The present study attempted to investigate the key pathways and genes which are associated with hypoxia in the human breast carcinoma cell line MDA-MB-231 with searched in gene expression omnibus (GEO) database for mRNA microarray data of MDA-MB-231 in normal and hypoxia condition.
Methods. Three GEO datasets GSE37340, GSE39042, and GSE42416 were downloaded from the Gene Expression Omnibus (GEO) database that these GEO profiles have of 9 cell lines in hypoxia condition and 8 cell lines in normal condition. The differentially expressed genes (DEGs) between MDA-MB-231 cell line in hypoxia and normal condition were analyzed by Geo2R software. Next, all the differentially expressed genes (DEGs) with p Results. 34 genes were found to be at least two datasets (i.e., SLC2A3, BNIP3, ENO2, PFKFB3, PLOD2, SLC2A1, HK2, ADM and etc.) that two genes among up regulated genes (HK2, ADM) were expressed in all three datasets.
Conclusion. These identified genes and pathways could help to understand the mechanism of development of(Triple-negative breast cancer) TNBC under hypoxia condition. Also HK2, ADM, CENP family, might be promising targets for the TNBC treatment.- انتشار مقاله: 31-02-1398

- چکیده انگلیسی: Ichthyoses as epidermal genodermatoses are a large group of keratinization disorders that affect the entire integument, which is typically characterized by visible scaling and inflammation on the skin. Nowadays, in addition to clinical criteria, new molecular diagnostic methods, such as next-generation sequencing, can help to differentiate the subgroups of ichthyoses more precisely. These disorders are mostly classified based on clinical and histologic features and molecular markers. Inherited ichthyoses were divided into two groups: non-syndromic ichthyosis and syndromic ichthyosis. Non-syndromic ichthyosis is a group of various skin diseases with genetic and clinical heterogeneity. In this group, ichthyosis vulgaris and recessive X-linked ichthyosis are common and are often of delayed onset. Correct diagnosis of the molecular defects resulted from ichthyosis is useful for the prediction of the prognosis, genetic counseling (accurate risk assessment), prenatal diagnosis, and a better understanding of skin biology. However, the most essential and promising advantage of a precise molecular diagnosis is using gene therapy for its treatment, which may be considered as a subcategory of personalized medicine. This review is focused on the different aspects of non-syndromic ichthyoses pathophysiology.

- چکیده انگلیسی: Background: Recently has been suggested that LINC01296 has an important role in tumor-promoting in different malignancies. We performed first meta-analysis to assess the association between the LINC01296 expression and clinicopathological criteria and the survival of patients with cancers. Methods: Relevant articles Identified by PubMed, EMBASE, Web of Science, and Scopus searching between December 2000 and 28 December 2018. Binomial data were evaluated by the odds ratio (OR) as the rapid statistic. The association between overall survival (OS) and the LINC01296 expression was evaluated using pooling the hazard ratio (HR) with its corresponding 95% confidence interval (CI). Results: Finally, 9 studies with 720 patients with cancer were included. The expression of LINC01296 showed a significant positive association with TNM stage (OR = 2.67, 95% CI = 1.83-3.88), tumor stage (OR= 2.22, 95% CI= 1.34-3.66) and lymph node metastasis (OR = 3.07, 95% CI = 2.23-4.21). A shorter OS was significantly associated with the expression of LINC01296 (HR = 3.95, 95% CI = 2.65-5.25) and lymph node metastasis (HR = 2.39, 95% CI =1.16-3.63). The OS did not show significant association with gender (HR = 0.83, 95% CI = -0.63-2.30) and tumor stage (HR= 2.66, 95% CI= -0.22-5.54). Conclusion: In conclusion, the results of this meta-analysis suggest that the expression of LINC01296 might be considered as a potential biomarker in patients with cancer.
