پروتئین PB2 ویروسهای آنفلوانزای A نقش مهمی در تعیین طیف میزبانی آنها دارد. در این مطالعه بخشی از ژن PB2 ویروسهای آنفلوانزای H9N2 جدا شده از جوجه مرغهای تجاری در استان تهران تکثیر و توالی نوکلئوتیدی و اسید آمینهای آنها تعیین گردیدند. تجزیه و تحلیل و مطالعه شجرهشناسی این توالیها و مقایسه آنها با برخی توالیهای موجود در بانک جهانی ژن از ویروسهای H9N2 انجام گردید. مطالعه شجرهشناسی به روی توالیهای ژن PB2 استفاده شده در این مطالعه نشان میدهد که جدایههای H9N2 ایران را میتوان در دودمان اروپایی-آسیایی طبقهبندی نمود؛ همان طوری که بر اساس نتایج حاصل از مطالعههای انجام شده به روی ژن HA نیز مشخص شده است. در پروتئین PB2 جدایههای H9N2 ایران در این مطالعه در محل 621 گلوتامیک اسید، در 701 آسپارتیک اسید و در 714 سرین قرار دارد. وضعیت مشابهی در مورد تحت تیپهای مختلف جدا شده از گونههای پرندگان در سطح دنیا نشان داده شده است. منشاء ژن PB2 جدایههای ایرانی متفاوت از امارات متحده عربی، پاکستان، عربستان سعودی (کشورهای خاورمیانه) و هند میباشد و احتمال دارد که اجداد ایزولههای H9N2 ایرانی دچار بازآرایی با سایر تحت تیپها شده باشند. توالی نوکلئوتیدی ژن PB2 این پنج جدایه ایرانی تشابه بالایی در حدود 5/97% تا 6/99% با یکدیگر دارند که نشان میدهد طی سالهای 1998 تا 2001 جهشهای بسیار کمی در آنها رخ داده است. مطالعه ژن PB2 در جدایههای سالهای اخیر از سایر استانها جهت بررسی تغییرات ممکن در آن و نیز مطالعه بیماریزایی جدایههای ایرانی در میزبان پستاندار قابل توصیه میباشد.

The PB2 protein of influenza A viruses has been shown as host range determinant. In this study PB2
gene of five H9N2 isolates from Tehran province during the period of 1998-2001 were partially amplified
and sequenced to do phylogenetic study. Iranian isolates could be classified in Eurasian lineage according to
their PB2 gene sequences. The PB2 genes of these isolates are different from those of Middle-East countries
such as the UAE, Pakistan and Saudi Arabia, as well as India, and it is probable that their ancestors have
undergone reassortment with other subtypes. The Nucleotide sequences of their PB2 are highly similar to
each other with 97.5-99.6% homology percentage showing the least mutations in their genes during the
period of 1998 to 2001. The PB2 of the Iranian isolates contain Glutamic acid-(E) in position 627, Aspartic

- چکیده انگلیسی: Bordetellosis or turkey coryza, caused by Bordetella avium, has been an issue for turkey industry since its first description in 1967 when it was reported for the first time. Bordetella avium causes a highly contagious upper respiratory disease in turkeys. Therefore, this study aimed to isolate and characterize this species from commercial and backyard turkeys in Tehran, Isfahan, and Northern provinces of Iran. For the purpose of the study, 625 tracheal swabs were taken from 425 commercial poults and 200 backyard poults aged 2-6 weeks from September 2016 to September 2018. The swabs were immediately plated on MacConkey and blood agar plates and then pooled (5 swabs/pool) in tubes, containing 2 mL distilled water, to perform direct polymerase chain reaction (PCR) for the identification of B. avium. A total of 17 swab pools were found to be positive for B. avium. A subset of seven positive samples were sequenced for the flanking region of piuA gene. The analysis of the sequences indicated that the sequences were 98%, 96%, and 98% similar to B. avium 197N (AM167904.1), 4142 (AY925058.1), and 4156 (AY925068.1) sequences, respectively. To the best of our knowledge, the current study is the first attempt toward the molecular detection and characterization of B. avium in Iran. It is highly recommended to perform further studies to isolate, characterize, and differentiate the regional isolates in order to help the developing turkey industry of Iran meet the increasing demands for protein in the diet of the citizenry.
