- چکیده انگلیسی: Statement of the Problem: Adequate compressive strength is an important characteristic for an ideal liner.
Purpose: This study aimed to assess the compressive strength of several commonly used liners.
Materials and Method: This in vitro, experimental study evaluated 120 samples fabricated of Dycal, Calcimol LC, Vitrebond, Activa Bioactive, and TheraCal LC (n=24) liners according to the manufacturers’ instructions. The samples were fabricated using a cylindrical stainless steel mold with 6±0.1 mm height and 4±0.1 mm internal diameter. Half of the samples in each group (n=12) underwent compressive strength test immediately after completion of their primary setting while the other half (n=12) underwent compressive strength test after 24 h. During this period, the samples were immersed in deionized water (grade 3) and incubated at 37±1°C and 100% humidity for 24 h. The compressive strength was measured using a universal testing machine. Data were analyzed using two-way ANOVA followed by Tukey’s post-hoc test.
Results: The compressive strength of the five liners was significantly different (p < 0.05). Calcimol LC showed maximum compressive strength both immediately after setting and after 24 h. The compressive strength at 24 h was significantly higher than the primary compressive strength in all groups (p < 0.05).
Conclusion: Within the limitations of this study, it seems that Calcimol LC, Activa Bioactive Liner, and TheraCal LC have adequate compressive strength and can be used alone to provide adequate support for the restorative materials.- انتشار مقاله: 13-11-1398

- چکیده انگلیسی: Background: The clinical success of sandwich technique depends on the strength of resin-modified glass ionomer cement (RMGIC) bonding to both dentin and resin composite. Therefore, the shear bond strength (SBS) of resin composite bonded to RMGIC utilizing different resin adhesives versus a GIC-based adhesive was compared. Materials and methods: In this in vitro study, 84 holes (5×2 mm) were prepared in acrylic blocks, randomly divided into seven groups (n=12) and filled with RMGIC (Light-Cured Universal Restorative, GC). In the Group I; no adhesive was applied on the RMGIC. In the Group II, non-etched and Group III was etched with phosphoric acid. In groups II and III, after rinsing, etch-and-rinse adhesive (OptiBond Solo Plus); in the Group IV; a two-step self-etch adhesive (OptiBond XTR) and in Group V; a one-step self-etch (OptiBond All-in-One) were applied on the cement surfaces. Group VI; a GIC-based adhesive (Fuji Bond LC) was painted over the cement surface and cured. Group VII; the GIC-based adhesive was brushed over RMGIC followed by the placement of resin composite and co-cured. Afterward; resin composite (Point 4) cylinders were placed on the treated cement surfaces. The specimens were placed in 100% humidity at 37 ± 1°C and thermo cycled. The shear bond test was performed at a cross-head speed of 1 mm/min and calculated in MPa; the specimens were examined to determine mode of failure. The results were analyzed using one-way ANOVA and Tukey test. Results: The maximum (24.62±3.70 MPa) and minimum (18.15±3.38 MPa) SBS mean values were recorded for OptiBond XTR adhesive and the control group, respectively. The pairwise comparisons showed no significant differences between the groups that bonded with different adhesives. The adhesive failure was the most common failure mode observed. Conclusion: This study suggests that GIC-based adhesive could be applied over RMGIC as co-cure technique for sandwich restorations in lieu of employing the resin adhesives

- چکیده انگلیسی: Case Description- The peresent case described sialocele associated with sialoliths and ectopic ossification in a 3-year-old female terrier dog.
Clinical Findings- In the general examination, an enlargement fluctuate mass was seen blow the jaw around the neck. Clinical parameters (TPR) and blood test results were normal. The radiograph was showed the fluid opacity mass with some radioopaque particles within it. The fine needle aspiration and then the cytologic investigation proved that the mass was filled with saliva.
Treatment and Outcome- The surgical technique was the best treatment for removing the affected mandibular and sublingual salivary glands. In histopathologic study, no salivary acini or ducts were observed. The mass wall lined with epithelium tissue in some area. The well differentiated trabecular bones were present in the inner surface of cyst. Some sialoliths in 2-4 mm dimeter were identified in the mass.
Clinical Relevance- Salivary sialocele is a collected saliva in a cavity formed of connective tissue. The most affected animal is dog in any age specialy in male, and rare in other species.- انتشار مقاله: 24-10-1396

- چکیده انگلیسی: Background: Adipose-derived stem cells (ADSCs) are a population of pluripotent cells used for tissue engineering purposes. The main purpose of the present study was to transdifferentiate the ADSCs to Schwann-like cells and to determine the intensity of apoptosis in ADSCs during the transdifferentiation process. Methods: ADSCs were isolated from the inguinal adipose tissue of adult rats and the identity of the undifferentiated ADSCs was confirmed by the detection of specific cells surface markers. The ADSCs were transdifferentiated by sequential administration of beta mercaptoethanol, all-trans retinoic acid and a mixture of forskolin, beta fibroblast growth factor, platelet derived growth factor and hergulin. The immunocytochemical properties of transdifferentiated Schwann-like cells were examined at specified time point. RT-PCR was used to investigate the gene expression of the undifferentiated and transdifferentiated ADSCs. Cell apoptosis was assessed with annexin/propidium iodide staining and 3-(4, 5-Dimethylthiazol- 2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Results: Expression of Schwann cell marker S100 was determined by immunocytochemical staining. RT-PCR analyses revealed that the induced ADSCs exhibited Schwann cell-specific markers such as S-100, P75, and glial fibrillary acidic protein on the 14th day. MTT assay and flow cytometry studies showed that of the total ADSCs in the differentiation medium, 50% of the cells died by apoptosis, but the remaining cell population remained strongly attached to the substrate and continued to differentiate. Conclusion: ADSCs could differentiate to Schwann-like cells in terms of morphology and phenotype. An increased cell death rate was noted and the principle mode of cell death was apoptosis.

- چکیده انگلیسی: Objective(s): We intended to determine whether the ovarian varicose which is one of the common etiologies of the pelvic congestion syndrome, has the ability to interfere with the DNA methylation reprogramming in the oocyte and thereby affect the oocyte quality or not.
Materials and Methods: Varicose model was induced according to the Turner’s method in the rats. Briefly, a 20-gauge needle was placed on the left renal vein and a thread was tied over both the needle and the renal vein medial to the insertion of the ovarian vein, and then the needle was removed. Evaluation of prooxidant-antioxidant balance (PAB) was assessed using specific kits and the expression level of the DNA methyltransferase genes Dnmt1, Dnmt3a and Dnmt3L was assessed by Real-time PCR. Immunofluorescent staining for 5-methylcytosine in the oocytes evaluated the global DNA methylation.
Results: A significant PAB increase in the ovaries from varicose group was seen. Real-time PCR demonstrated a remarkable decrease in the expression of the Dnmt3a and Dnmt3L which are responsible for de novo DNA methylation in the oocytes. Immunofluorescent staining for 5-mC showed a reduction in the fluorescence intensity in the oocytes collected from the varicose group.
Conclusion: Our findings from Real-time PCR and immunocytochemistry suggest that the epigenetic parameters in the oocyte could be affected by varicose induction and these epigenetic alteration has the potential to affect the oocyte quality. We suggest that the epigenetic changes could happen in the oocytes after the induction of ovarian varicose and lead to the oocyte quality reduction or even infertility.- انتشار مقاله: 11-07-1396

- چکیده انگلیسی: Objective(s): Several researchers have reported the relationship between infertility in male and varicocele for so many years but the implication of varicocele in female patients is remains elusive. Here, we aim to examine the effects of unilateral varicose ovarian vein on antioxidant capacity and oocyte quality of rat ovary after the experimental creation of varicocele in female rats.
Materials and Methods: In this study, thirty adult female albino rats were divided into three equal groups: Group 1 as the control group has 10 rats, Group 2 as the sham group has 10 rats and they underwent a sham operation and finally Group 3 has the varicocele group has 10 rats. Antioxidant assays for superoxide dismutase, glutathioneperoxidase and catalase were performed using specific assay kits and gene expression for Bax, Bmp-15, Hsp-27 and Gdf-9 was done via real time PCR.
Results: The adverse effects of the experimentally induced varicocele were reported and recorded on the left ovary compared to the right sided ovary (no varicocele induction) in the varicocele group. Real time PCR data shows that the expression of Gdf-9, Hsp-27 and Bmp-15 genes were all significantly reduced at P≤0.05.
Conclusion: The results of this study show that reduced gene expression of Bmp-15, Gdf-9 and Hsp-27, increased gene expression of bax and an imbalance between pro-oxidant/ antioxidant ratio are few of the several mechanisms by which varicocele may lead to infertility in female.- انتشار مقاله: 10-06-1395

- چکیده انگلیسی: Objective(s):Although previous studies have confirmed the beneficial effects of human umbilical cord matrix stem cell (hUCM) transplantation post myocardial infarction (MI), but this stem cell resource has no potential to induce angiogenesis. In order to achieve the process of angiogenesis and cardiomyocyte regeneration, two required factors for cardiac repair agents were examined namely; hUCM and VEGF on an infarcted heart. The main objective of this research is to investigate the combinatory effect of dhUCM and VEGF transplantation on an infarcted heart.
Materials and Methods:45 min of ligating the left anterior descending coronary artery, the MI-induced
animals received 50 μl PBS, 5 μg VEGF, 5×106 hUCM cells alone, combined with 5 μg VEGF and 5×106 differentiated hUCM cells alone or combined with 5 μg VEGF through intramyocardial injection. MI group, without hUCM and VEGF served as the control group. Left ventricular function and angiogenesis
were also evaluated.
Results: After eight weeks post MI, there were significant rise in left ventricular ejection farction in dhUCM+VEGF group compared to the other treated and non-treated groups (P<0.05). Fibrosis tissue was markedly lower in the dhUCM+VEGF and hUCM+VEGF groups compared to the other treated and non-treated groups (P<0.05). Despite these benefits, vascular density in dhUCM+VEGF group was not markedly different compared to VEGF and hUCM+VEGF groups. The transplanted hUCM and dhUCM cells survived and migrated to the infarcted area.
Conclusion: Our findings demonstrated that the dhUCM cells transplantation combined with VEGF were more efficient on an infarcted heart.- انتشار مقاله: 09-04-1394

- چکیده انگلیسی: Objective(s): The present day challenge is how to obtain germ cells from stem cells to treat patients with cancer and infertility. Much more efforts have been made to develop a procedure for attaining germ cells in vitro. Recently, human umbilical cord-derived mesenchymal stem cells (HUMSCs) have been introduced with higher efficacy for differentiation. In this work, we tried to explore the efficacy of HUMSCs and some effective products of placental cells such as transforming growth factors. This study is aimed to optimize a co-culture condition for HUMSCs with placental cells to obtain primordial germ cells (PGCs) and reach into oocyte-like cells in vitro.
Materials and Methods: In this experimental study, HUMSCs and placental cells were co-cultured for 14 days without any external inducer in vitro. Then HUMSCs were assessed for expression of PGC markers; Octamer-binding transcription factor 4(OCT4),Tyrosine-protein kinase Kit (CKIT), Stage specific embryonic antigen 4 (SSEA4) , DEAD (Asp-Glu-Ala-Asp) box polypeptide 4( DDX4) and oocyte specific markers; Growth differentiation factor-9( GDF9), Zona pellucida glycoprotein 3(ZP3). The pertinent markers were assessed by immunocytochemistry and Q-PCR.
Results: Co-cultured HUMSCs with placental cells (including amniotic and chorionic cells) presented Oct4 and DDX4, primordial germ cells specific markers significantly, but increment in expression of oocyte-like cell specific markers, GDF9 and ZP3 did not reach to statistically significant threshold.
Conclusion: Placental cell supplementsTransforming growth factor (TGF α, β) and basic fibroblast growth factor (bFGF) in a co-culture model can provide proper environment for induction of HUMSCs into PGCs and expression of oocyte-like markers.- انتشار مقاله: 06-11-1393

Recent reports suggest that opium sellers cheat their customers by adding lead to the opium. Contaminated opium can threaten the health of consumers. The present study aimed to compare the serum level of lead between opium dependents and a control group. Methods: This was a cross-sectional study in which 50 opium dependents aged 20 to 60 years old were compared with a control group of 43 non-dependents who were matched with the case group in terms of sex and age. The serum level of lead and liver function tests including serum total bilirubin, AST, ALT, Alkaline-phosphatase and hemoglobin were measured for all subjects. Findings: The mean level of serum lead concentration in opium dependents and controls was 3929.358 ± 147.67 and 3532.721 ± 1141.53, respectively and the difference was not statistically significant. There was no significant correlation between serum level of lead and age, duration of opium dependency, serum total bilirubin, hemoglobin, AST, ALT, and Alkaline-phosphate. Conclusion: Although there was no significant relationship between opium consumption and serum level of lead, the concentration of lead in dependents' serum was higher than controls. Further studies are needed to approve this relationship to be used for screening and on time diagnosis of opium dependents. Keywords: Opium, Addiction, Lead poisoning, Serum level.
