- چکیده انگلیسی: Oxygen is an essential part of the most important metabolic pathways in aerobic organisms. Oxygen delivery is merely dependent on blood, rendering blood loss a devastating event. Traumatic pre-hospital liver bleeding is a major cause of early trauma deaths in human and animals, with no established therapeutic method yet. Increasing intra-abdominal pressure (IAP) has been shown to reduce liver bleeding by half. Although reduction of blood loss could be in favor of blood oxygen delivery, however, the complex interaction between increased IAP and respiratory mechanics during severe hemorrhagic shock remained unclear. We used a novel model of liver trauma in 16 rabbits and randomly assigned them to either normotensive abdomen group or increased IAP by fluid infusion (HA) groups (n=8 each). Liver size and the amount of liver injury were evaluated. Various blood oxygenation parameters were recorded. Both groups were identical in terms of the liver size and injury. The HA group had significantly lower shock index. Arterial oxygen capacity and oxygen content were higher in the HA group. No significant statistical difference was seen between groups in terms of abdominal perfusion pressure; alveolar pressure of oxygen; dissolved oxygen in blood plasma; alveolar to arterial oxygen tension gradient; arterial to alveolar oxygen pressure ratio; the ratio between partial pressure of arterial oxygen and fraction of inspired oxygen; and respiratory index. In conclusion, the novel therapeutic method of increasing IAP by fluid infusion in a rabbit model of liver hemorrhage preserved blood oxygenation better than the classic therapeutic method.

- چکیده انگلیسی: Despite promising results observed using silicone rubber chamber, no previous comprehensive work was performed on behavior of the conduit. Present study aimed at further functional, histomorphometrical and immunohistochemical assessment of nerve regeneration in the same animal along a 10-mm rat sciatic nerve gap. Fifty- four male Wistar rats were divided into three experimental groups (n = 18), randomly: Sham-operation (NC), Transected control (TC) and silicone conduit (SIL). In NC group after anesthesia left sciatic nerve was exposed through a gluteal muscle incision and after haemoestasis the muscle was sutured. In TC group left sciatic nerve was exposed the same way and transected proximal to the tibio-peroneal bifurcation leaving a 10-mm gap. In SIL group left sciatic nerve was transected the same way and proximal and distal stumps were each inserted into a silicone tube. Each group was subdivided into three subgroups of six animals each and were studied 4, 8, 12 weeks after surgery. Functional analysis showed significant improvement of nerve function in SIL group than in TC group (P < 0.05). Morphometric indices and immuohistochemistry indicated there were significant differences (P < 0.05) between SIL and TC groups 12 weeks after surgery. Silicone entubulation technique has offered the hope of providing a method for achieving the peripheral nerve regeneration in the least harmful way that is available, easily performed. Using silicone tubes in bridging of nerve defects could be promising because it is inert and does not induce extensive scarring or degeneration after implantation.

- چکیده انگلیسی: Objective- The healing effect of chitosan supported cerium oxide nanoparticles (nano-CeO2) on cutaneous experimental excisional wound infected with pseudomonas aeruginosa was studied in rat.
Design- Experimental Study.
Animals- Sixty male albino rats.
Procedures- The chitosan supported nano-CeO2 hydrogel was prepared and characterized using Fourier-transform infrared spectroscopy (FT-IR), Energy Dispersive X-ray Spectroscopy (EDX), X-ray powder diffraction (XRD), Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM). The wound was infected with p. aeruginosa and in positive control (PC) group it was treated with silver sulfadiazine ointment with no treatment in negative control (NC) group. The infected wound was treated with chitosan suspension-gel, cerium oxide nanoparticles solution or chitosan supported-CeO2 nanoparticle gel in the treatment groups. The rats were randomized into five groups (n=12). Each group was subdivided into three subgroups of four animals each and studied 3, 7 and 14 days post-wounding.
Results- Plantimetry, histopathological assessments and hydroxyprolin content showed better and faster healing of the wounds in the treatment groups, particularly in chitosan supported-CeO2 nanoparticle gel (ChCe) group compared to the NC group (P< 0.05). Using by Ferric Reducing Antioxidant Power (FRAP) the total antioxidant capacity was statistically higher in treatment groups compared to NC group (p< 0.05). The bacterial numbers were significantly lower in ChCe and cerium oxide nanoparticles solution (Ce) groups compared to chitosan (CH) group on day 3 post-wounding (P< 0.05).
Conclusion and Clinical Relevance-Topical application of the chitosan supported nano-CeO2 particles on the infected wound enhanced tissue total antioxidant capacity, reduced the bacterial count, accelerated proliferation and migration of fibroblasts and keratinocytes, increased hydroxyproline level and neovascularization scale of the healing wound. Chitosan supported nano-CeO2 hydrogel could be suggested as an alternative therapy for treating of the infected wound.- انتشار مقاله: 15-05-1396

- چکیده انگلیسی: Objective- Adipose tissue is an appropriate source for isolation of cells with stem-cell-like properties. In the present long-term study, the effects of the omental adipose-derived nucleated cells (OADNCs) fraction were compared to those of the undifferentiated cultured bone marrow stromal cells (BMSCs) on sciatic nerve regeneration. Design- Experimental in vivo study. Animals- Fifty male White Albino rats. Procedures- The rats were divided into four experimental groups, randomly: Normal control (NC), inside-out vein graft (IOVG), the OADNC and the BMSC groups. A 10-mm sciatic nerve defect was bridged using an inside-out vein graft. In OADNC and BMSC groups, the vein was filled with OADNCs and BMSCs, respectively. Functional studies of sciatic function index, electrophysiologic, morphometric and immunohistochemistry assessments were performed at 16, 20 and 24 weeks after surgery. Results- There was no significant differences (P > 0.05) between BMSC and OADNC groups concerning recovery of the regenerated nerves, amplitude and time delay of electromyography. Compared to IOVG, OADNCs enhanced the nerve regeneration similar to undifferentiated BMSCs. Conclusion and Clinical Relevance- It was concluded the long-term morphometric results could be more reliable in histological study of sciatic nerve regeneration.

Conclusion and Clinical Relevance- Inside-out vein graft technique has offered the hope of providing a biological method for achieving the peripheral nerve regeneration in the least harmful way that is available, easily performed and affordable. It also averts the need for foreign materials that are likely to provoke a foreign body reaction.

- چکیده انگلیسی: Objective(s): To investigate the effect of topical administration of alpha-lipoic acid into chitosan conduit on peripheral nerve regeneration using a rat sciatic nerve transection model.
Materials and Methods: Forty five Wistar rats were divided into three experimental groups randomly. A 10-mm gap of sciatic nerve was bridged with a chitosan conduit following surgical preparation and anesthesia. In treatment group, the conduit was filled with 30 µl alpha-lipoic acid (10 mg/kg/bw).It was filled with 30 µl phosphate buffered saline solution in control group. In Sham group sciatic nerve was just exposed.
Results: The recovery of nerve function was faster in treatment group than in control, at 4 and 8 weeks after surgery (P-value<0.05). Conduction velocity was better in treatment group than in control group at 4 and 12 weeks (P-value<0.05). Recovery index was higher in treatment group than the control group, 8 weeks after surgery (P-value <0.05). Greater nerve fiber diameter, axon diameter, and myelin sheath thickness were observed in treatment group compared to control group at 8 and 12 weeks after surgery (P-value<0.05). The immunoreactivity of regenerated axons and myelin sheath in treatment group were far more similar to sham group.
Conclusion: Alpha-lipoic acid when loaded in a chitosan conduit could improve transected sciatic nerve regeneration in rat.- انتشار مقاله: 27-12-1393
