- چکیده انگلیسی: Acetaminophen (acetyl-para-amino phenol; APAP)-induced hepatotoxicity is the most common form of drug-induced liver injury (DILI) worldwide. APAP is also used as a model drug to assess hepatoprotective strategies against DILI. In the current study, the potential cytoprotective effects of Allium cepa (Onion) extract (OE) was investigated in APAP-treated hepatocytes. Isolated hepatocytes were prepared with the collagenase perfusion of rat liver. Isolated hepatocytes (10 mL, 106 cells/mL) were incubated in the Krebs Henseleit buffer (pH = 7.4) in continuously rotating 50 mL round bottom flasks, under an atmosphere of carbogen (95% O2 and 5% CO2) in a 37 °C water bath. Cytotoxicity, ROS formation, and mitochondrial membrane potential collapse were assessed as oxidative stress markers. APAP administration to rat hepatocytes (500 µM) was accompanied by cytotoxicity, ROS formation, depletion of cellular glutathione (GSH) reservoirs, and mitochondrial depolarization. It was found that OE administration (100 µL) significantly reduced cell death, ROS formation, and its consequences, such as the decrease in cellular GSH and mitochondrial injury induced by APAP. These results indicate that the crude extract of Allium cepa exhibits hepatoprotective action, probably through antioxidative properties and protecting vital cellular organelles such as mitochondria.

- چکیده انگلیسی: Eremostachys laciniata, having rich flavonoids content, is expected to have a considerable antioxidant effects. We used ACMS (Accelerated cytotoxic or protective mechanism screening technique) to evaluate possible antioxidant effect of E. laciniata rhizome against oxidative cell damages induced by different types of oxidative stress such as iron-8-hydroxyquinolin (IQ) complex and copper in freshly isolated liver cells. The extracts were prepared with n-hexane, dichloromethane and methanol. Hepatocytes were isolated from male Sprague-Dawley rats by a two-step collagenase perfusion. Cell viability was measured by trypan blue exclusion method. DPPH (2, 2-diphenyl-1-picrylhydrazyl) assay was used to evaluate the antioxidant activity. ROS formation was measured by using DCFDA (2, 7-dichlorofluorescin diacetate) probe, mitochondrial membrane potential (MMP) was assessed by rhodamine 123 fluorescence and lipid peroxidation was determined by thiobarbituric acid reactive substances (TBARS) assay. The MET extract was demonstrated to possess a significant radical scavenging activity (RC50%=0.212). Unlike MET extract, the n-hexane and dichloromethane extracts showed toxic effects in cell suspensions. The MET extract significantly decreased cell death and ROS formation induced by IQ complex and copper and demonstrated protective effects against copper-induced mitochondrial membrane potential collapse and lipid peroxidation. The protection induced by MET extract can be attributed to antioxidant characteristics of phenylethanoids content.

- چکیده انگلیسی: Overdose of acetaminophen causes severe hepatic necrosis in humans and experimental animals. Studies on its hepatotoxicity remain a very active area since some of current data are still uncertain. In this study, freshly isolated rat hepatocytes were used to determine the effects of garlic extract and its component, allicin on the acetaminophen-induced cell cytotoxicity and to compare with the effect of N-acetyl cysteine as a standard treatment. Garlic extract was prepared via a standard method and its allicin and allyl mercaptan contents were determined using analytical and preparative high performance liquid chromatography (HPLC). Rat hepatocytes were isolated using collagenase perfusion and mitochondrial membrane potential and cell cytotoxicity were determined using Rhodamine 123 fluorescence and trypan blue exclusion, respectively. Inclusion of garlic extract and/or N-acetyl cysteine resulted in a reduction in the loss of mitochondrial membrane potential as well as cell death which occurred after acetaminophen addition and therefore, illustrated considerable hepatoprotective effects without significant differences between two treatments. In contrast, pure allicin was not effective significantly. The hepatoprotective effects of garlic extract may be due to the compounds other than allicin such as allyl mercaptan, as allicin has been shown to transform to allyl mercaptan as a major metabolite.
