- چکیده انگلیسی: κ-casein is a glycosilated protein in mammalian milk that plays an essential role in the milk micelles. Control of κ-casein expression reflects this essential role, although an understanding of the mechanisms involved lags behind that of the other milk protein genes. Transcriptional regulation, a first mechanism for controlling the development of organisms, is carried out by transcription factors binding sites (TFBSs). We determined the 5’ region for Iranian Dromedaries and bactrianus camel κ-casein gene. We also determine their regulatory TFBSs regions. This region contained 14 potential TFBSs The four of them C/EBP-α, Oct1, MGF/STAT5 and TPB have almost perfectly conserved.

- چکیده انگلیسی: Brucellosis is a zoonotic disease in human and animals. Brucella melitensis is one of the most pathogenic species of Brucella in goat and sheep. Omp31 is an outer membrane protein of Brucella that acts as an immunogenic protein. Cytokines are glycoproteins with low molecular weight that play the role of an immune adjuvant and regulate immune responses. Interleukin-2 is one of the most important cytokines, which are secreted by the white blood cells and involved in T cell immune responses. In the present study, a chimeric Omp31-Interleukin2 recombinant protein was generated by means of genetic engineering techniques. This chimeric coding sequence was amplified by using specific primers and using Splicing Overlap Extension (SOE) PCR technique. The fusion of the two mentioned proteins was accomplished using a rigid linker. The generated chimeric IL2-Omp31 fragment was TA cloned, and then subcloned into pEt22b vector as an expression vector. The chimeric protein was successfully expressed in E. coli BL21 (DE3) and confirmed by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and also Western-blotting analysis. Finally, in order to assess the antigenic features of the recombinant chimeric IL2-Opm31 protein, its secondary structure and antigenicity were predicted in silico.

- چکیده انگلیسی: Foot-and-mouth disease (FMD) is a severely contagious viral disease that mainly affects cloven-hoofed
livestock and wildlife. This study quantifies the cytokines mRNA expression of vaccinated guinea pigs with
FMD type O inactivated vaccine. Blood samples were collected from eight guinea pigs at 7 and 28 days after the
first vaccination. Extracted mRNAs were reverse-transcribed into cDNA and analyzed for quantification of IFN-
γ, TNF-α and IL-10 expression using relative real-time PCR assay. Our results showed that all of the genes were
upregulated. The expression of TNF-α and IL-10 genes significantly increased (P<0.05) in day 28th in
comparison to the day 7th post the first vaccination. It can be concluded that the vaccine induced immune
responses by increasing expression of the cytokines. Therefore, effects of DNA vaccines on immune system also

- چکیده انگلیسی: Brucellosis is a well-known infection in domestic animals which caused by Brucella bacterium. Due to
serious economic and medical consequences of this disease, various efforts have been made to prevent the
infection through the use of recombinant vaccines based on Brucella outer membrane protein (OMP)
antigens. The objectives of the present study were cloning, sequencing and epitope prediction of Omp25
and BLS genes as two major Brucella melitensis antigens. The full-length open reading frame (ORF) of
Omp25 and BLS genes were amplified and cloned into pTZ57R/T vector. Phylogenetic analysis of
sequenced genes showed that both genes were nearly similar in different Brucella species. Several online
prediction softwares were used to predict B and T-cells epitopes, secondary and tertiary structures,
antigenicity ability and enzymatic degradation sites. Bioinformatic tools used in the current study were
confirmed by the results of three different experimental epitope predictions. Bioinformatic analysis
identified five and two B-cell and two and one T-cell epitopes for Omp25 and BLS antigens, respectively.
Finally, according to the antigenicity ability and proteosomal recognition site common B and T-cell epitope
was predicted for Omp25 (154-162 amino acids) and BLS (37-48 and 119-139 amino acids). Results of this
