- چکیده انگلیسی: Mycobacterium tuberculosis (Mtb) is considered to be a major public health concern and a successful intracellular pathogen associated with high mortality worldwide. The Bacillus Calmette-Guerin (BCG) vaccine is the only available vaccine for the prevention of tuberculosis (TB) and tubercular meningitis in children. However, BCG is not adequately effective in the treatment of the adults affected to TB. According to the literature, there are controversial data on the potential role of B cells. B cells and humoral immune response play a key role in the amplification of the host immune response against TB. This review study aimed to discuss B cells and humoral immune responses in TB infection and assess its application as a therapeutic option. The monitoring of various B cell phenotypes in TB could be a reliable marker for the prediction of TB in individuals, especially in the latent form. According to the findings, the CMI response (especially Th1 activities) is not sufficient for efficient protection against TB, and B cells and Abs influence the innate immunocytes and Th1, while playing a pivotal role in various outcomes of exposure with tubercle bacilli. Although B cells may contribute to Mtb in the development of active TB, further investigations are required regarding the effects of B cells and humoral immunity on TB pathogenesis and the targeted harmful humoral-mediated response. Moreover, B cells and antibodies could be proper biomarkers to promote the studies regarding the detection of reliable diagnostic tools for the reactivation of latent TB, as well as use as a new generation of therapeutic options.

- چکیده انگلیسی: Background: The diagnostic methods which are used for acute ocular toxoplasmosis are very important; if the treatment is delayed, it sometimes leads to loss of vision. Fewstudies have been performed to evaluate serological tests used in the diagnosis of acute ocular toxoplasmosis. Objective: To evaluate the immunoglobulin (Ig) M, G and IgG avidity tests for diagnosis of acute ocular toxoplasmosis in the northeast of Iran. Methods: A cross-sectional study was carried out from January 2014 to December 2016. After an opthalmic examination was conducted by a retina specialist, 16 typical acute and 34 typical chronic ocular toxoplasmosis cases were included in this study. Information on clinical manifestations, age and occupation was recorded. Anti-Toxoplasma IgG, IgM and IgG avidity tests were administered on serum samples using the ELISA method. Results: Blurring of vision in all patients was the most clinical presentation. The IgG avidity test could diagnose all acute and recent cases. However, three false positive and one false negative result occurred using the IgM test by ELISA. The false negative result in all likelihood occurred because the patient was at the beginning stage of the infection. Conclusion: The result of this study showed that IgM is not a reliable marker of acute disease. Repetition of the serology tests was proposed in cases with clinical manifestations without detectable antibody titer after approximately two weeks. IgG avidity testing results coincided with clinical diagnosis and it could therefore considered to be a reliable method to differentiate between recently acquired and chronic ocular toxoplasmosis.

- چکیده انگلیسی: Objective(s): Despite treatment with antibiotics and vaccination with BCG, tuberculosis (TB) is still considered as one of the most important public health problems in the world. Therefore, designing and producing a more effective vaccine against TB seems urgently. In this study, immunogenicity of a fusion protein which consisting or comprising CFP-10 from Mycobacterium tuberculosis and the Fc-domain of mouse IgG2a was evaluated as a novel subunit vaccine candidate against TB.
Materials and Methods: The genetic constructs were cloned in pPICZαA expression vector and recombinant vectors (pPICZαA-CFP-10: Fcγ2a and pPICZαA-CFP-10:His) were transformed into Pichia pastoris. To evaluate the expression of recombinant proteins, SDS-PAGE and immunoblotting were used. The immunogenicity of recombinant proteins, with and without BCG were assessed in BALB/c mice and specific cytokines against recombinant proteins (IFN-γ, IL-12, IL-4, IL-17 and TGF-β) were evaluated.
Results: The levels of IFN-γ and IL-12 in mice that received recombinant proteins was higher than the control groups (BCG and PBS). Thus, both recombinant proteins (CFP-10:Fcγ2a and CFP-10:His) could excite good response in Th1-cells. The Fc-tagged protein had a stronger Th1 response with low levels of IL-4, as compared to CFP-10:His. However, the highest level of Th1 response was observed in groups that were vaccinated with BCG (prime) and then received recombinant protein CFP-10: Fcγ2a (booster).
Conclusion: The results demonstrated that binding mice Fc-domain to CFP-10 protein can increase the immunogenicity of the subunit vaccine. Further studies, might be able to design and produce a new generation of subunit vaccines based on the Fc-fused immunogen.- انتشار مقاله: 12-11-1395

- چکیده انگلیسی: Objective: In this study, antimicrobial activities of an ethanol extract of Tribulus terrestris aloneand in combination with Capsella bursa-pastoris and Glycyrrhiza glabra were examined in vitro against six pathogens namely Streptococcus mutans, Streptococcus sanguis, Actinomyces viscosus, Enterococcus faecalis Staphylococcus aureus, and Escherichia coli.
Materials and methods: Antibacterial activities of the extracts were examined using disc and well diffusion methods and the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of ethanol extracts were determined against these microorganisms using agar and broth dilution methods. Chlorhexidine was used as positive control.
Results: Tribulus terrestris extract exhibited good antibacterial activity against all bacteria. Antibacterial activity of mixed extract was evaluated and exhibited that mixed extract was more effective against all bacteria than any of the cases alone which indicates the synergistic effect between these three extracts (p˂0.05). No strain showed resistance against these extracts. In agar dilution, Tribulus terrestris exhibited MIC values ranging from 35.0 to 20.0 mg/ml and mixed extract showed MIC values ranging from 12.5 to 5.0 mg/ml. The results of broth dilution method were consistent with the findings of the agar dilution method.
Conclusion: This in-vitro study was a preliminary evaluation of antibacterial activity of the plants. It provided scientific evidence to support uses of T. terrestris and its mixture with C. bursa-pastoris and G. glabra for the treatment of oral infections. - چکیده انگلیسی: Objectives: Oral infections and dental caries are still considered as serious public health problems and inflict a costly burden to health care services around the world and especially in developing countries.
Materials and Methods: In the present study, we evaluated the antibacterial activity of Glycyrrhiza glabra (G. glabra) against oral pathogens by diffusion methods and determined the minimum inhibitory concentration (MIC) by both broth and Agar dilution methods and minimum bactericidal concentration (MBC) by broth dilution methods.
Results: In this study, G. glabra extract showed good antibacterial activity against six bacteria. No strain in this study showed resistance against this extract.
Conclusion: G. glabrais suggested as an appropriate candidate to help us in order to control dental caries and endodontic infections.- انتشار مقاله: 10-11-1390
