- چکیده انگلیسی: Detection of genetically modified organisms (GMOs) in food is an important issue for all the subjects involved in food control and customer’s right. Due to the increasing number of GMOs imported to Iran during the past few years, it has become necessary to screen the products in order to determine the identity of the consumed daily foodstuffs. In this study, following extraction of genomic DNA from processed foods sold commercially in Iran, qualitative PCR was performed to detect genetically modified maize. The recombinant DNA target sequences were detected with primers highly specific for each investigated transgene such as CaMV35s gene, Bt-11, MON810 and Bt-176 separately. Based on the gel electrophoresis results, Bt-11 and MON810 events were detected in some maize samples; while in none of them Bt-176 modified gene was detected. The results demonstrate for the first time the presence of genetically modified maize in Iranian food products, reinforcing the need for the development of labeling system and valid quantitative methods in routine analyses.

- چکیده انگلیسی: Background: The role of mesenchymal stem cell in cellular therapy is the subject of interest for many researchers. The differentiation potential of MSCs and abilities in modulations of the recipient’s immune system makes them important cells in tissue regenerative studies. MSCs by releasing the proinflammatory cytokines play important role in immunomodulatory systems; however the signaling pathways for releasing of these mediators are not well understood. Glutathione has been shown to play a role in modulation of cytokines in hepatogenic differentiation.
Objective: In the current study we aimed to investigate the effects of buthionine sulfoximine (BSO, inhibitor for glutathione synthesis) and N-acetylecystin (NAC, an inhibitor for ROS generation) on proinflammatory cytokines production in a hepatogenic differentiation model.
Results: BSO and NAC significantly decreased IL-6 and TNF-α levels at 14 days of differentiation, whereas, NAC decreased the levels of IL-8 at days 2 and 14 of differentiation. Moreover, intracellular glutathione level during the differentiation was depleted.
Conclusion: Our current study suggests a novel role of GSH as an immunopharmacological regulatory molecule during hepatogenic differentiation. Finally, this information may shed some light on the understanding of MSCs responses in transplantation and cell therapy in diseases such as chronic hepatic diseases.- انتشار مقاله: 15-05-1395

- چکیده انگلیسی: The study of the interaction between DNA and small molecules such as drugs is one of the current general interest and importance. In this paper, the electrochemical investigation of the interaction between some flavonoids such as rutin, quercetin, and hesperidin with dsDNA on the surface of Supramolecular Ionic Liquid grafted on the Graphene Oxide Modified Glassy Carbon Electrode (SIL-GO/GCE) is reported for the first time. The apparent binding constant (K) of the interaction between flavonoids and dsDNA was calculated using the current titrations. The apparent binding constants (K) of rutin, quercetin and hesperidin were calculated to be 4.3×105, 2.1×105 and 9.2×1-6 M-1, respectively. Furthermore, the electrochemical behavior of rutin on the surface of SIL-GO/GCE was studied in details using cyclic voltammetry and linear sweep voltammetry. The mechanism of the electrochemical redox reaction of rutin was proposed. When DNA was added into flavonoid solutions, their cathodic peak currents were decreased with few changes in the peak potentials. Furthermore, the interaction between rutin and bovine serum albuminwas studied using differential pulse voltammetry. In conclusion, the SIL-GO/GCE provides a promising platform for the study of the interaction between DNA and small molecules.

- چکیده انگلیسی: Recently, with increasing public awareness of food safety, developments of some valid and rapidly analytical methods are required for the determination of residual pesticides in crops.This study was conducted on validation of a reliable, accurate and precise method for multi analysis of pesticides residue in melon as a main fruit in Iranian food basket. A reliable, and valid method based on spiked calibration curves and modified QuEChERS sample preparation was developed for determination of 12 pesticide residues in melon by Gas Chromatography Mass Spectrometry (GC/MS). The interested pesticides are, diazinon, dimethoate, chlorothalonil, pirimicarb, chlorpyrifos-methyl, metalaxyl, α-endosulfan, ß -endosulfan, permethrin 1, 2 and fenvalerate 1, 2 that are belonged to different chemical groups of pesticides. Triphenylmethane (TPM) is used as internal standard. Use of spikedsamples for constructing the calibration curve substantially reduced adverse matrix-related effects. The recovery of pesticides at 3 concentration levels (n=3) was in range of 79.8-97.6%. The method was proved to be repeatable with RSD lower than 20%. The limits of quantification for all pesticides were The recoveries and repeatabilities were in accordance with the criteria set by SANCO Guideline (Commission of the European Communities, 2006). Using spiked samples for preparation of calibration curves is a sensitive, selective, rugged, rapid and accurate method in pesticides analysis.
