- چکیده انگلیسی: Objective(s) In the present study, the effects of aqueous-ethanolic extract from Rosa damascena on heart rate and contractility were examined. Materials and Methods Isolated guinea-pig hearts were perfused through aorta in a Langendorff mode. Heart rate (HR) and contractility were determined in the presence of four concentrations of the extract (0.1, 0.2, 0.4 and 1.0 mg %) and isoprenaline (1, 10, 100 nM and 1 μM) in comparison with baseline values in the presence and absence of propranolol (n= 10 for each group). Results Both isoprenaline and the extract caused increase in heart rate and contractility (P< 0.05 to P< 0.001). The percent increased in HR due to the final concentration of isoprenaline in the absence of propranolol was significantly greater than that of the extract (P< 0.01). Propranolol caused significant reduction in both HR and contractility (P< 0.05 for both) but this effect was significantly reversed by isoprenaline and the extract (P< 0.05 to P< 0.001). The percent increased in heart contractility due to the final concentration of the extract in the absence and presence of propranolol was significantly greater than that of isoprenaline (P< 0.05 for both cases). There was significant correlation between both HR and heart contractility with concentration of isoprenaline and the extract (P< 0.05 to P< 0.001). Conclusion In conclusion this study showed a relatively potent inotropic and chornotropic effect for Rosa damascena on isolated guinea-pig heart.

- چکیده:
- چکیده انگلیسی: Objective In previous studies, the relaxant, anticholinergic (functional antagonism), antihistaminic and its stimulatory effects on b-adrenoceptors of Nigella sativa have been demonstrated on guinea pig tracheal chains. In the present study, the relaxant effects of hydro-ethanolic, macerated aqueous (MA) and lipid-free macerated aqueous (LFMA) extract of Nigella sativa on tracheal chains of guinea pigs were examined. Materials and Methods The relaxant effects of four cumulative concentrations of each extract (0.8, 1.2, 1.6 and 2.0 g%) was compared with saline as negative control and four cumulative concentrations of theophylline (0.2, 0.4, 0.6 and 0.8 mM) on precontracted tracheal smooth muscle of guinea pig (60 mM KCl in group 1 and 10 µM methacholine in group 2, n=6 for each group). Results In group 1 all concentrations of theophylline, the last two concentrations of MA and the last three concentrations of LFMA extracts showed significant relaxant effects compared to that of saline (p<0.05 – p<0.005). Two final concentrations of hydro-ethanolic extract caused contraction in comparison with saline in this group. In group 2 all concentrations of theophylline, MA and LFMA and the last three concentrations of hydro-ethanolic extracts showed significant relaxant effects relative to that of saline (p<0.005 – p<0.001). The relaxant effect of different concentrations of MA, the last three concentrations of hydro-ethanolic and LFMA extract was significantly greater in group 2 as compared with group 1. However, there was no significant difference in the relaxant effect of different concentrations of theophylline, between group 1 and 2. In both groups, the relaxant effect of most concentrations of MA, hydro-ethanolic LFMA extracts was significantly less than those of theophylline (p<0.05 – p<0.001). In group 1 and 2, the relaxant effect of all concentrations of hydro-ethanolic extract was significantly lower than most concentrations of others (p<0.05 – p<0.01). There was a significant positive correlations between the relaxant effects and concentrations for theophylline and all extracts (except hydro-ethanolic extract in group1) in both groups (p<0.05 – p<0.001). Conclusion These results showed that mainly water soluble substances of Nigella sativa were responsible for the relaxant effect of the plant on tracheal chains of guinea pigs.

- چکیده:
- چکیده انگلیسی: Objective: This is the first study to evaluate the effect of carvacrol on pulmonary function tests (PFT), and total and differential white blood cell (WBC) counts in healthy volunteers.
Materials and Methods: Thirty healthy volunteers were recruited based on the inclusion and exclusion criteria. The subjects were treated with two doses of carvacrol (1 and 2 mg/kg/day) for 1 month. Total and differential WBC counts and PFT were examined before and after the treatment period.
Results: There were no statistically significant differences in terms of total and differential WBC counts between pre and post-treatment with the two doses of carvacrol. Also, results of PFT tests indicated that administration of 1 mg/kg/day carvacrol had no effect on PFT parameters when comparing post-treatment values with pre-treatment values. Treatment with 2 mg/kg/day carvacrol for 1 month increased forced expiratory volume in first second (FEV1) (p<0.05). However, the changes in total and differential WBC counts as well as PFT values after 1-month treatment were not significantly different between two groups.
Conclusion: The results of this study indicated that treatment of healthy individuals with 1 and 2 mg/kg carvacrol for 1 month has no negative effects on total and differential WBC counts nor PFT values.- انتشار مقاله: 09-05-1397

Objective: Pharmacological effects of carvacrol such as its anti-inflammatory activities have been shows. In this study the effects of carvacrol on serum levels of total protein (TP), phospholipase A2 (PLA2) and histamine in sensitized guinea pigs was evaluated.
Materials and Methods: Sensitized guinea pigs were given drinking water alone (group S), drinking water containing three concentrations of carvacrol (40, 80 and 160 µg/ml) or dexamethasone. Serum levels of TP, PLA2 and histamine were examined I all sensitized groups as well as a non-sensitized control group (n=6 for each group).
Results: In sensitized animals, serum levels of TP, PLA2 and histamine were significantly increased compared to control animals (p<0.05 to p<0.001). Significant reduction in TP, PLA2 and histamine levels were observed in treated groups with the two higher concentrations of carvacrol but dexamethasone treatment only decreased serum level of PLA2 (p<0.001). Although the effect of the lowest concentration of the extract was less than that of dexamethasone (p<0.05 for TP and p<0.001 for PLA2), the effects of the two higher concentrations on PLA2 were similar to dexamethasone and on TP (p<0.01) and histamine (p<0.001) were higher than those of dexamethasone.
Conclusion: These results showed that carvacrol reduced serum levels of TP, PLA2 and histamine in sensitized guinea pigs which may indicate an anti-inflammatory effect of this agent in inflammatory disorders such as asthma.- انتشار مقاله: 04-05-1395

- چکیده:
- چکیده انگلیسی: Objective: Asthma is an airway complex disease defined by reversible airway narrowing and obstruction, chronic airway inflammation, airway hyperresponsiveness, and tissue remodeling. The purpose of this study was to determine the effect of Brassica napus L. (B. napus) on airway pathologic changes in a rat model of asthma.
Materials and Methods: Twenty-four rats were divided into 4 groups: control, asthmatic, asthmatic treated with 0.5 mg/kg B. napus oil, and asthmatic treated with 0.75 mg/kg B. napus oil. To induce the experimental asthma, rats in groups 2, 3, and 4 received an i.p. injection of ovalbumin and aerosolized ovalbumin. Simultaneously, rats in groups 3 and 4 received B. napus oil daily by gavage. After 31 days, in all groups, thoracotomy was done and lung tissue samples were taken. For pathological evaluation, microscopic slides were prepared. The eosinophil numbers in the submucosal layer and thicknesses of smooth muscle layer of bronchioles were detected.
Results: Eosinophil numbers in the submucosal layer, as well as smooth muscle layer thicknesses were significantly lower in the rat group treated with 0.75 mg/kg B. napus oil as compared with asthmatic group (p<0.01, p<0.05).
Conclusion: B. napus could be useful as adjuvant therapy in rat model of asthma. This effect was probably related to its antioxidants componentsthat reduce the levels of inflammatory mediators such as leukotrienes, IL-4, IL-5, and IL-13.- انتشار مقاله: 04-06-1391
