- چکیده انگلیسی: Allium Jesdianum (AJ) is the native plant mostly grown in Middle East region that has the excellent pharmacological properties. In this study, we evaluated the hepatoprotective effect of hydroalcoholic extract of AJ on liver injury induced by Bromobenzene (BB) in male mice. Animals were randomly divided into five groups, control group received normal saline plus olive oil, groups 2-4 received(500, 1000 and 2000 mg/kg) AJ extract plus BB for 5 days and 5th group received BB (460 mg/kg). On the fifth day all groups received hexobarbital sodium (25 mg/kg, i.p). it should be noted that sleeping time of all mice were recorded. After 24 hours mice were sacrificed. By serum and tissue biomarkers such as alanine aminotransferase (ALT), aspartate aminotransferase(AST), reduced glutathione(GSH), malondialdehyde(MDA) and histopathological studies were measured. The results showed that BB significantly increased sleeping time, ALT, AST and MDA levels besides and decreased GSH level compared with control group. AJ extract at doses1000and 2000 mg/kg showed a significant alter in all studied endpoints and dose 2000 mg/kg showed a marked improvement in histopathological examination. The present finding showed that administration of the hydroalcoholic extract of AJ could prevent of hepatotoxicity induced by BB via improvement serum and tissue parameters and histopathological alterations in liver tissue.

- چکیده انگلیسی: Objective(s): The purpose of this study was to investigate cytotoxicity and membrane toxicity effects induced by Mesobuthus eupeus venom (MEV) on the HT-29 cell line.
Materials and Methods: To determine the in vitro cytotoxicity via MTT assays, HT-29 (as cancer cell line) and Hek-293T (as normal cell) were treated through different concentrations of MEV, and cytotoxicity effects were then measured through assessment of mitochondrial membrane potential (ΔΨm), reactive oxygen species (ROS) generation, and apoptosis induction. The colony formation assay was performed to measure the antiproliferative effect of MEV on HT-29 cells. Nuclei alterations were also observed during apoptosis following DAPI staining. Besides, atomic force microscopy (AFM) was used to detect alterations in morphology and ultrastructure of the cells at a nanoscale level.
Results: According to MTT and clonogenic assays, MEV caused a significant decrease in cell viability and proliferation of HT-29 cells while it did not have any impact on normal cells and the IC50 value was found to be 10 µg/ml. Induction of apoptosis was also confirmed by flowcytometric analysis in HT-29 cells. Moreover, the results indicated that MEV had led to a suppression of proliferation and induction of apoptosis through increased ROS and depolarization of mitochondria. Furthermore, AFM imaging demonstrated apoptosis cell death after being treated with MEV in HT-29 cells.
Conclusion: This study showed that MEV had an antiproliferative effect on HT-29 cells by inducing apoptosis through the mitochondria signaling pathway. These findings suggested that MEV could be used as a promising natural remedy for cancer treatment.- انتشار مقاله: 13-03-1398

- چکیده انگلیسی: Objective(s): Bisphenol A (BPA) as a synthetic compound is applied in many plastic industries. BPA has been reported to have endocrine-disrupting feature with cytotoxic effects. The study aimed to evaluate the efficiency of Naringin against testicular toxicity induced by BPA in adult rats.
Materials and Methods: The animals were assigned into six groups of control, BPA-treated (50 mg/kg), BPA+Naringin-administrated (40, 80, 160 mg/kg) and Naringin-treated (160 mg/kg) for 30 days. At the end of experiments, testicular weight, total testicular protein, epididymal sperm count, testicular enzymes, serum follicle-stimulating hormone (FSH), luteinizing hormone (LH), testosterone and estradiol, testicular enzymatic and non-enzymatic antioxidants and histopathology of testis tissue were evaluated by their own methods.
Results: The results showed a reduction in testicular weight, total testicular protein, epididymal sperm count, testicular enzymes (alkaline phosphatase and lactate dehydrogenase) and decrease in the serum TSH, LH, testosterone and estradiol in BPA-administrated rats. Furthermore, BPA reduced the enzyme activities of glutathione peroxidase, superoxide dismutase, and catalase in testis tissue. Also, BPA caused an induction in lipid peroxidation and increase in reactive oxygen species levels, whereas it decreased the glutathione content of testis tissue. Histological findings exhibited seminiferous tubules vacuoles, atrophy and separation of the germinal epithelium in BPA-administrated rats. Oral administration of Naringin along with BPA normalized the biochemical, morphological and histological changes and reduced the testicular toxic condition.
Conclusion: These results demonstrated that Naringin significantly managed male reproductive toxicity by antioxidant capabilities, preventing morphological modifications and escalating defense mechanism, thereby reducing oxidative stress from BPA-induced damage.- انتشار مقاله: 21-11-1396

- چکیده انگلیسی: Background: Organophosphates are pesticides that are used widely in agriculture and industry. Because of the ease of access and abundant use of these pesticides, many cases of intentional and accidental poisoning of these compounds are reported. The aim of this comprehensive study was to evaluate the symptoms and treatment of patients referred to Razi hospital as the main center for poisoning in the South west of Iran.
Methods: In this study, patients with Organophosphate poisoning referred to Razi hospital during the years 2006 to 2012 was included. The information collected from their records includes reference data, demographic data, clinical manifestations and treatment options.
Results: Of all 173 cases, 46.2% were male. The average age of cases was 24.31 years and most of them were from rural areas. 86% of patients had deliberately consumed a toxic substance. The first clinical symptoms of more than half of the patients were nausea and vomiting. 72.8% of patients needed antidote; a combination of atropine and pralidoxim was administered in 70% of cases. 16.2% required intubation and 80% admitted to ICU. In 52% of cases a specified pesticide was found and 4% of the cases deceased.
Conclusion: Organophosphate poisoning in most cases occurred deliberately as a suicidal attempt. It was more common in rural areas. Need for admission to intensive care unit was more common in cases under the age of twenty. Treatment of patients with both antidote atropine and pralidoxime was a very suitable therapeutic model.- انتشار مقاله: 14-07-1395
