- چکیده انگلیسی: Objective(s): Study of non-coding RNAs is considerable to elucidate principal biological questions or design new therapeutic strategies. miRNAs are a group of non-coding RNAs that their functions in PI3K/AKT signaling and apoptosis pathways after T cell activation is not entirely clear. Herein, miRNAs expression and their putative targets in the mentioned pathways were studied in the activated CD4+T cells.
Materials and Methods: Herein, proliferation rate and IL-2 secretion were measured in treated and untreated cells by IL-2. Putative targets of up-regulated miRNAs were predicted by bioinformatics approaches in the apoptotic and PI3K/AKT signaling pathways. Then the expression of two putative targets was evaluated by quantitative RT-PCR.
Results: Proliferation rate of treated cells by IL-2 increased in a dose- and time- dependent manner. Naive and activated CD4+T cells induced by different dose of IL-2 secreted abundant amounts of IL-2. Also, in IL-2 un-induced cells (IL-2 depleted cells) after 3 days, decrease of proliferation has been shown. In silico analysis predicted putative targets of up-regulated miRNAs such as AKT1, AKT3 and apoptotic genes in the activated cells induced or un-induced by IL-2. Decrease of AKT3 was shown by Q-RT-PCR as a potential target of miRNAs overexpressed in IL-2 depleted cells. But there was no significant difference in AKT1 expression in two cell groups.
Conclusion: Our analysis suggests that decrease of AKT3 was likely controlled via up-regulation of specific miRNAs in IL-2 depleted cells. Also it seems that miRNAs play role in induction of different apoptosis pathways in IL-2 induced and un-induced cells.- انتشار مقاله: 20-09-1393

- چکیده انگلیسی: Hemophilia B is an X-linked recessive bleeding disorder caused by heterogeneous mutations in factor IX
gene. In about one-third of cases it arises by a new mutation in germ-line cells. In this study carrier testing
was performed for females of a family with only one affected individual by single strand conformation polymorphism (SSCP). Results indicated that the SSCP band shift in the propositus was de novo and his mother and also sisters were not carrier. This finding was also confirmed by sequencing.

- چکیده انگلیسی: MicroRNAs (miRNAs) exert a critical influence on physiological and pathological processes through posttranscriptional
modification of their mRNA targets. They play important roles in tumorigenesis and are considered to
be potential diagnostic and prognostic biomarkers with various cancers. MiR-200c and miR-9 are regulatory elements
that can have dual impacts as oncogenes and/or tumor suppressor genes. MiR-200c regulates two transcription factors,
ZEB1 and ZEB2, while miR-9 is a regulatory factor for the E-cadherin protein which has a critical function in cell-cell
junctions and is inhibited by two transcription factors ZEB1 and ZEB2. In this study, expression levels of miR-200c
and miR-9, ZEB-1, ZEB-2 and E-cadherin were assessed in 30 non-small cell lung cancers (NSCLCs) by real-time
qPCR. MiR-9 was down-regulated significantly in tumor tissues compared to normal adjacent tissues, while there was
no significant change in expression level of miR-200c. On the other hand, ZEB1 demonstrated significant increase
and ZEB2a decrease at the mRNA level. These results indicate roles for miR-9 and ZEB1 in genesis of lung cancer,
although clinico-pathological associations were not evident. Further studies are necessary to assess implications for
