- چکیده انگلیسی: Objective(s): -methyl-D-aspartate NMDA receptor (NMDAR) and aquaporin 4 (AQP4) are involved in the molecular cascade of edema after traumatic brain injury (TBI) and are potential targets of studies in pharmacology and medicine. However, their association and interactions are still unknown.
Materials and Methods: We established a rat TBI model in this study. The cellular distribution patterns of AQP4 after inhibition of NMDAR were determined by Western blotting and immunoreactive staining. Furthermore, the regulation of NMDA receptor 1 by AQP4 was studied by injection of a viral vector targeting AQP4 by RNAi into the rat brain before TBI.
Results: The results suggest that AQP4 protein expression increased significantly (P<0.05) after TBI and was down-regulated by the NMDAR inhibitor MK801. This decrease could be partly reversed using the NMDAR agonist NMDA. This indicated that AQP4 mRNA levels and protein expression are regulated by the NMDA signaling pathway. By injection of AQP4 RNAi viral vector into the brain of TBI rat models, we found that the mRNA and protein levels of NMDAR decreased significantly (P<0.05). This suggested that NMDAR is also regulated by AQP4.
Conclusion: These data suggested that the inhibition of AQP4 down-regulates NMDAR expression, which might be one of the mechanisms involved in edema after TBI.- انتشار مقاله: 25-10-1396

- چکیده انگلیسی: Objective(s): Occlusal trauma is one of the most common forms of oral biting dysfunction. Long-term occlusal trauma could weaken the stomatognathic system; especially damage one’s masticatory muscle. Through using the rat model, this study investigated the trophic effect of ciliary neurotrophic factor (CNTF) on injured masseter muscle. Materials and Methods: Male Wistar rats (n=36) were randomly divided into five experimental groups and one control group (6 rats per group). Animals in the experimental group were cemented modified crowns on their mandibular first molars to artificially induce occlusal trauma in 1, 3, 7, 14, and 28 days. Control group was sham-treated with forced mouth-opening for about 5 min, while no crowns were placed. After 28 days of treatment, all rats were euthanized and their masseter muscle was collected. Through immunofluorescence and real-time quantitative PCR, the expression of desmin, CNTF, and CNTFRα was investigated in rat masseter muscle. The microstructure of masseter muscle was observed by transmission electron microscope. Results: The expression of desmin showed a time-dependent decrease on traumatic and non-traumatic sides masseter, until reached the nadir at the 14th day, then restored to its normal level at the 28th day; however, the expression of CNTF and CNTFRα on the traumatic and non-traumatic sides increased from day 7, reached the peak at the 14th day, and returned to normal level on the 28th day. Conclusion:CNTF, as an important neurotrophic factor, was tightly associated to the restoring of rat injured masseter muscle, which provides new target and treatment method for clinical application.

- چکیده انگلیسی: A generation method for the rail random irregularity with arbitrary wavelength interval (WI) is developed, and its accuracy and efficiency are demonstrated. Then a moving wheel-rail-bridge interaction element is derived to establish the finite element equations of motion for the train-rail-bridge interaction system, and the flow chart of assembly and calculation for the system equations is given. According to the sub-interval principle, the influences of the irregularities with the large WI and the sub-intervals on the dynamic responses are analyzed by a numerical example, and the sensitive WI of each response is discussed. The results indicate that the bridge acceleration and the contact force are both more sensitive to the irregularity with WI (1~5 m). The irregularity with WI (0.1~1 m) has less influence on the car body acceleration but mainly contributes to the rail acceleration. However, all the irregularities with wavelengths in interval (1~150 m) can have significant influences on the car body acceleration. Meanwhile, the transient jump of wheel can be simulated and should be taken into account for the derailment risk assessment.

- چکیده انگلیسی: To determine changes in bacterial composition in skin ulcerations in farmed sea cucumber Apostichopus japonicus, full-length 16S rRNA gene sequencing of different skin samples of A. japonicus was performed on a Pacific Biosciences (PacBio) RS platform. The fact that one of them varies in bacterial diversity than the other two may suggest different stage of skin ulceration, but the answer is inconclusive due to inefficiency in size of sampling pool. After comparing them with three skin samples from three healthy sea cucumbers, respectively, the majority of the results were distinguishable at the species level. The bacterial composition, which increased in diversity during the possible early stage of skin ulceration, exhibited a decrease in diversity at possible advanced stages, except for the predominant Saccharicrinis carchari (846.85 ± 5.75‰). This work may have the first clue implicating Ornithinibacillus contaminans as a potential causative pathogen of skin ulcerations in A. japonicus.

- چکیده انگلیسی: Most bioenergy feedstock studies focus on maximizing aboveground biomass production. Cropland with fertile soils can produce high aboveground biomass yields but its diversion to bioenergy causes greenhouse gas emissions from direct and indirect land use changes. Here, we analyze three grassland experiments that minimize land use changes by using abandoned and degraded agricultural land. We find that soil and root carbon storage is a greater determinant of the climate change mitigation potential of biofuels than aboveground biomass, and tends to be higher for treatments with high plant diversity. Aboveground biomass yield ranged from 450-650 g ha-2 yr-1 for the productive treatments with moderate intensification, but its climate benefit via converting into biofuels and displacing fossil fuels can be substantially reduced by the rebound effect of fuel market. Because of high soil and root C storage rates (152-483 g CO2 ha-2 yr-1), many treatments are carbon negative even without the fossil fuel displacement benefit. To effectively mitigate greenhouse gas emissions, we should focus on increasing belowground carbon storage and explore the potential benefits of high-diversity plant species mixtures.
