- چکیده انگلیسی: BACKGROUND AND AIM: The calcifying epithelial odontogenic tumor (CEOT) is a rare entity described by Pindborg and represents < 1% of all odontogenic tumors. Nearly 200 cases of this neoplasm have been reported to date. It mostly occurs in the posterior mandible associated with an impacted tooth, most often a mandibular molar. Patients are usually between 30 and 50 years of age, with no sex predilection. CASE REPORT: Presented here is a case of an intraosseous CEOT, which occurred in a 16-year-old female in the body of the mandible extending from tooth nos. 32 to 47 with no specific clinical manifestations. CONCLUSION: CEOT accounts for < 1% of all odontogenic tumors. Approximately 200 cases have been reported today. This case report describes the clinical, radiographic and microscopic features of a large CEOT in a 16-year-old girl, which although was big in size, had no specific clinical manifestations. The patient reported a relatively rapid evolution of the tumor in the mandibular region, which is a unique manifestation.

- چکیده انگلیسی: Background: Chondroitinase ABC (cABC), due to its loosening impact on the extracellular matrix scaffold, has been used to enhance regeneration of injured axonal tracts after spinal cord injury (SCI). However, cABC thermal instability at physiological temperature has limited its clinical application. The disaccharide trehalose has been shown to increase the stability of cABC in body temperature. Therefore, the present study was conducted to assess the effect of combined trehalose and cABC on inflammation, lipid peroxidation and histopathological changes following SCI. Methods: Male Wistar rats were randomized into six groups (n=12) including sham, SCI, vehicle, trehalose, cABC and trehalose + cABC. In sham group, only laminectomy was performed. Other groups underwent laminectomy followed by contusion spinal cord injury. Twenty four hours after treatment, the level of IL-1β, Myeloperoxidase (MPO) and Malondialdehyde (MDA) were measured. Histopathological changes were also scored. Results: Spinal cord injury in rats resulted in severe trauma characterized by increase in inflammation, oxidative stress, neutrophils infiltration, hemorrhage, necrosis and edema. The levels of IL-1β, MDA and MPO were 260.3±16.4 nmol/mg protein, 1.2±0.06 mU/mg protein and 18.9±0.7 pg/mg protein, respectively in the vehicle group. However, combined treatment with cABC and trehalose reduced the amount of these factors significantly to 142.4±17 nmol/mg protein, 0.57±0.03 mU/mg protein and 13.8±0.4 pg/mg protein, respectively (P<0.05). In addition, treatment with cABC and trehalose improved histopathological alterations. Conclusions: The present data suggest that coadministration of trehalose and cABC exhibits neuroprotection effect through reducing inflammation and tissue injury events associated with SCI.

- چکیده انگلیسی: Background: Breast cancer is formed of a neoplastic component (epithelial) and a non-neoplastic component (stroma). Stromal- stromal and tumor- stromal interactions have been shown in the regulation of cancer cell growth, metastatic capacity and outcome of treatment. Tumor-associated macrophages (TAMs) are a component of tumor stroma reactionsand are considered as an important component of breast cancer tumor tissue which approximately form 50 % to 80 % of tumor tissue.In this study, the frequency of CD68 positive cells in association with other factors such as age, tumor size, ER, PR, Ki-67, Her2-neu receptors, stage and grade in invasive carcinoma tissues were morphologically and statistically evaluated. The frequency of CD68 was also discussed in relation to the number of involved lymph nodes. Method: A total of 50 invasive breast cancer patients with and without axillary lymph nodes involvement were studied. IHC staining for CD68 and Ki-67 markers was performed. For each tumor, 5 fields with different density of CD68 were counted under 400x by optical microscopy and the average of the five fields was taken as the percentage. Patients were divided into the two groups of low infiltration and high infiltration based on the percentage of CD68.By the same way, patients were divided into the two groups of low infiltration and high infiltration based on Ki67 percentage. Results: CD68 positive cells had significant correlation with ER negative and higher Ki67. No significant correlation was found between CD68 positive cells and the number of involved lymph nodes, age, size, HER2neu, PR, stage and grade. Conclusion: It seems that the presence of CD68 -positive macrophages in invasive breast cancers and nearby lymph nodes is associated with a worse prognosis.

- چکیده انگلیسی: Background: Following spinal cord injury, reactive astrocytes upregulate chondroitin sulfate proteoglycans (CSPGs) which act as a barrier to neuronal repair and regeneration. Therefore, enzymatic digestion of CSPGs by chondroitinase ABC (cABC) is a key strategy in the treatment of spinal cord injury. Furthermore, cABC has been shown to attenuate post spinal cord injury inflammation and may decrease astrocytes activation. Thus, this study was conducted to investigate the effect of cABC on astrocytes level through measuring glial fibrillary acidic protein (GFAP) in an animal model of spinal cord injury. Methods: spinal cord injury was performed through contusion method. Sixty male rats (220-250 g) were divided into three groups. The first group was just subjected to spinal cord injury. In the second group, phosphate buffered saline (6 µl) was immediately injected in lesion site after spinal cord injury. The third group received cABC (6 µl, 10 U/ml) intrathecally after spinal cord injury. Then, GFAP, CSPGs and chondroitin 4-sulfate (C4S) levels were measured at 4 hours and 1, 3 and 7 days after spinal cord injury using immunohistochemical method. Results: GFAP level increased in all groups until 7 days post spinal cord injury, but no significant difference was observed between enzyme- treated group and two other groups at similar time point (p˃0.05). CSPGs level increased significantly in the first and second groups whereas it decreased considerably in enzyme- treated group. In addition, due to enzymatic digestion of CSPGs, C4S level in the third group increased significantly. Conclusion: cABC enzyme decreased CSPGs level through degradation without any…… considerable effect on their source which is reactive astrocytes.

- چکیده انگلیسی: Background & Aims: This study was performed to investigate possible relationships between the manifestation of stromal cells (fibroblasts and/or myofibroblasts) by focusing on expression of their matrix metalloproteinase 9 (MMP9) and possible angiogenesis based on CD31 and CD34 antigen expression during the various steps of hyperplastic changes to precancerousstate and invasive breast cancer. Methods: Our study included 50 females with invasive ductal carcinoma. Samples were obtained by mastectomy or biopsy and were immunohistochemically stained for the MMP9, CD31, and CD34 antibody. microvessel count (MVC) was also carried out on samples. Statistical analysis of the data was performed using ANOVA and Student's t-test (P < 0.05). Findings were compared with our "Breast Cancer Data Bank" for reevaluation of their clinical staging. Results: Positive significant correlations were observed between expression of MMP9 and invasive ductal carcinoma in situ (DCIS) and fibrocystic disease ± ductal intraepithelial neoplasia (FCD ± DIN) areas (P=0.001). MMP9 expression in invasive areas was more strongly positive than precancerous areas. Statistically significant correlations were observed between MMP9 expression and CD31 in grade II in invasive areas. MVC was evaluated by CD31 antibody. It was found to be inversely related to increased MVC from invasive areas, DCIS, DIN, and normal areas (P<0.001). No significant difference was observed in MVC based on age, tumor size or steroid receptors in stroma of an invasive cancer, DCIS, and FCD ± DIN. Conclusion: MMP9 expression in invasive areas was more strongly positive than precancerous areas, and negative in normal areas. Angiogenesis can be observed before any significant changes in preinvasive breast lesions. The elevated content of microvessel count of the tumor may be an indicator for worse prognostic factor. The progression from epithelial hyperplasia toward DCIS, and then, invasive carcinoma seems too complex to be assumed a linear progression

- چکیده انگلیسی: A heterogeneous group of CNS tumors are characterized by mixed neuroepithelial and mesenchymal features. Glial tumors manifesting this phenomenon are referred to as gliosarcoma. These tumors are usually mistaken for cerebral metastases or meningioma at operation. Their histological studies have revealed an admixture of gliomatous and sarcomatous tissues, which leads to a biphasic pattern. The mesenchymal component can present in different forms such as fibrosarcoma, undifferentiated pleomorphic sarcoma, chondro-osteogenic, and myogenic differentiation, as well as angiosarcomatous and liposarcomatous types. Squamous differentiation, adenoid formations and glandular structures may also be displayed.
Herein, we report a rare case who was admitted to the emergency room with decreased consciousness resembling methadone poisoning. Clinical work-up showed a temporoparietal mass on radiological investigation. Histopathological evaluation of the brain mass revealed a gliosarcoma with adenoid formations and a mesenchymal component, which manifested as chondrosarcomatous differentiation. Immunohistochemical studies confirmed the histologic diagnosis through positivity for EMA, GFAP, S100, and vimentin expression in different components.- انتشار مقاله: 14-12-1398

- چکیده انگلیسی: Background & Objective:This study aimed to investigate the effect of decellularized allogeneic bone graft enriched by periosteal stem cells (PSCs) and growth factors on the bone repair process in a rabbit model, which could be used in many orthopedic procedures.
Methods: In this experimental study, a critical size defect (CSD) (10 mm) was created in the radial diaphysis of 40 rabbits. In group A, the defect was left intact with no medical intervention. In group B, the defect was filled by a decellularized bone graft. In group C, the defect was implanted by a decellularized bone graft enriched with platelet growth factors. In group D, the defect was treated by a decellularized bone graft seeded by periosteal mesenchymal stem cells (MSCs). Also, in group E, the defect was filled by a decellularized bone graft enriched with platelet growth factors and periosteal MSCs. Radiological evaluation was done on the first day and then in the second, fourth, and eighth weeks after the operation. The specimens were harvested on the 28th and 56th postoperative days and evaluated for histopathological criteria.
Results: The radiologic and microscopic analysis of the healing process in bone defects of the treated groups (C, D, and E) revealed more advanced repair criteria than those of groups A and B significantly (P<0.05).
Conclusion: Based on this study, it appears that implantation of concentrated PSCs in combination with growth factors and allogeneic cortical bone graft is an effective therapy for the repair of large bone defects.- انتشار مقاله: 29-10-1397

- چکیده انگلیسی: Background & Objective:Basal cell carcinoma (BCC) is classified into BCC1 or low risk (nodular, superficial type) and BCC2 or high risk (micronodular, morpheaform, infiltrative, and basosquamous types) based on clinical behavior. This study attempts to evaluate immunohistochemical (IHC) findings and clinical features associated with local aggressiveness and recurrence in BCC lesions.
Methods:This is a cross-sectional descriptive study conducted on 42 paraffin blocks (22 BCC1, 20 in BCC2) at Pathology Department of Afzalipour Teaching Hospital. First, demographic features of the patients were recorded and pathology blocks were classified by two dermatopathologists based on histopathological types of BCC1 and BCC2. Then, primary monoclonal antibodies including CD10, CD1a, SMA, Ki67, CD34, and P53 were utilized for IHC study. We compared BCC1 and BCC2 according to IHC markers, demographic features of patients, and tumoral features.
Results:The mean number of Langerhans cells (LCs) within epidermis above tumor mass was 14+1.92 and 4.7±1.23 in BCC1 and BCC2, respectively; these results show a significant difference between the two groups (P=0.001). P53 was positive in 41.13±6.39% and 74.5 ±6.26% of the tumor cells in BCC1 and BCC2 groups, which was statistically significant (P=0.001). Also, the mean number of blood vessels was 14.40±1.30 and 21.40±1.97 in BCC1 and BCC2, that was statistically significant (P=0.005).
Conclusion: Higher numbers of angiogenesis (SMA positive) and positive P53 were observed in BCC2 than BCC1. Also, more active positive CD1a cells were observed in BCC1 compared to BCC2.- انتشار مقاله: 20-12-1396

- چکیده انگلیسی: Background and Objective: In recent years, due to increasing number of patients with non-healing skin ulcers, skin substitutes have been used. Skin substitutes contain living cells causing faster and more effective wound healing. Therefore, research on the use of autologous and allogeneic cells such as fibroblasts in skin substitutes has attracted attentions. However, there are discrepancies in the immune responses to allogeneic fibroblasts. Therefore, we aimed to review the immune responses to allogeneic fibroblasts.
Methods: Donor fibroblasts were isolated from the skin of three rats. Nine recipient rats which were subcutaneously injected with three different regimens, were divided into three groups: Group 1; phosphate buffered saline (PBS) without cells (control), group 2: allogeneic fibroblasts of one animal source suspended in phosphate buffered saline, and group 3; phosphate buffered saline containing mixed allogeneic fibroblasts of three animal sources. The skin samples were biopsied at 1, 3 and 7 days after injection and studied histopathologically.
Results and Conclusion: No signs of redness and edema were observed in the injection sites. In pathology examination, changes such as vasculitis, eosinophils and lymphocytes accumulation around fibroblasts, fibroblast apoptosis and transplant rejection at the injection site were not observed in either group.
Subcutaneous injection of allogeneic fibroblasts in rats can be introduced as a promising approach for wound healing as they do not stimulate the immune system.- انتشار مقاله: 07-03-1396

- چکیده انگلیسی: Background and Objectives: The mesenchymal stem cells derived from peripheral blood (PB) have been recognized as a promising source for allogeneic cell therapy. The aim of this study was to investigate the isolation, growth and differentiation ability of peripheral blood-isolated mesenchymal stem cells.
Methods: The mononuclear cells were purified from fresh peripheral blood using density gradient centrifugation then cultured in a suitable medium, expanded and characterized. In the following, these cells were cultured in specific adipogenic and osteogenic differentiation media.
Results and Conclusion: In spite of the absence of any stimulating factor, the cells adhered to the flasks and developed a rather homogeneous, spindle-shaped morphology after consecutive passages. The cells were confirmed to have mesenchymal phenotype by expression of specific markers (CD90, CD105, and CD73) and absence of CD45 marker, which is specific for hematopoietic stem cells. They could differentiate into lineage-specific committed cells (osteoblasts and adipocytes).
According to the findings, the conventional, labour-intensive and time-consuming approaches are not necessary to obtain an optimal number of cells from peripheral blood. This relatively accessible and minimally invasive source of stem cells may open a new era for practical exploitation in regenerative medicine.- انتشار مقاله: 22-03-1396
