چکیده:
هدف: برآوردها نشان میدهد که در دنیا تقریبا سی میلیون راس بز و در ایران حدودا پنج میلیون راس بز کرکی در حال نگهداری و پرورش است. یکی از با اهمیتترین این نژادها بز کرکی راینی است. هورمونهای استروئیدی از طریق تنظیم رونویسی ژنهای هدف وظایف فیزیولوژیکی خود را انجام میدهند. به طوریکه گیرندههای این هورمونها از طریق شبکههای رونویسی در پوکی استخوان، التهاب و سرطان نقش ایفا میکنند. گیرنده استروژن (ER) یک فاکتور رونویسی است که واسطه عملکرد هورمون استروژن در بسیاری از فرآیندهای فیزیولوژیکی و آسیب شناختی است. دو گیرنده استروژن وجود دارد، ESR1 و ESR2که در اندامهای تولید مثلی واقع شده اند. میزان تولید مثل، به ویژه باروری، یکی از مهمترین صفات اقتصادی در تولید حیوانات است و توسط عوامل ژنتیکی و محیطی تنظیم میشود. لذا، هدف این پژوهش بررسی پروفایل بیانی ژن ESR2 در بافتهای مختلف بز کرکی راینی با استفاده از Real Time PCR بود. مواد و روشها: نمونه گیری از بافتهای بیضه، تخمدان، رحم، پستان، مغز، کلیه و قلب یک راس بز نر و یک راس بز ماده در کشتارگاه انجام شد (از هر بافت 3 تکرار). از بافتهای مورد نظر استخراج RNA کل انجام و کیفیت و کمیت آن ارزیابی شد. سپس cDNA ساخته و Real Time PCR اجرا شد. همچنین از ژن خانه دار GAPDH به عنوان کنترل استفاده شد. منحنیهای ذوب بررسی و تجزیه و تحلیل دادههای حاصل از Real Time PCR انجام شد. نتایج: نتایج منحنیهای Real Time PCR و مشاهده نتایج الکتروفورز محصولات PCR روی ژل آگارز 2 درصد نشان داد که ژن ESR2 در بافتهای بیضه، تخمدان، رحم، پستان، مغز، کلیه و قلب بیان شده است. بیشترین میزان بیان در تخمدان و سپس مغز و رحم مشاهده شد و کمترین سطح بیان مربوط به بافتهای کلیه، قلب و بیضه بود. نتیجهگیری: با توجه به نتایج پژوهش حاضر و نتایج سایر محققین میتوان نتیجه گرفت که ESR2 نقش مهمی در باروری مادهها دارد، در پژوهش حاضر مشخص شد ESR2در بافت تخمدان نسبت به بافتهای دیگر بسیار بیشتر بیان میشود. لذا، ESR2 به احتمال زیاد برای باروری مادهها بسیار ضروری هستند و نتایج این پژوهش را برای پژوهشهای آینده جهت توصیف نقش ژن ESR2 به عنوان یک ژن کاندیدا برای باروری بهتر و فیزیولوژی طبیعی در حیوانات اهلی، به ویژه بز فراهم کرده است.

Objective Estimates show that in the world about 30 million goats and in Iran about five million Cashmere goat are bred. One of the most important of these breeds is the Raini Cashmere goat. Steroid hormones perform their physiological functions by regulating the transcription of target genes. Receptors for these hormones play a role in osteoporosis, inflammation and cancer through transcription networks. Estrogen receptor (ER) is a transcription factor that mediates the function of estrogen in many physiological and pathological processes. There are two estrogen receptors, ESR1 and ESR2, which are located in the reproductive organs. Reproduction rate, especially fertility, is one of the most important economic traits in animal production and is regulated by genetic and environmental factors. Therefore, the aim of this study was to investigate the expression profile of ESR2 gene in different tissues of fluffy goats using Real Time PCR.
Materials and methods Sampling of testis, ovarian, uterine, breast, brain, kidney and heart tissues was performed from a male and a female goat (3 replicates from each tissue). Total RNA was extracted from the tissues and the quality and quantity of the extracted RNA were evaluated. The cDNA was then synthesized and Real Time PCR was performed. The GAPDH housekeeping gene was used as a control. Melting curves were examined and data obtained from Real Time PCR were analyzed.
Results The results of Real Time PCR curves and observation of electrophoresis of PCR products on 2% agarose gel showed that ESR2 gene was expressed in testis, ovary, uterine, breast, brain, kidney, and heart tissues. The highest level of expression was observed in the ovary and then the brain and uterus and the lowest level of expression was related to kidney, heart and testis tissues.
Conclusions According to the results of the present research and the results of other studies, it can be concluded that ESR2 plays an important role in female fertility, because in the present study it was found that ESR2 is much more expressed in ovarian tissue than other tissues. Therefore, ESR2 is likely to be essential for female fertility, and the results of this study provide the basis for future research to describe the role of the ESR2 gene as a candidate gene for better fertility and normal physiology in domestic animals, especially goats.
