- چکیده انگلیسی: Evidence has shown that liver disease caused by hepatitis viruses can be more aggressive and severe in HIV infected subjects. Therefore, the present cross-sectional study aimed to evaluate the seroprevalence of HDV infection among HIV/HBV co-infected clients in Shiraz, southwest Iran. In this study, 178 patients co-infected with HBV and HIV individuals were enrolled. The diagnosis of HIV infection was documented based on serological assays. The demographic and complementary data were collected by a questionnaire. HBsAg and HDV Ab were detected by commercial quantitative enzyme linked immunosorbent assay kits according to the manufacturer’s instructions. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were also measured. The mean age of the participants was 37.4±7.4 years (range 22-63). 175 (98.4 %) patients were male and 3 (1.6 %) were female. Among 178 patients co-infected with HIV/HBV, 35 cases (19.7%, 95% CI: 14%-25%) were anti-HDV positive and 143 (80.3%) were negative for anti-HDV. HDV exposure in HIV/HBV co-infected patients was associated with blood transfusion (P=0.002, OR: 14.3) and prison history (P=0.01, OR: 2.31) but not with age, marital status, unsafe sex contact, and injection drug abuse. Our data showed a relatively high prevalence of HDV infection in HIV infected population in Shiraz, Iran. The high frequency of HDV Ab in patients with blood transfusion and prison history reveals that HDV transmission occurs more frequently in the parental route than sexual contacts; therefore, blood screening for HDV diagnosis in the high-risk group is recommended.

- چکیده انگلیسی: Background: Herpes simplex viruses (HSV) are human pathogens that establish lytic and latent infections. Reactivation from latency occurs intermittently, which represents a life-long source for recurrent infection. The role of immune factors in the control of recurrent symptomatic HSV lesions is complex and the exact role of cytokines remains unclear.
Objective: To assess the levels of tumor necrosis factor-α (TNF-α) and interleukin-10 (IL-10) along with anti-herpetic IgG and IgM, in the symptomatic reactivation of HSV infection.
Methods: Thirty-six patients with recurrent symptomatic herpes infection were selected as the study group and thirty-two healthy individuals with no history of symptomatic labial herpes infection enrolled as the control group. Skin swabs were obtained from lip and skin lesions for viral culture. Confirmation of HSV cytopathic effect was carried out using PCR assay. The levels of TNF-α, IL-10, IgG and IgM were measured using ELISA.
Results: The level of TNF-α was significantly lower in individuals with recurrent symptomatic herpes infection in comparison with the controls (p=0.04). Also a significant elevation was observed in the levels of specific IgG in patients compared to controls (p<0.05).
Conclusion: The decreased level of TNF-α and increased levels of IgG in individuals with a history of symptomatic reactivation of HSV infection is suggestive of a probable shift in favor of the Th2 immune response.- انتشار مقاله: 14-05-1395

- چکیده انگلیسی: Background: IL-10 is an anti-inflammatory cytokine which is involved in tumorigenesis. Over production of IL-10 and elevated number of IL-10 generating mononuclear cells in breast tumor tissue has already been shown.
Objective: To determine the association of IL-10 promoter polymorphisms with increased risk of breast cancer and its association with breast cancer prognostic factors.
Methods: Peripheral blood samples from 275 female breast cancer patients and 320 cancer free controls were used to detect three single nucleotide polymorphisms in IL-10 promoter region ( -1082, -819, -592 ) by PCR method.
Results: The frequency of genotypes and alleles of three mentioned regions of IL-10 promoter and their haplotypes (GCC, ATA, and ACC) showed no statistically significant difference between patients and controls. In the case of prognostic factors, progesterone receptor (PR) status exhibited significant relation with -1082 genotypes (P=0.03) and haplotypes (P=0.02). -1082 AA genotype was associated with negative PR expression whereas AG and GG genotypes of this site were positively associated with PR expression. Similarly GCC haplotype correlated with positive PR expression and ATA and ACC with negative PR expression.
Conclusion: The data of this study showed that IL-10 promoter gene polymorphisms may not be considered as one of the risk factors for breast cancer in Iranian patients.- انتشار مقاله: 16-05-1395

Virus (HCV) infection, natural drug resistance remains a challenging obstacle for their impacts. The data regarding
protease inhibitors (PIs) resistance in Iran population are limited. The aim of this study was to investigate the variations
in NS3 protease of HCV from non-responder patients. Methods: In this cross-sectional study, 14 HCV infected patients
with genotype 1(N=5) and 3(N=9) who have not responded to Interferon-related regime were enrolled from Liver
Clinic, Shiraz. The NS3 protease region was amplified by Nested-PCR followed by product gel extraction. Besides,
some amplified protease regions were cloned into a cloning vector to improve the sensitivity of mutation detection.
Both crude and cloned sequences were then introduced into sequencing. The obtained sequences were compared with
the NS3 reference sequences and analyzed by Geno2pheno available software to find possible substitutions. In the
end, the phylogenetic tree was constructed. Results: Among variations responsible for PIs resistance, only one out of
14 (7%) sample who was infected with genotype 1a, harbored R117C+N174S double mutation, which causes reduced
susceptibility to Telaprevir. Any another resistance mutation was not found among the studied population. The most
frequent substitutions were determined as I52M(N=9), S102A(N=9), S166A(8) and V170I(8) for genotype 3a, and
F147S/A(4) for genotype 1. However, some uncharacterized substitutions on scored position, including I132L(N=1),
I170V(N=3) and N174S(N=2) were also determined among sequences. Phylogenetic analysis demonstrated that the
protease region has enough power to correctly classify enrolled samples into relevant clusters on the tree. There were 2,
3 and 9 cases of sub-genotypes 1a, 1b, and 3a, respectively. Conclusion: A low frequency of PIs resistance mutations
in our HCV infected population is a hopeful point of starting these drugs in HCV infected patients.- انتشار مقاله: 03-10-1397
