- چکیده انگلیسی: Introduction: burn wounds are common happenings which cause many people have irreversible defects and to be death around the world every year. In this study, we evaluated the effects of Melilotus officinalis and Alpha ointment in the healing of burn wounds and compare their findings with silver sulfadiazine (SSD).Methods: Seventy-five male Sprague-Dawley rats were randomly divided into five groups, and similar burn ulcers were produced on the back of rats. The first group of rats received no treatment, base gel was used topically to group II, in groups III-V, Alpha, SSD and Melilotus officinalis preparations were applied, respectively. Wound healing, contraction and histopathological examination were evaluated at the end of 7, 14, and 21 days.Results: Alpha ointment was equally effective as Melilotus officinalis gel, while its efficacy was better compared to SSD for all parameters of wound healing on days 7, 14 and 21. Alpha and Melilotus officinalis preparations as less expensive medications which significantly improve the quality of wound healing and scar formation and also are more appropriate treatment choices than SSD.Conclusion: Alpha and Melilotus officinalis are recommended as alternative to SSD, especially in patients with low economical backgrounds or in those who show adverse reactions to SSD.

- چکیده انگلیسی: Valproic acid (VPA) is a widely administered drug against epilepsy and several other neurological disorders. On the other hand, liver injury is a deleterious side effect associated with VPA. Oxidative stress seems to play a critical role in VPA-induced hepatotoxicity. The current investigation was designed to evaluate if N-acetylcysteine (NAC) and dithiothreitol (DTT) as thiol reducing agents have any protective effects against VPA-induced liver injury. Isolated rat hepatocytes (in vitro) were exposed to increasing concentrations of VPA (25, 50, 100, 150, and 250 µM) and markers of cytotoxicity were evaluated. Furthermore, animals received VPA (250 and 500 mg/kg, i.p for 15 consecutive days) (in vivo) and markers of liver injury were monitored. It was found that 250 µM of VPA caused marked cytotoxicity toward isolated hepatocytes as judged by trypan blue exclusion test. Moreover, markers of oxidative stress including glutathione depletion and lipid peroxidation were detected in VPA-treated hepatocytes. On the other hand, VPA caused a significant increase in plasma markers of hepatotoxicity in drug-treated group. Liver histopathological changes and markers of oxidative stress were also detected in VPA-treated animals. It was found that administration of NAC (1 mM), and DTT (1 mM) significantly alleviated VPA-induced cytotoxicity (In vitro). NAC (250 and 500 mg/kg) and DTT (15 and 30 mg/kg) also significantly mitigated VPA hepatotoxicity (In vivo). The data obtained from the current investigation indicate potential therapeutic properties of thiol reductants against VPA-induced liver injury.

- چکیده انگلیسی: Omeprazole (OMP) and ketoconazole (KTZ) have been shown to activate the AHR signaling pathway in spite of the fact that they bind to the receptor with low or no affinity. The aim of this study was to investigate whether KTZ and OMP can act as indirect activator of AHR. In order to evaluate the effects of KTZ and OMP on AHR signaling, we measured cytochromes p450 (CYP1A1) enzyme activity by ethoxyresorufin-O-deethylase (EROD) assay as endpoint. FICZ, at 1nM concentration, caused a transient elevation in the catalytic activity of CYP 1A1. KTZ and OMP were found to be inducer of CYP1A1 at concentrations above 50 µM. At early time of incubation (3hr), a dose-dependent inhibition of FICZ-induced EROD activity was seen. When OMP or KTZ were added together with FICZ, a prolonged activation of CYP1A1 was observed at later time of incubation (24h). Taken together, our findings support the earlier observation that we shown that CYP 1A1 inhibitors can act as an AHR activator though inhibition of metabolic degradation of FICZ.

- چکیده انگلیسی: Statement of the Problem: Oral mucositis (OM) is a common side effect of anti-cancer drugs and needs significant attention for its prevention.Purpose: This study aimed to evaluate the healing effects of olive leaf extract on 5-fluorouracil-induced OM in golden hamster.Materials and Method: OM was induced in 63 male golden hamsters by the combination of 5-fluorouracil injections (days 0, 5 and 10) and the abrasion of the cheek pouch (days 3 and 4). On day 12, hamsters were received topical olive leaf extract ointment, base of ointment, or no treatment (control) for 5 days. Histopathology evaluations, blood examinations, and tissue malondialdehyde level measurement were performed 1, 3 and 5 days after treatments.Results: Histopathology score and tissue malondialdehyde level were significantly lower in olive leaf extract treated group in comparison with control and base groups (p= 0.000). Significant decreases in white blood cell, hemoglobin, hematocrit , and mean corpuscular volume and an increase in mean corpuscular hemoglobin concentration were observed in olive leaf extract treated group in comparison with control and base groups (p< 0.05).Conclusion: Our findings demonstrated that daily application of olive leaf extract ointment had healing effect on 5-fluorouracil induced OM in hamsters.
