سگهای مبتلا به لیشمانیوز احشایی از مدتها پیش به عنوان مهمترین مخازن بیماری ناشی از لیشمانیا اینفانتوم شناخته شده اند. این مطالعه به منظور ارزیابی شیوع سرمی عفونت ناشی از لیشمانیا اینفانتوم در سگهای بدون علایم بالینی در مناطق مرکزی و جنوب غربی ایران و ارزیابی فاکتورهای خطر همراه این بیماری انجام شد. به این منظور، اقدام به تهیه نمونههای سرمی از 548 قلاده سگ از سه استان واقع در این مناطق و ارزیابی این نمونهها با رقت 1:80 تا 1:20480 با استفاده از تست آگلوتیناسیون مستقیم (DAT) گردید. 53 مورد از سگها (67/9%)، آنتیبادی ضد لیشمانیا اینفانتوم را در رقتهای سرمی حد اقل 1:80 نشان دادند. شیوع سرمی عفونت در سگهای خانگی به طور معنیداری کمتر از میزان آن در سگهای آزاد (نگهبان یا ولگرد) بود (P<0.001). میزان عفونت در سگهای مسنتر از دو سال، به طور معنیداری در مقایسه با سگهای جوانتر بالاتر بود (P<0.001). اختلاف معنیداری در شیوع سرمی عفونت در سگهای نر یا ماده مشاهده نشد. مطالعه حاضر، نشان دهنده عفونت نسبتا بالای ناشی از لیشمانیا اینفانتوم در سگها در مناطق مورد بررسی بوده و اهمیت بررسی سگهای آزاد را به عنوان مخازن عفونت مشخص میسازد. بالا بودن میزان عفونت در سنین بالا و در سگهای ولگرد، احتمال آلودگی سگها را در مواجهه با آلودگیهای محیطی و مخازن دیگر بیماری نشان میدهد.

Clinically infected dogs have been identified as the main reservoir hosts of visceral leishmaniasis (VL)
caused by Leishmania infantum. Recently asymptomatic infected dogs were regarded to be as important as
clinically ill dogs. This study was aimed to determine the seroprevalence of L. infantum infection among
asymptomatic dogs in the southwest and central parts of Iran and to investigate possible risk factors
associated with this infection. 548 serum samples were collected from dogs in three Iranian provinces and
subjected to direct agglutination test (DAT) in dilutions of 1:80 to 1:20480. Fifty three (9.67%) of the dogs
had detectable anti-L. infantum antibodies at dilutions of ≥1:80. Living status of the dogs (household or free roaming) was a potential risk factor for the infection; seroprevalence was significantly higher in free roaming dogs (P<0.001). Dogs of more than 2-year-old had a significantly higher infection rate in comparison with younger dogs (P<0.001). No significant statistical differences were seen between seroprevalences of the male and female dogs. The results of this study show relatively high sero-prevalence of L. infantum infection in evaluated regions and higher seroprevalence in old and free roaming dogs, which shows the importance of environmental contamination and access of the dogs to the other reservoir hosts.- انتشار مقاله: 27-07-1389

- چکیده انگلیسی: Ocular lesions are a common manifestation of toxoplasmosis in cats. In this survey, 10 healthy Syrian
mice were infected through intraperitoneal inoculation of bradyzoites of Toxoplasma gondii (Tehran strain).
Mice were killed 30 days after inoculation and the suspension of their brains was prepared, after confirmation
of the infection. Seven kittens were fed with the infected mice brain suspension. The kittens were 2.5–3
months old and had body weights of 650–900 g. All kittens shed oocyst 7–8 days after infection, and had
antibody titers as high as 1/320–1/1280 30 days after the challenge. Serum chemistry and complete blood
count were determined in all of the kittens. Examination of eyes was done with ophthalmoscope twice during
one week. Only one kitten showed unilateral anterior uveitis on day 16 after the infection was diagnosed

- چکیده انگلیسی: Mediterranean visceral leishmaniasis is a zoonotic disease caused by Leishmania infantum and transmitted by the bites of infected female sand flies. Iran is one of the endemic areas of this disease. Dogs and canines are the major reservoir hosts of Leishmania infantum in the new and old world, including Iran. If visceral leishmaniasis is left untreated, it may result in a 90% mortality rate. The identification and elimination of infected dogs are efficient ways to control this disease. The diagnostic methods used to identify these animals cannot yield 100% detection. Therefore, in the present study, we used a multiepitope recombinant protein (PQ10) to distinguish between symptomatic and asymptomatic infections caused by Leishmania infantum in animal reservoirs (dogs). The coding sequence of PQ10 recombinant protein was sub-cloned in pET28 expression vector and was commercially synthesized by GENERAY Biotechnology, China. In the following process, sequencing was performed with plasmid-specific primers and followed by the expression, optimization of expression. The purified recombinant protein was confirmed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot. The efficacy of recombinant PQ10 protein was evaluated by an indirect enzyme-linked immunosorbent assay (ELISA) test using 150 dog sera (25 symptomatic positive sera, 25 asymptomatic positive sera, 80 negative sera, and 20 sera of dogs with other infectious diseases). Direct agglutination test (DAT) as the standard method was used to compare and determine specificity and sensitivity. The results indicated that the sensitivity and specificity of ELISA using infected dog sera were 94% and 86%, respectively. Positive and negative predictive values for dog sera were reported as 87.03% and 93.47%, respectively. This protein was able to identify 92% of asymptomatic dogs with visceral leishmaniasis. The results showed that the recombinant protein PQ10 is able to identify positive cases of canine visceral leishmaniasis, especially asymptomatic cases.
