- چکیده انگلیسی: Background: Human CD34 is a transmembrane glycoprotein which is expressed in human hematopoietic stem cells (HSCs) and the small- vessel endothelial cells of a variety of tissues. CD34 plays a critical role as a marker for diagnosis and classification of leukemia. Anti CD34 antibodies are used for isolation and purification of HSCs from bone marrow, peripheral blood and cord blood.
Objective: To characterize a newly produced monoclonal antibody against a human CD34 peptide.
Methods: Anti CD34 monoclonal antibody (Clone 2C10-D3) was purified from mouse ascitic fluid and hybridoma cell culture supernatants by affinity chromatography and its immune reactivity was examined by ELISA. The purified antibody was further characterized using Western blot and flow cytometry on TF1 (Human Erythroblast) cell line.
Results: ELISA experiment revealed that the antibody recognized CD34 peptide. Western blot analysis on TF1 cell lysate confirmed the reactivity of the antibody with a 42 KDa protein. Blocking the antibody with a saturating concentration of specific CD34 peptide resulted in loss of its activity with TF1 lysate in Western blot. The 2C10-D3 antibody reacted with TF1 cells in flow cytometry in a similar manner to a commercial anti CD34 monoclonal antibody.
Conclusions: Our data suggest that the anti CD34 monoclonal antibody (Clone 2C10-D3) is an appropriate antibody to study the CD34+ cells by flow cytometry and Western blot.- انتشار مقاله: 15-05-1395

- چکیده انگلیسی: Background: Nasal continuous positive airway pressure (NCPAP) is an effective method of ventilation in newborns with respiratory distress syndrome (RDS). Using an appropriate nasal skin care protocol is identical to prevention or reduction of nasal skin breakdown in those who receive NCPAP. This study aimed to investigate the effectiveness of an evidence-based clinical care protocol on nasal skin integrity in preterm newborns who receive NCPAP.
Materials and Methods: A Randomized Controlled Trial was used to conduct the study. A cohort of 110 preterm newborns with a gestational age (GA) of 25 to 36 weeks, receiving nasal continuous positive airway pressure (NCPAP) for RDS in the neonatal intensive care unit of a university teaching hospital were selected to perform the study. They were randomly assigned to a protocolized nasal skin care (group A) or to a group receiving the routine care (group B). Nasal skin integrity of the preterm neonates, were measured on a daily basis for 10 days using the Neonatal Skin Condition Scale (NSCS) 24 hours after placement of NCPAPs in both groups.
Results: Each intervention and control group included 55 neonates. 65.50% of neonates in the control group and 47.30% of neonates in the intervention group were male. Repeated measures analysis showed thatNSCS scores were significantly lower in intervention group receiving nasal skin care in accordance with the protocol than the control group receiving the routine nasal skin care (P=0.000).
Conclusion: In this study, the protocolized care reduced nasal skin breakdown in the preterm newborns receiving NCPAP. Therefore, it can be used as an effective method in nasal skin care in neonates who are treated by NCPAP.- انتشار مقاله: 26-08-1395

Born of a child with hearing loss is a stressful and unexpected experience for the parents. Understanding the experience of the mothers of children with hearing loss is a crucial to provide an appropriate care for this group of mothers.This study aimed to understand the concerns of the mothers who have a child with congenitally hearing loss.
Materials and Methods
In this qualitative content analysis, 35 mothers with child hearing loss participated in the semi-structural and deep interviews through a purposeful sampling method. Data were analyzed by using qualitative content analysis provided by Graneheim et al.
Results
Concerns of mothers of children with hearing loss were categorized in three main themes: ‘the concerns of nature of hearing loss’, ‘the social concerns’ and ‘concerns about future’. Remarkably, the participants live with fear of the possibility of another deaf child birth in the next pregnancy or next generations were the most important concern of mothers.
Conclusion
Mothers of deaf children are experiencing the varieties of concerns. To cope with these concerns, they need a special consideration by society and health professionals. Modification of the concerns can decrease the child-related stress and consequently encourage them to involve in child care.- انتشار مقاله: 15-01-1396
