- چکیده انگلیسی: Objective: The aim of the present study was to evaluate the possible protective effect of Plantago major (P. major) extract against doxorubicin (DXR)-induced renal inflammation in rats.
Materials and Methods: 80 male albino rats were randomly divided into 8 groups as follows: control, DXR, Ext (extract) 600, Ext1200, dexamethasone+DXR, vitamin E+DXR, Ext600+DXR, and Ext1200+DXR. Duration of the study was 35 days and DXR was intravenously injected on the 7th day of the experiment. Tumor necrosis factor-alpha (TNF-α) production and monocyte chemoattractant protein-1 (MCP-1) expression levels were assessed in the left kidney. Serum creatinine concentration and osmolarity were determined on the 1st, 14th, 21st, 28th and 35th days of the experiment.
Results: DXR caused a significant increase in renal expression of MCP-1 and TNF-α production compared to control animals. Administration of dexamethasone, vitamin E and P. major extract significantly improved the expression of these inflammatory mediators compared to DXR group. Compared to day 1 in DXR group, serum osmolarity showed a significant increase on days 21, 28 and 35. Also, on these days, serum osmolarity in DXR group was significantly higher than that on the same days in control group. In Vit E+DXR and Ext 1200+DXR groups, there was no significant changes in serum osmolarity among different days of the study. However, in these groups, serum osmolarity on days 21, 28 and 35 showed a significant decrease compared to the same days in DXR group.
Conclusion: Present results suggest that hydroethanolic extract of P. major protected renal tissue against DXR–induced renal inflammation.- انتشار مقاله: 22-09-1396

- چکیده انگلیسی: Objective: Angiotensin II (Ang II), the main product of renin-angiotensin system (RAS) has a well-known role in cardiovascular regulation. Over-production of Ang II is one of the important underlying mechanisms of hypertension. In this study, the effect of crocin on cardiovascular responses in rats with acute hypertension induced by Ang II was evaluated.
Materials and methods: Rats were divided into six groups (n = 6): 1) Control: rats that received saline, 2) Ang II: rats that received Ang II (300 ng/kg) infused in two min, 3) Losartan (Los) + Ang II : rats that received Los (10 mg/kg, i.v) before Ang II, and 4-6) Crocin (Cro) + Ang II groups: rats that received three doses of crocin (50, 100 and 200 mg/kg, slow i.v) 10 min before Ang II. Femoral artery and vein were cannulated for recording of cardiovascular parameters and injection of drugs, respectively. Systolic blood pressure (SBP), mean arterial blood pressure (MAP) and heart rate (HR) were continuously recorded by power lab system. After injection of reagents and extracts, maximum changes (∆) of MAP, SBP and HR were recorded and compared with control group.
Results: Ang II (300 ng/kg) increased maximal changes in MAP, SBP and HR compared to control group (p<0.001) and Los significantly attenuated these effects of Ang II (p<0.001). - چکیده انگلیسی: Objective: Inflammation and oxidative stress is considered to have a crucial role in induction of nephropathy. Curcuma longa (C. longa) and Nigella sativa (N. sativa) have anti-inflammatory and antioxidant effects. This study was designed to investigate the effect of mixed hydro-alcoholic extract of N.sativa and C. longa on the oxidative stress induced by Adriamycin (ADR) in rat kidney.
Material and Method: The animals were divided into 6 groups: control (CO), ADR, Adriamycin+ Vitamin C (ADR+VIT C), C. longa extract+ Adriamycin (C.LE+ADR), N. sativa extract+ Adriamycin (N.SE+ADR) and C. longa extract+ N. sativa extract + Adriamycin (N.S+C.L+ADR). ADR (5mg/kg) was injected intravenously, whereas VITC (100mg/kg) and extract of C. longa (1000mg/kg) and N. sativa (200mg/kg) were administrated orally. Finally, the renal tissue, urine and blood samples were collected and submitted to measure of redox markers, osmolarity and renal index.
Results: The renal content of total thiol and superoxide dismutase (SOD) activity significantly decreased and Malondialdehyde (MDA) concentration increased in Adriamycin group compared to control group. The renal content of total thiol and SOD activity significantly enhanced and MDA concentration reduced in treated-mixed extract of C. longa and N. sativa along with ADR group compared to ADR group. The mixed extract did not restore increased renal index percentage induced by ADR. There also was no significant difference in urine and serum osmolarity between the groups.
Conclusion: hydro-alcoholic extracts of N.sativa and C.longa led to an improvement in ADR-induced oxidative stress and mixed administration of the extracts enhanced the aforementioned therapeutic effect.- انتشار مقاله: 18-12-1393

- چکیده انگلیسی: Objective: Neuroimmune factors contribute on the pathogenesis of sickness behaviors. Nigella sativa (NS) has anti-inflammatory, anti-anxiety and anti-depressive effects. In the present study, the effect of NS hydro-alcoholic extract on sickness behavior induced by lipopolysaccharide (LPS) was investigated.
Materials and Methods: The rats were divided into five groups (n=10 in each): (1) control (saline), (2) LPS (1 mg/kg, administered two hours before behavioral tests), (3-5) LPS-Nigella sativa 100 , 200 and 400 mg/kg (LPS-NS 100, LPS-NS 200 and LPS-NS 400, respectively). Open- field (OF), elevated plus maze (EPM) and forced swimming test (FST) were performed.
Results: In OF, LPS reduced the peripheral crossing, peripheral distance, total crossing and total distance compared to control (pConclusion: The results of the present study showed that the hydro-alcoholic extract of NS reduced the LPS-induced sickness behaviors in rats. - چکیده انگلیسی: Objective: Rosa damascena mill L. (R. damascena) is a well-known plant with fragrant effects. Several therapeutic effects of this plant on respiratory, gastrointestinal and nervous systems have been reported. It is also suggested to have beneficial effect on cardiovascular system especially blood pressure regulation. The present study was carried out to evaluate acute cardiovascular effect of hydro-alcoholic extract of R. damascena.
Materials and Methods: Thirty-two male Wistar rats were randomly divided into four groups (n= 8 for each group). After anesthesia, a catheter was inserted into the femoral artery and blood pressure and heart rate (HR) were continuously recorded by a power lab system. Animals received three doses of hydro-alcoholic extract (250, 500, and 1000 mg/kg) via peritoneal (i.p). After 30 min, systolic blood pressure (SBP), mean arterial pressure (MAP) and HR were recorded and maximal changes were compared to control group.
Results: Injection of all doses of the extract did not significantly change HR compare to control group. The SBP, dose dependently, was decreased by all doses of the extract and the maximal response was significant compared to saline group (p
Conclusion: This study provides evidence of a hypotensive effect of hydro-alcoholic extract of R. damascena with no significant effect on HR. - چکیده انگلیسی: Objective: The use of drugs with herbal origin is increasing for treatment of gastrointestinal (GI) disorders. Rosa damascena (R. damascena) is a well-known plant suggested to have beneficial effect on GI system. In this study, the effect of aqueous fraction of R. damascena on the contractionsofisolated guinea pig ileum was investigated.
Materials and Methods: Aqueous fraction of plant was obtained from ethanolic extract after ethyl acetate and n-butanol fractions were discarded. To evaluate effect of this fraction on ileum contraction, guinea pig ileum was removed and mounted on organ bath and its contraction was recorded. Effect of various concentrations (0.66, 0.83, and 1.3 mg/ml) of aqueous fraction on ileum contraction in comparison with Ach in presence and absense of atropine, a muscarinic antagonist of cholinergic, was evaluated. The response of ileum to 1 µg/ml of acetylcholine was considered as 100% response.
Results:Our results showed that aqueous fractions of R. damascena dose-dependently increased basal guinea pigs ileum contractions (pConclusion: It is concluded that aqueous fraction of R. damascena has mild excitatory effect on ileum contraction and this fraction may be beneficial as a mild laxative agent.- انتشار مقاله: 05-10-1391

- چکیده انگلیسی: Objective: Rosa damascena mill L (R. damascena) is a medicinal plant mostly known in the world for its perfume. It also has beneficial effects on stress, tension and depression. In this experiment antidepressant effect of ethanolic extract of R. damascena by forced swimming test (FST) was evaluated. Material and Methods: Animals received ethanolic extract (15, 60 and 90 mg/kg, i.p), imipramine (15mg/kg, i.p; positive control), or saline (negative control). Thirty min post- injection, immobility and swimming times were measured and compared in the different studied groups. Results: Intraperitoneal injection of lower concentration of extract (15 mg/kg) did not change swimming and immobility times compared to the control group. The higher concentrations of extract (60 and 90 mg/kg) significantly increased immobility time and decreased swimming time. Therefore ethanolic extract at tested doses had no antidepressant effect in this study. Conclusion: Although ethanolic extract did not have antidepressant effect, we cannot rule out this effect altogether. In our opinion, antidepressant effect is masked by CNS depression effect of ethanolic extract of R. damascena.

- چکیده انگلیسی: Objective: Rosa damascena mill L (R. damascena) is an ornamental plant that has several therapeutic (such as sedative and hypnotic) effects. It also heals depression, grief, nervous stress and tension. In the present study we evaluated antidepressant-like effect of R. damascena using forced swimming test (FST). Material and Methods: Three doses of aqueous extract (15, 60 and 90 mg/kg) was injected intraperitoneally. After 30 min of injection, immobility and swimming times were measured and compared with control (negative control) and imipramine (positive control). Results: The results showed that low dose (15mg/kg) of extract significantly increased swimming time and decreased immobility time. However, the two high doses of extract (60 and 90mg/kg) had no significant effect on these parameters. Conclusion: These results proposed antidepressant-like effect of low concentration of aqueous extract of R. damascena.
