- چکیده انگلیسی: Objective(s): Despite the good results of anticancer activities by curcumin, there are some hurdles that limit the use of curcumin as an anticancer agent. Many methods were examined to overcome this defect like the use of the dendrosomal curcumin (DNC). There is increasing evidence that miRNAs play important roles in biological processes. In this study, we focus on the roles of microRNA-21 in the anti-cancer effects of DNC in breast cancer.
Materials and Methods: Also, we have used different methods such as MTT, apoptosis, cell cycle analysis, transwell migration assay and RT-PCR to find out more.
Results: We observed that miR-21 decreased apoptotic cells in both cells (from 6.35% to 0.34 % and from 7.72% to 1.32% orderly) and DNC increased it. As well as, our findings indicated that cell migration capacity was increased by miR-21 over expression and was decreased by DNC. The combination of miR-21 vector transfection and DNC treatment showed lower percentage of apoptotic cells or a higher level of penetration through the membrane compared with DNC treatment alone. Furthermore, DNC induced a marked increase in the number of cells in sub G1/G1 phase and a decrease in G2/M phase of the cell cycle in both; but, we observed reverse results compared it, after transfection with miR-21 vector.
Conclusion: We observed that miR-21 suppress many aspects of anti-cancer effects of DNC in breast cancer cells, it seems that co-treatment with DNC and mir-21 down-regulation may provide a clinically useful tool for drug-resistance breast cancer cells.- انتشار مقاله: 23-01-1396

- چکیده انگلیسی: Many kinds of mutations in mitochondrial (mt) DNA have been reported to be related to the development of Diabetes Mellitus (DM), this type of diabetes has also been shown to be influenced by other genetic factors and/or environmental factors. Among them, tRNALeu(UUR) and its adjacent mtDNA NADH dehydrogenase subunit 1(ND1) region within the mt genome are linked to high susceptibility to DM. A point mutation at 3243 base pair (bp) in the mt tRNA Leu(UUR) is commonly referred to as a syndrome of mitochondrial myopathy, Encephalopathy, Lactic acidosis, and Stroke-like episodes (MELAS). In the current study, we have assessed
the frequency of the A3243G in Iranian diabetic type 2 patients. DNA was obtained from peripheral leukocytes of 154 patients with diabetes Mellitus type2 (l50 with type 2 and 4 with gestational diabetes) and 40 control subjects. Insulin concentration from patients’ blood was measured using Radioimmunoassay procedure. Patients showed fasting blood sugar (FBS) between 150-230 mg/dl, body mass index (BMI) between 19-32 Kg/m2 and insulin concentration 0.9-2.35 mg/ml. PCR-RFLP, single strand conformation polymorphism
(SSCP) and sequencing methods were used to detect the A3243G or other mutations in the mitochondrial
tRNALeu (UUR) gene. A3243G mutation was not detected in patients. SSCP results showed a new pattern of PCR product in 6 patients. The C3316T transition mutation in the ND1 mitochondrial gene was confirmed in selected samples (n=6) by sequencing. No differences were observed between the two groups for C3316T and A3243G mutations (P=0.348). The mt C3316T mutation did not have any effect on the clinical finding of type 2 diabetes carrying this mutation. These data together with clinical characteristics of the patients may

- چکیده انگلیسی: The D-loop region is a hot spot for mitochondrial DNA (mtDNA) alterations, containing two hypervariable segments, HVS-I and HVS-II. In order to identify polymorphic sites and potential genetic background accounting for Hypertrophic CardioMyopathy (HCM) disease, the complete non-coding region of mtDNA from 31 unrelated HCM patients and 45 normal controls were sequenced. The sequences were aligned upon the revised Cambridge Reference Sequence (rCRS) and any incompatibilities were recorded as numerical changes in homoPolymeric C Tract (PCT), single base substitutions, insertions and deletions (Indels). Nucleotide substitutions were found to make up the majority of the mutations, rather than indels. We drew significantly
high transition rate (81.8%) versus lower frequency of transversions (18.2%). 12 polymorphisms were identified in this study which had not been published in the MitoMap database. PCT changes at position 303-309 were detected in 83% of our samples. Our results suggest that an increased level of HVS-I and HVS-II substitutions may be an indicator of mitochondrial DNA instability. Furthermore, mtDNA mutations
may play an important role in pathogenesis of cardiac arrest which has remained unexplained for long.

- چکیده انگلیسی: This paper reports on the findings of a study designed to investigate the cultural and social complexities governing the compliment responses among the Persian native speakers. 200 Persian respondents took a 24-item Discourse Completion Task (DCT) while 15 native field workers were also set responsible for collecting the examples of complimenting exchanges they either observed or participated in. The results suggested a significant effect for the treated intervening social variables of age, gender, educational background, social distance, and relative power as well as compliment topics in determining the type of compliment response. The responses were further suggestive of the contextual effects of the three systems of hierarchical, solidarity and deferential as well as a newly coined system as kinship system.

- چکیده انگلیسی: Liver is the largest and most vital organ in human body and able to detoxify several chemical compounds. Acute liver failure (ALF) is a rare devastating process that can leads to urgent liver transplantation. In this study we introduce 3 children of a family with an accidental intoxication of Cassia Floribunda which causes ALF. The first case is a four-year-old girl with an icter's complaints and drowsiness after eaten plant in the garden while playing with her sister and friend 3 days ago. At the beginning her GCS was 10, vital signs were normal, sclera was icteric and pupil has normal size and was reactive to the light. Initial tests showed leukopenia, anemia, thrombocytopenia, increase of liver enzymes , coagulopathy , hypoglycemia. With diagnosis, fulminant liver deficiency and hepatic encephalopathy, the patient transferred to PICU and GCS reduced. her general appearance became better ,consciousness increased, lab test findings improvedand and discharged after 11 days.Second and third cases presented to emergency with similar complaints without icteric, vital signs and other examinations were normal . Initial tests reported reduced platelets, increase liver enzymes , Without hypoglycemia. treatment was started for patient by intravenous ranitidine and monitored in emergency . Two days after discharged. But in third case platelets increased.ALF in children is a serious problem that can lead to death or need to liver transplantation. When evaluate the causes of ALF, poisoning due to plants should be kept in.
