Cashmere goat are bred. One of the most important of these breeds is the Raini Cashmere goat. Steroid hormones perform their physiological functions by regulating the transcription of target genes. Receptors for these hormones play a role in osteoporosis, inflammation and cancer through transcription networks. Estrogen receptor (ER) is a transcription factor that mediates the function of estrogen in many physiological and pathological processes. There are two estrogen receptors, ESR1 and ESR2, which are located in the reproductive organs. Reproduction rate, especially fertility, is one of the most important economic traits in animal production and is regulated by genetic and environmental factors. Therefore, the aim of this study was to investigate the expression profile of ESR2 gene in different tissues of fluffy goats using Real Time PCR.
Materials and methods
Sampling of testis, ovarian, uterine, breast, brain, kidney and heart tissues was performed from a male and a female goat (3 replicates from each tissue). Total RNA was extracted from the tissues and the quality and quantity of the extracted RNA were evaluated. The cDNA was then synthesized and Real Time PCR was performed. The GAPDH housekeeping gene was used as a control. Melting curves were examined and data obtained from Real Time PCR were analyzed.
Results
The results of Real Time PCR curves and observation of electrophoresis of PCR products on 2% agarose gel showed that ESR2 gene was expressed in testis, ovary, uterine, breast, brain, kidney, and heart tissues. The highest level of expression was observed in the ovary and then the brain and uterus and the lowest level of expression was related to kidney, heart and testis tissues.
Conclusions
According to the results of the present research and the results of other studies, it can be concluded that ESR2 plays an important role in female fertility, because in the present study it was found that ESR2 is much more expressed in ovarian tissue than other tissues. Therefore, ESR2 is likely to be essential for female fertility, and the results of this study provide the basis for future research to describe the role of the ESR2 gene as a candidate gene for better fertility and normal physiology in domestic animals, especially goats.- انتشار مقاله: 21-08-1399

Extracted RNA were immediately stored at -80°C.The Quality and quantity of RNA were evaluated and cDNA was synthesized and Real Time PCR was performed. PCR Products were electrophoresed on 1.5% agarose gel. Melting curves from Real Time PCR were examined and were evaluated different levels of expression in the studied different tissues.
Results
The results of Real Time PCR curves and observation of electrophoresis of PCR products on agarose gel showed that the leptin gene was expressed in all the tested tissues and the highest level of expression was observed in adipose tissue (4.5) and liver (3.7) and the lowest level was detected in heart (1.4).
Conclusions
These results may show that leptin plays a particular role in fat metabolism. Further studies are needed to clarify role of leptin in the physiology of fat metabolism and other materials. This would help us to better understand the mechanisms for the known effect of nutritional factors and body fatness on various functions.- انتشار مقاله: 24-12-1399

- چکیده: هدف: سه ژن باروری BMPR1B (یا ALK6 معروف به FecB روی کروموزوم 6)، GDF9 (معروف به FecG روی کروموزوم 5) و BMP15 (معروف به FecX روی کروموزوم X) شناسایی شده است. ژن های پروتئین مورفوژنتیک استخوان (BMP) اعضای فوق خانواده فاکتور رشد تبدیل کننده بتا (TGF-β) هستند که سیتوکین های چند منظوره با عملکرد 2 برابر هستند و در انواع سلولها بیان میشوند. BMP ها در اصل بر اساس توانایی آنها در تولید استخوان در جای غیر معمول هنگام کاشت در داخل بافت نرم در داخل بدن شناسایی میشوند. آنها همچنین در رشد و توسعه جنین، هموستازی، ترمیم الگوسازی بافتهای مختلف، تمایز سلولی و آپوپتوز نقش دارند. ژن BMP15 با افزایش میتوز در سلول گرانولوزا، سرکوب بیان گیرنده هورمون محرک فولیکول و تحریک بیان لیگاند کیت، تکثیر و تمایز سلول گرانولوز را تنظیم میکند. از این رو، این ژن نقشی اساسی در باروری پستانداران ماده دارد. هدف این مطالعه پروفایل بیانی mRNA مختص بافت ژن BMP15 در بز کرکی راینی با استفاده از Real Time PCR بود. مواد و روشها: نمونههای بافتی (از هر بافت 3 تکرار) از تخمدان، رحم، شاخ رحم، اویداکت، ماهیچه اسکلتی، چربی زیرپوستی، قلب، جگر، شش، کلیه، طحال و غده فوق کلیه در هنگام کشتار در کشتارگاه از دو راس بز تهیه شد. استخراج RNA کل و سنتز cDNA انجام شد شد. میزان نسبی بیان ژن BMP15 با استفاده از واکنش Real Time PCR به روش SYBR Green به دست آمد. ژن GAPDH به عنوان ژن کنترل داخلی استفاده شد. برای تجزیه و تحلیل دادههای حاصل از Real Time PCR از روش پیفافل استفاده شد. نتایج: بیان ژن BMP15 در بافتهای تخمدان، رحم، شاخ رحم، اویداکت، ماهیچه اسکلتی، چربی زیرپوستی، قلب، جگر، شش، کلیه، طحال و غده فوق کلیه بز کرکی راینی با استفاده از روش PCR در زمان واقعی نشان داد که این ژن فقط در تخمدان بیان میشود و در بافتهای مطالعه شده دیگر بیان نمیشود. نتیجهگیری: با توجه به نتایج پژوهش حاضر و نتایج سایر محققین میتوان نتیجه گرفت که ژن BMP15 نقش مهمی در تخمدان و در نتیجه در باروری دارد، چرا که در پژوهش حاضر مشخص شد که ژن BMP15 فقط در بافت تخمدان بیان میشود. لذا، ژن BMP15 به احتمال زیاد برای باروری مادهها بسیار ضروری است و نتایج این پژوهش اساسی را برای پژوهشهای آینده جهت توصیف نقش ژن BMP15 به عنوان یک ژن کاندیدا برای باروری بهتر و فیزیولوژی طبیعی در حیوانات اهلی، به ویژه بز فراهم کرده است.
- چکیده انگلیسی: Objective Three fecundity genes have been identified, namely bone morphogenetic protein receptor type 1B (BMPR1B; or activin-like kinase 6, ALK6) known as FecB on chromosome 6, growth differentiation factor 9 (GDF9) known as FecG on chromosome 5, and bone morphogenetic protein 15 (BMP15) known as FecX on chromosome X. Bone morphogenetic protein (BMP) genes are members of the transforming growth factor-beta (TGF-β) super-family which are multifunctional cytokines with a 2-fold function and are expressed in a variety of cells. BMPs were originally identified based on their ability to produce ectopic bone formation when implanted within the soft tissue in vivo. They also play roles in embryonic development, homeostasis, repairing of various tissue patterning, cell differentiation, and apoptosis. The BMP15 gene regulates granulose cell proliferation and differentiation by promoting granulosa cell mitosis, suppressing follicle-stimulating hormone receptor expression, and stimulating the kit ligand expression; hence, this gene plays a pivotal role in female fertility in mammals. The aim of this study was to investigate tissue-specific mRNA expression profile of BMP15 gene in Raini cashmere goat using Real Time PCR. Materials and Methods Tissue samples (3 repetitions of each tissue) were taken from ovary, uterus, uterine horn, oviduct, skeletal muscle, subcutaneous fat, heart, liver, lung, kidney, spleen and adrenal during slaughter at the slaughterhouse from two goats. RNA extraction from tissues and cDNA synthesis were performed. Relative gene expression of BMP15 was done applying Real Time PCR using SYBR Green method. GAPDH gene was used as housekeeping gene. Pfaffl method was used to analyze achieved data. Results BMP15 gene expression in ovary, uterus, uterine horn, oviduct, skeletal muscle, subcutaneous fat, heart, liver, lung, kidney, spleen and adrenal tissues using Real Time PCR in studied goats demonstrated that BMP15 gene is expressed only in the ovary tissue and is not expressed in other studied tissues. Conclusions Based on the results of the present study and the results of other researchers, it can be concluded that BMP15 gene plays an important role in ovary and as a result in fertility, because in the present study it was found that BMP15 is expressed only in ovary tissue. Therefore, BMP15 is likely to be essential for female fertility, and the results of this study provide a basis for future research to describe the role of BMP15 gene as a candidate gene for better fertility and normal physiology in domestic animals, especially goats.

هدف: گیرنده استروژن (ER) یک فاکتور رونویسی است که واسطه عملکرد هورمون استروژن در بسیاری از فرآیندهای فیزیولوژیکی و آسیب شناختی است. بیان گیرنده استروژن (ESR1)، که گیرنده استروژن آلفا (ER-α) را کد می کند، مختص بافت است. به عنوان مثال، بیان ESR1 در آندومتر و غده پستانی بسیار زیاد است و در جفت و پوست کم است. این ژن روی کروموزوم شماره 6 انسان قرار دارد و به نامهای Era، ESRA، ESTRR، و NR3A1 نیز معروف است. در چندین پژوهش نشان داده شده است که واریانتهای ژنتیکی در این ژن با حساسیت به سرطان پستان همبستگی دارند. هدف این پژوهش بررسی میزان بیان ژن ESR1 در بافتهای مختلف بز کرکی راینی با استفاده از Real Time PCR بود.
مواد و روشها: تعداد یک راس بز نر و یک راس بز ماده برای نمونه برداری از بافتها انتخاب شدند. نمونهها از بافتهای قلب، کلیه، مغز، تخمدان، رحم، پستان و بیضه (از هر بافت 3 تکرار) در هنگام کشتار در کشتارگاه تهیه شد. RNA کل استخراج و cDNA ساخته شد. برای برررسی میزان نسبی بیان ژن ها از واکنش Real Time PCR به روش SYBR Green استفاده شد. در این مطالعه از ژن GAPDH به عنوان ژن کنترل داخلی استفاده شد. برای تجزیه و تحلیل دادههای حاصل از Real Time PCR از روش پیفافل استفاده شد.
نتایج: مطالعه بیان ژن ESR1 در بافتهای قلب، کلیه، مغز، تخمدان، رحم، پستان و بیضه بز کرکی راینی با استفاده از روش PCR در زمان واقعی نشان داد که این ژن در تمامی بافتهای بررسی شده بیان شده است. بیشترین سطح بیان در بافتهای کلیه، تخمدان، رحم و بیضه و کمترین سطح بیان در بافت مغز و قلب مشاهده شد
نتیجهگیری: با توجه به نتایج پژوهش حاضر و نتایج سایر محققین میتوان نتیجه گرفت که گیرندههای استروژن نقش مهمی در اسپرماتوژنز و باروری نرها دارند، چرا که در پژوهش حاضر مشخص شد که ESR1 در بافتهای تولیدمثلی نسبت به بافتهای غیرتولیدمثلی بسیار بیشتر بیان میشود. لذا، استروژن و گیرندههایش به احتمال زیاد برای باروری نرها بسیار ضروری هستند و نتایج این پژوهش اساسی را برای پژوهشهای آینده جهت توصیف نقش ژن ESR1 به عنوان یک ژن کاندیدا برای باروری بهتر و فیزیولوژی طبیعی در حیوانات اهلی، به ویژه بز فراهم کرده است.- چکیده انگلیسی: Abstract
Objective
The estrogen receptor (ER) is a transcription factor that mediates the function of the hormone estrogen in many physiological and pathological processes. The expression of the estrogen receptor (ESR1), which encodes the alpha estrogen receptor (ER-α), is tissue specific. For example, the expression of ESR1 in the endometrium and mammary gland is very high and low in the placenta and skin. The gene is located on human chromosome 6 and is also known as Era, ESRA, ESTRR, and NR3A1. Several studies have shown that genetic variants in this gene are associated with breast cancer sensitivity. The aim of this study was to investigate ESR1gene expression in different tissues of Raini Cashmere goat using Real Time PCR.
Materials and Methods
One male and one female goats were selected for tissue sampling. Samples were taken from tissues of the heart, kidneys, brain, ovaries, uterus, breast and testicles (3 repetitions of each tissue) during slaughter at the slaughterhouse. RNA was extracted and cDNA was synthesized. Real Time PCR was performed using SYBR Green method to study relative gene expression. GAPDH gene was used as housekeeping gene. Pfaffl method was used to analyze achieved data.
Results
The study of ESR1 gene expression in the tissues of the heart, kidney, brain, ovary, uterus, mammary gland and testis of goat using Real Time PCR showed that this gene is expressed in all studied tissues. The highest levels of expression were observed in the kidney, ovary, uterus and testis tissues, and the lowest levels of expression were observed in the brain and heart tissues.
Conclusions
Based on the results of the present study and the results of other researchers, it can be concluded that estrogen receptors play an important role in spermatogenesis and male fertility, because in the present study it was found that ESR1 is much more expressed in reproductive tissues than in non-reproductive tissues. Therefore, estrogen and its receptors are likely to be essential for male fertility, and the results of this study provide a basis for future research to describe the role of ESR1 gene as a candidate gene for better fertility and natural physiology in domestic animals, especially goats.- انتشار مقاله: 09-11-1398

- چکیده: هدف: کالپاستاتین در بافت عضله اسکلتی حیوانات یافت شده است و میتواند با سرکوب فعالیت کالپاین مانع از رشد بی رویه سلولی شود. دخالت کالپاینها در آپوپتوزیز موضوعی است که هنوز مورد بحث است، اگرچه دخالت آنها محدود به انواع خاصی از سلولها و محرکهای خاص است. مطالعات مختلف نشان داده که این ژن در عملکرد رشد و کیفیت گوشت نقش دارد، لذا هدف این پژوهش بررسی میزان بیان ژن کالپاستاتین در بافتهای مختلف بز کرکی راینی با استفاده از Real Time PCR بود. مواد و روشها: در مجموع 21 نمونه بافتی شامل ماهیچه، کلیه، قلب، جگر، شش، طحال و چربی (از هر بافت 3 تکرار) از 3 بز کرکی راینی در هنگام کشتار در کشتارگاه تهیه شد. RNA کل استخراج و cDNA ساخته شد. برای برررسی میزان نسبی بیان ژن ها از واکنش Real Time PCR به روش SYBR Green استفاده شد. در این مطالعه از ژن گلیسرآلدئید 3 فسفات دهیدروژناز (GAPDH) به عنوان ژن کنترل داخلی استفاده شد. برای تجزیه و تحلیل داده های حاصل از Real Time PCR از روش پیفافل استفاده شد. نتایج: برای ژن کالپاستاتین قطعه bp89 و برای GAPDH قطعه bp101 در همه نمونهها مشاهده شد. نتایج Real Time PCR حاصل از این پژوهش نشان داد که ژن کالپاستاتین در تمامی بافتهای بررسی شده (ماهیچه، کلیه، قلب، جگر، شش، طحال و چربی) بیان شده است و بیشترین سطح بیان در بافت قلب، طحال و جگر و کمترین سطح بیان در بافت چربی مشاهده شد. نتیجهگیری: نتایج نشان میدهد که ژن کالپاستاتین در بافتهای مختلف بز گسترده است. همچنین این پژوهش اساسی را برای انجام پژوهشهای بیشتر در مورد کالپاستاتین در بز کرکی راینی ایجاد نمود. پیشنهاد میشود که این مطالعه با تعداد دام بیشتر، جنسهای مختلف، سنین متفاوت و مراحل فیزیولوژیکی گوناگون در نژادهای مختلف بز انجام شود تا بتوان به یک نتیجهگیری جامعی دست یافت.
- چکیده انگلیسی: Objective Calpastatin has been found in skeletal muscle tissue of animals and could prevent unrestrained cellular growth by suppressing calpain activity. The involvement of calpains in apoptosis has been a subject of debate, although their involvement is limited to certain cell types and to specific stimuli. Various studies have shown that this gene is involved in growth performance and meat quality, therefore the aim of this study was to investigate Calpastatin gene expression in different tissues of Raini cashmere goat using Real Time PCR. Materials and Methods Totally 21 tissue samples including muscle, adipose, kidney, spleen, liver, lung and heart tissues were taken from 3 Raini cashmere goats. RNA was extracted and cDNA was synthesized. Real Time PCR was performed using SYBR Green method to study relative gene expression. GAPDH gene was used as housekeeping gene. Pfaffl method was used to analyze achieved data. Results For calpastatin gene 89bp fragment and for GAPDH 101bp fragment were observed in all studied tissues. Real Time PCR results of this study showed that Calpastatin gene is expressed in all studied (muscle, adipose, kidney, spleen, liver, lung and heart) tissues and the highest level of expression was observed in heart, spleen and liver tissues and the lowest level was seen in adipose tissue. Conclusions Results showed that Calpastatin are widespread in different tissues of goat. This study would also lay a foundation for further Calpastatin research in Raini cashmere goat. It is suggested that this study be conducted with greater number of livestock, different sexes, different ages and different physiological stages in different breeds of goats in order to reach a comprehensive conclusion.

نتایج: نتایج حاصل از این بررسی نشان داد که ژن DLK1 در تمامی بافتهای بررسی شده (بافت ماهیچه، چربی، کلیه، طحال، جگر و مغز) بیان شده است و بیشترین سطح بیان در بافت ماهیچه و کلیه و کمترین سطح بیان در بافت طحال و مغز مشاهده شد.
نتیجهگیری: این پژوهش اساسی را برای انجام پژوهشهای بیشتر در مورد Dlk1 در بز کرکی راینی و سایر نژادهای دیگر بز و حتی دام و طیور بومی ایجاد نمود. پیشنهاد میشود که این مطالعه با تعداد دام بیشتر، جنسهای مختلف، سنین متفاوت و مراحل فیزیولوژیکی گوناگون در نژادهای مختلف بز انجام شود تا بتوان به یک نتیجهگیری جامعی دست یافت.- چکیده انگلیسی: Objective
Dlk1 plays an important role in regulating skeletal muscle development and regeneration, differentiation of adipocytes, regulation of muscle cell growth, hepatoblast proliferation and blood cells formation. The aim of this study was to investigate Dlk1 gene expression in different tissues of Raini cashmere goat using Real Time PCR.
Materials and Methods
Tissue samples (18) including muscle, kidney, spleen, brain, liver and back fat tissues were taken from 3 Raini cashmere goats. RNA was extracted and cDNA was synthesized. Real Time PCR was performed using SYBR Green method to study relative gene expression. Beta actin gene was used as housekeeping gene. Pfaffl method was used to analyze achieved data.
Results
Results of this study showed that Dlk1 gene is expressed in all studied (muscle, kidney, spleen, brain, liver and back fat) tissues and the highest level of expression was observed in muscle and kidney tissues and the lowest level was seen in spleen and brain tissues.
Conclusions
This study would lay a foundation for further Dlk1 research in Raini cashmere goat and other goat breeds and also native animal and poultry. - چکیده: هدف: اپیژنتیک شامل تغییرات ژنتیکی قابل توارث است که بر الگوی بیان ژنها تاثیر دارد، اما این تغییرات به دلیل تغییر در توالی DNA نمیباشند. بهعلاوه، موجودات از طریق برخی از مکانیسمهای ملکولی و سلولی در مقابل عوامل بیماریزا مقاومت نشان میدهند. با توجه به نتایج مطالعات انجام شده به نظر میرسد که بین مکانیسمهای اپیژنتیکی و دفاعی ارتباط وجود داشته باشد. لذا، هدف تحقیق حاضر، بررسی چگونگی ارتباط اپیژنتیک و سیستم دفاعی موجودات و ارائه جدیدترین پیشرفتها در زمینة مطالعات انجام شده بر روی تنظیم اپیژنتیکی ارتباط موجود ـ میکروب (شامل باکتری و قارچ)، با تمرکز بر نقش تغییرات هیستون و متیلاسیون DNA ژنوم میزبان در مقاومت نسبت به بیماری و تجهیز سیستم دفاعی بود. بنابراین، مواردی از قبیل، مکانیسمهای ایجاد کننده تنظیمات اپیژنتیکی، نقش متیلاسیون DNA در اثر متقابل گیاه ـ میکروب، نقش متیلاسیون DNA بواسطه RNA در ارتباط گیاه ـ میکروب، تغییرات هیستونی، تغییرات هیستونی پس از ترجمه در اثرمتقابل گیاه ـ میکروب، نقش اپیژنتیک و microRNAs در سلامت حیوان، کنترل اپیژنتیکی تجهیز سیستم دفاعی و توارث فرا نسلی و تنظیم اپی ژنتیکی در عوامل بیماریزای گیاه و تاثیر آن بر بیماریزایی مورد بحث قرار گرفت. نتایج: توارث اپیژنتیکی میتواند جانوران یا گیاهانی را ایجاد کند که از طریق مکانیسمهای بلند مدت و کوتاه مدت خود را با شرایط خاص محیطی تطبیق دهند. بررسی نتایج مطالعات انجام شده توسط پژوهشگران مختلف شواهدی گویا در مورد مشارکت مکانیسمهای اپیژنتیکی در تجهیز دفاع فرانسلی (transgenerational defense priming) فراهم ساخته است. نتیجهگیری: درک بیشتر ما از نحوة مشارکت مکانیسمهای اپیژنتیک در سیستم دفاعی جانور یا گیاه و نحوة تعامل عوامل بیماریزا با این مکانیسم میتواند احتمالات جدیدی را برای استراتژیهای نوین حفاظت از محصولات فراهم سازد.
- چکیده انگلیسی: Objective Epigenetics contains studying inheritable genetic variations that affect on gene expression pattern, but these variations do not create because of changes in DNA sequence. In addition, organisms are resistant to pathogens through some molecular and cellular mechanisms. Thus, the purpose of this review was to overview interaction between epigenetics and defense system of organisms and highlight recent findings in epigenetic regulation of organism-microbe interaction with focusing on role of histone modifications and DNA methylation of host genome in resistance to disease and defense priming. Hence were discussed creating mechanisms of epigenetics regulations, role of DNA methylation in plant-microbe interaction, role of RNA-mediated DNA methylation in plant-microbe interaction, histone modifications, post-translational modifications of histones in plant-microbe interaction, role of epigenetics and microRNAs in animal health, epigenetic control of defense priming and transgenerational inheritance and epigenetic regulation in plant pathogenes and effects on pathogenesis. Results It was demonstrated that epigenetic inheritance can provide animals and plants with long-term and short-term mechanisms to adapt to specific environmental conditions. Reviewing results of performed studies by different researchers have provided clear evidence for the involvement of epigenetic mechanisms in transgenerational defense priming. Conclusions Moreover, more understanding of how epigenetic mechanisms contribute to animal and plant defence and how pathogens may counteract this reaction can provide new possibilities for novel production protection strategies.
