Background and objectives: Human adenoviruses cause a wide range of diseases, from self-limiting and mild infections to some life-threatening infections. Many studies have shown that components derived from medicinal plants have antiviral activity. Pistacia genus is rich in phenolic compounds and has antioxidant and antimicrobial effects. The aim of this study was to investigate antioxidant potential and antiviral effects of ethanol and crude extracts and different fractions of Pistacia atlantica Desf. leaves on adenovirus. Methods: Crude P. atlantica leaf extract was prepared by maceration with 80% ethanol. Hexane, chloroform, ethyl acetate and n-butanol fractions were prepared using liquid-liquid extraction method. Toxicity on HEp-2 cells and anti-adenoviral activity of the extract/fractions were evaluated by MTT colorimetric methods. The concentration that caused 50% viral inhibition (IC50) and 50 % cytotoxicity concentration (CC50) were evaluated using regression analysis. Selectivity index (SI), as a marker of antiviral activity, was calculated. To determine antioxidant activity the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay was used. Results: Ethyl acetate fraction showed the highest antioxidant activity with IC50of 1.54±0.12 μg/mL in DPPH scavenging assay. Based on our results, IC50 of P. atlantica crude extract on adenovirus was 15.72 μg/mL, with SI of 8.09; n-butanol fraction showed the highest anti-adenoviral activity among the fractions with IC50of 16.38 µg/mL and SI of 26.5. Conclusion: The ethanol extract and n-butanol fraction of P. atlantica leaves showed inhibitory effects on adenovirus and could be a new promising anti-adenovirus agent.- انتشار مقاله: 03-09-1398

- چکیده انگلیسی: Background: An efficient, promoted tri-component catalytic reaction between barbituric acid (or N,N-dimethyl barbituric acid), 4-hydroxy coumarin, and a wide range of aryl aldehydes using zinc oxide nanowires (ZnO NWs) to obtain some new 4-hydroxychromenylarylmethyl-6-hydroxypyrimidine-2,4-diones is described. Method: The reactants were successfully condensed via three C-C bond formation by zinc oxide nanowires (ZnO NWs) as an efficient, environmentally safe and recyclable nano catalyst to produce target molecules. In addition, the biological effects of synthesized products by the use of DPPH and acyclovir as positive controls and also Hep-2, vero cell, HSV-1, and adenovirus as four applied cell lines have been evaluated. Results: The results showed that synthesized products have anti-oxidant, cytotoxic and anti-viral activities and can offer promising prospect as biologically active agents. Conclusion: This achievement in an efficient and eco-friendly synthesis of novel analogous of hybrid molecules in aqueous media with special biological properties may engross chemists and pharmacologists as well as pharmacists in future.

- چکیده انگلیسی: Objective: Influenza virus, which is associated with high level of morbidity and mortality, has been recently considered a public health concern; however, the methods of choice to control and treat it are limited. Our previous study showed anti-influenza virus activity of pomegranate peel extract (PPE). In this study, the mechanism through which PPE acts against influenza virus A/Puerto Rico/8/34 (H1N1; PR8) was investigated.
Materials and Methods:Ethyl alcohol extract of pomegranate (Punica granatum L.) peel was prepared, and the action mechanism of PPE in inhibiting influenza replication was studied by time-of-drug-addition assay, virucidal activity, RNA replication, hemagglutination inhibition assay, viral mRNA expression, and western blot analysis.
Results: PPE inhibited viral polymerase activity, viral RNA replication, and viral protein expression but could not affect hemagglutination inhibition and virucidal activity. According to time-of-drug-addition assay results, PPE inhibited the virus adsorption and early steps of influenza replication.
Conclusion: This study demonstrated that the antiviral effect of PPE on influenza virus is most probably associated with inhibition of viral adsorption and viral RNA transcription.- انتشار مقاله: 16-04-1397

- چکیده انگلیسی: Objective: Influenza A virus infections are still a major health problem and the choices available for the control and treatment of the disease are limited. This research evaluated in vitro and in vivo antiviral effects of Peganum harmala L. seeds (PHS) extract against influenza A virus.
Materials and Methods: In this research, in vitro anti-influenza A virus activity of the extract was assessed in Madin-Darby canine kidney (MDCK) cells. In order to evaluate anti-influenza activity of PHS extract in vivo, BALB/c mice were infected with 5LD50 of mouse-adapted influenza virus (H1N1; PR8) and received 200 mg/kg/day of PHS extract or 20 mg/kg/day oseltamivir. Lungs of seven mice per group were removed on day 3 post-infection and lung virus titers were determined by qRT-PCR. Mice survival, body weights and general conditions were observed for up to 14 days post-infection.
Results: The results demonstrated that, the ethanolic extract of PHS possesses high activity against influenza virus with IC50 value of 15.7 (CI95%:11.7-21) μg/ml in MDCK cells. Our results also showed that, oral administration of PHS extract (200 mg/kg/day) or oseltamivir (20 mg/kg/day) to infected mice, increased the survival rate, reduced body weight loss, and decreased lung virus titer.
Conclusion: Based on our findings, P. harmala seeds extract can inhibit influenza A virus replication in vitro and in vivo. Therefore, isolation and characterization of the plant’s active compounds and investigation of the underlying mechanisms of its antiviral action are highly suggested.- انتشار مقاله: 09-12-1395

Objective: There are a number of published data indicating in vitro anti-HSV activity of some of Iranian herbal extracts with no systematic review to discuss these results. Therefore, this article was aimed to review and discuss the methods carried out and the phytochemistry and bioactivity of the extracts used and also conclusions provided in these publications.
Materials and Methods: Published articles both in English (from Medline, Science Direct, EMBASE, Scopus, Pro Quest, Google scholar, Cochrane Library) and in Persian (from SID, Iran Medex and Magiran) databases, from 1966 to October 2014 were incorporated in this review. The in vitro studies that lacked CC50, IC50, were excluded.
Results: Only 42 published reports were found to examine Iranian herbs against HSV replication in vitro. Seventeen out of 42 studies in which 23 kinds of medicinal plants were subjected to crude extraction were included. The review of data showed that some of the herbal extracts including Hyssopus officinalis methanolic extract, Melissa officinalis aqueous extract, Quercus persica L. hydroalcoholic extract and Securigeras ecuridaca methanolic extract with selective index (SI) of 234, 877, >778 and 250, respectively were highly effective against HSV in vitro.
Conclusion: More comprehensive studies using more advanced methods are needed to be done to achieve promising anti-HSV agents from the bioactive compounds isolated from these herbs.- انتشار مقاله: 10-04-1394

- چکیده انگلیسی: Background and aims: Human adenoviruses can cause a diversity of clinical diseases, but there is no antiviral therapy formally approved by adenovirus infections. Thus, antiviral agents derived from medicinal plants which are effective against adenoviruses infections are urgently required. Therefore, this research was aimed to evaluate in vitro antiadenovirus activity of pomegranate (Punica granatum L.) peel extract. Methods: In this research, crude ethanol extract of pomegranate peel was prepared. Anti-adenovirus activity of the extract was evaluated on Hela cell line using MTT (3-[4,5-dimethylthiazol–2-yl]-2,5-diphenyltetrazolium bromide) assay. The 50% inhibitory concentration (IC50) and 50% Cytotoxicity Concentration (CC50) of the extract were determined using regression analysis. To determine antioxidant activity, total phenol content, and flavonoids content of the extract, the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay; Folin-Ciocalteu method and aluminum chloride colorimetric method was used, respectively. Results: The CC50 and IC50 of the extract were 165±10.1 and 18.6±6.7µg/ml, respectively. The selectivity index (SI), the ratio of CC50 and IC50, was 8.89. The IC50 of DPPH radical was 7.7±1.21 μg/ml, compare with butylated hydroxytoluene (BHT), with IC50 of 25.41±1.89 μg/ml. The total phenol and Flavonoid contents were 282.9 mgGAE/g and 136.6mg/g, respectively. This study revealed that the pomegranate (Punica granatum L.) Conclusion: peel extract exhibited Anti-adenovirus activity, with SI value of 8.9, suggesting its potential use as Anti-adenovirus agents. Also this extract with high phytoconstituents could be a promising source of medicinally important natural compound.
