Recent investigations have examined the titanium dioxide nanoparticles (TiO2 NPs) in biomedical applications, mostly in the cancer treatment field. This study aimed to evaluate the effects of simulated microgravity combined with TiO2 NPs in MDA-MB-231 cells proliferation for the first time. In other words, this study examined the utility of the microgravity environment in nano-therapy.
Materials and Methods: The MDA-MB-231 human breast cancer cell line and TiO2 NPs were purchased. The 2D clinostat was applied for the simulation of the microgravity. The morphological studies, MTT cytotoxicity assay, Acridine orange/Ethidium bromide double staining studies and flow cytometry analysis were utilized.
Results: The MTT assay, the morphological studies, Acridine orange/Ethidium bromide double staining studies and flow cytometry analysis confirmed the apoptosis-inducing effect of microgravity in combination with TiO2 NPs. The IC50 of simulated microgravity in the presence of TiO2 NPs was determined to be 130 µM. Furthermore, MDA-MB-231 cells exposed to microgravity adopted a different phenotype.
Conclusion: Based on our observation, although the relative mechanisms need to be explored further, microgravity can strictly affect the TiO2 NPs effects on MDA-MB-231 cells. The significance of this study lied in the fact that simulating microgravity can be a powerful physical cure for cancer therapy and open new horizons for the studies in the field of biology, biophysics, and medicine.- انتشار مقاله: 24-11-1397

- چکیده انگلیسی: Background and objectives: The goal of this study was to compare the cytotoxic and antioxidant activities of the corms extracts of two Crocus species (C. sativus L. and C. pallasii subsp. haussknechtii). Methods: n-Hexan, chloroform and methanol extracts of the corms were prepared. Cytotoxic activities of the extracts against human breast cancer cell lines MDA-MB-231 and MCF-7 were evaluated via MTT assay and dual acridine orange/ethidium bromide fluorescent staining test. Antioxidant activities of the extracts were assessed by DPPH method. Results: The results showed that the methanol extracts of the corms from both species had cytotoxic activity against the breast cancer cells. Cytotoxic activity of the corms extract of C. sativus L. was significantly higher than C. pallasii subsp. haussknechtii. n-Hexan and chloroform extracts from the corms of C. pallasii subsp. haussknechtii and Crocus sativus L. showed no anti-proliferative activity which indicated the polar nature of the cytotoxic agent(s) in the extracts. The results of dual acridine orange/ethidium bromide fluorescent staining test revealed that the methanol extracts of both species corms destroyed cancer cells through apoptosis. Moreover, the methanol corm extracts of the two species showed little antioxidant activity. Conclusion: It can be concluded that C. sativus L. may possess more significant apoptotic properties compared to C. pallasii subsp. haussknechtii.

- چکیده انگلیسی: Background and objectives: Salvia suffruticosa is a perennial plant from Lamiaceae family. Many Salvia species have been employed as medicinal plants; despite the medicinal potentials of S. suffruticosa, there is limited studies regarding its phytochemical profile or biological properties. The aim of the present study was to investigate the chemical constituents of the essential oil and extract of the plant and evaluate its biological activities. Methods: Essential oil from the aerial parts of the plant was extracted by hydrodistillation and analyzed using gas chromatography/mass spectroscopy. Isolation of compounds from methanol and petroleum ether fractions was achieved by using column chromatography with different stationary phases. The structures of the isolated compounds were elucidated by NMR techniques. Cytotoxicity potentials were evaluated using MTT assay and acridine orange/ethidium bromide staining method. Antioxidant activity was assessed by DPPH method. Results: Hydrocarbon sesquiterpenes were identified as the predominant components of the oil, with β-caryophyllene (27.35%), bicyclogermacrene (22.15%), germacrene-D (9.49%) and β-farnesene (9.08%) as the major constituents. Phytochemical analysis of the extract resulted in isolation of lupeol (1), β-sitosterol (2), stigmasterol (3), caffeic acid (4) and 1-feruloyl-β-D-glucopyranose (5). Among the tested samples, lupeol demonstrated the most potent inhibitory activity toward breast cancer cell lines including MCF-7, T-47D and MDA-MB-231 with IC50 values equal to 33.38±2.6, 36.70±3.1 and 23.66±1.4 μg/mL, respectively; caffeic acid with IC50 value of 12.1±1.2 μg/mL showed the most potent radical scavenging activity. Conclusion: The results of this study suggested S. suffruticosa as a promising source of bioactive compounds useful in prevention and treatment of cancer.

- چکیده انگلیسی: Chromanone derivatives demonstrate remarkable cytotoxity against a varieties of cancer cell lines. Novel 9-[hydroxy(substitutedphenyl)methyl]-2,2-dimethyl-2,3,8,9- tetrahydro-4H,10H-pyrano[2,3-f]chromene-4,10-diones as Glyasperin analogues were synthesized in four steps from known 4-chromone 1. The key step was the preparation of chromane dione 5a by regioselective intramolecular cyclization reaction in 85% yield. Condensation of 5a with substituted aromatic aldehydes afforded corresponding alpha hydroxybenzyl analogues 6a-6e. The cytotoxic study of the synthesized compounds against breast cancer human cell line (T47D) showed moderate cytotoxic activities (IC50=16-40 μM) compared to the positive control drug vincristin (IC50=2.5 μM).

Microalgae biomass production and optimization is necessary to overcome food security problems and address environmental topic as well as high-value compounds production. In this investigation the influence of three culture media (BBM, BG-11 and Guillard f/2) on the growth and biomass production of the Picochlorum sp. RCC486 microalgae was evaluated.
One- way ANOVA with Tukey’s HSD and Fisher’s LSD tests were used to test the effect of different culture media on optical density. Then the maximum optical density and biomass production by Picochlorum sp. RCC486 was evaluated using response surface methodology. A historical data of response surface methodology was used to find the optimum response, which yields the highest biomass production.
Microalgal growth in Guillard f/2 medium reached highest cell count in the considered time period and the microalgae presented highest biomass production in comparison with other two culture media (BBM and BG-11). Additionally, biomass productivity(58 mg L-1d-1 ), specific growth rate (0.681d-1), dry weight (1.516 g L-1) and cell density(52.9 ×106 cells mL-1) were highest in comparison with other two culture media. The predicted optical density and biomass production using historical data design were close to their experimental values corresponding to 1.50313 and 1.976gL-1 respectively. while the concentrations of nitrogen source and phosphorus source were achieved in concentrations of 1345.55 mg L−1, 6.983 mg L−1 respectively. Under the optimum condition, the biomass yield was1.961 gL-1 which is 0.76% less than the predicted value. Also the study reported that the biomass production of Picochlorum sp. RCC486 improved from 1.516 g L-1 in defined Guillard f/2 medium to 1.961 g L-1 in modified Guillard f/2 medium resulting 22.69% increase. Thus, Picochlorum sp. RCC486 biomass content could be enhanced by optimizing nitrogen and phosphorus concentrations.- انتشار مقاله: 10-05-1397
