- چکیده انگلیسی: In this research, we investigated the antidiabetic activity of Vitis elegans rhizome, which is used as traditional treatment for diabetes mellitus. The methanol, chloroform, petroleum ether, and hexane extracts of the plant root were obtained through serial exhaustive extraction and were analyzed by Thin Layer Chromatography (TLC). Glycogen phosphorylase (GP) assay was done to determine the inhibitory effects of respective extracts on GP enzyme. Total phenol content was measured using the Folin-Ciocalteu method and brine shrimp test was done to evaluate the toxicity of the extracts. Evaluation of TLC plates alone and after spraying with different reagents indicated that terpenoid was the major component of the sample followed by alkaloid and phenol. Chloroform extract applied more inhibitory effects on GP enzyme activity with percentages of 39.65 in concentration of 2.5 mg/ml. This suppression effect was higher than glucose, as a standard inhibitory agent in the body. The highest amount of phenol was found in the methanol extract, equal to 49 mg GAE g-1. Petroleum ether, chloroform and methanol extracts were considered as non-toxic solvents with LC50 values of 8.9, 3.5 and 3.7 mg/ml respectively. While hexane with 0.089 mg/ml LC50 value was classified as toxic extract. Based on the results of this study, we concluded that Vitis elegans rhizome, has the potential to be further studied for anti-hyperglycemic properties.

- چکیده: مقدمه و هدف: سرطان پستان دومین سرطان شایع در زنان بعد از سرطان ریه میباشد. با توجه به اینکه ترکیبات گیاهی قرنهاست که برای درمان سرطان استفاده میشوند. هدف از این مطالعه بررسی اثر سمیت سلولی عصاره هیدروالکلی آویشن دنایی بر روی رده سلولهای سرطانی MCF-7 بود. روش تحقیق: سلولهای سرطانی MCF-7 و سلولهای طبیعی فیبروبلاست در محیط کشت DMEM حاوی سرم جنین گاو و آنتیبیوتیک کشت گردیدند. این سلولها در مجاورت دوزهای مختلف عصاره هیدروالکلی آویشن دنایی (156/0، 312/0، 625/0، 25/1، 5/2 میلیگرم بر میلیلیتر) قرار گرفتند و به مدت 24، 48 و 72 ساعت انکوبه شدند. پس از پایان انکوباسیون، از روش تغییر یافته آزمون رنگ سنجی MTT جهت تعیین سمیت سلولی عصاره استفاده شد. نتایج و بحث: نتایج حاصل از تست MTT نشان داد که عصاره هیدروالکلی آویشن دنایی اثر ضد سرطانی وابسته به دوز و زمان بر سلولهای سرطانی MCF-7 دارد، به طوری که با افزایش غلظت عصاره و انکوباسیون 72 ساعت بیشترین درصد مرگ سلولی مشاهده شد ( 05/0 > P). عصاره گیاه سمیت سلولی قابل توجهی را بر روی سلولهای طبیعی فیبروبلاست نشان نداد.لذا به نظر میرسد با تحقیقات بیشتر در آینده، میتوان از ترکیبات آن در درمان سرطان بهره جست. توصیه ی کاربردی/ صنعتی:با توجه به اینکه مصرف داروهای گیاهی برای درمان بسیاری از بیماریها در حال افزایش است، عصاره هیدروالکلی آویشن دنایی به دلیل داشتن خاصیت آنتیاکسیدانی میتواند در درمان سرطان در کنار دارو درمانی استفاده شود.
- چکیده انگلیسی: Background & Aim:Breast cancer is the second most common cancer in women after lung cancer. Given that the herbal ingredients are used for centuries to treat cancer,The aim of this study was to determine the cytotoxic effect of hydroalcholic extract from Thymus daenesisCelak on MCF-7 cancer cells line. Experimental: Breast cancer cells MCF-7 and natural fibroblast cells were cultured in DMEM medium containing fetal bovine serum and antibiotics. The cells were exposed to different doses ofhydroalcholic extract ofThymus daenesisCelak(0.156, 0.312, 0.625, 1.25, 2.5mg/ml)and incubated for 24, 48 and 72 hours, respectively. After incubation, the modified MTT colorimetric test was used to determine cytotoxicity. Results: The results of MTT test showed thathydroalcholic extractof Thymus daenesisCelak has dose- and time-dependent anti-cancer effect on MCF-7 cancer cells, so that by increasing the concentration and 72h incubation, the most cell death was observed (P<0.05). Plant extract did not show significant cytotoxicity on natural fibroblast cells. Then, it seems that its compounds can be used in treating cancer through more future research. Recommended applications/industrie: With regard to the increasing use of herbal medicines to treat many diseases, hydroalcholic extract of Thymus daenesis Celak can be used to treat cancer with drug therapy due to having antioxidant properties.

- چکیده انگلیسی: AbstractBackground: Dietary pattern is an effective way of studying the effect of diet on diseases. We investigated the association between dietary patterns and blood pressure (BP) in adults aged 20-50 years.Methods: In a cross-sectional design, 418 individuals were selected through stratified multistage random sampling from households living in different regions of Shiraz. Information on demographic characteristics, anthropometric features, dietary intakes, and systolic (SBP) and diastolic (DBP) blood pressure was gathered. Dietary patterns were determined using a validated food frequency questionnaire.Results: Three dietary patterns were specified: vegetable (high in vegetables and legumes), Western-like (high in meat, sugarsweetened beverages, salty and sweet snacks, refined grains, high-fat dairy), and Mediterranean-like (rich in low-fat dairy, fruit, vegetables, nuts, olive, fish, and low in hydrogenated fats). After adjustment for confounders, Mediterranean-like dietary pattern had an inverse association with SBP (β=-0.24; 95% CI: -5.25, -1.27) and DBP (β=-0.17; 95% CI: -3.65, -0.20) in males but not females. Vegetable and Western-like dietary patterns were not associated with BP in either sex after adjusting for confounders. Positive relationships were observed between BP and body mass index (r=0.28 and 0.33 for SBP and DBP, P<0.001), waist circumference (r=0.51 and 0.45 for SBP and DBP, P<0.001), and waist-to-hip ratio (r=0.54 and 0.44 for SBP and DBP, P<0.001). Dietary energy and carbohydrates were positively and fats inversely associated with BP. Among micronutrients, vitamin E had a significant inverse association with BP.Conclusion: Mediterranean-like dietary pattern may lower the risk of hypertension in Shiraz males.

- چکیده انگلیسی: Introduction: Today, development of professionalism is acritical aim of medical schools. Studies have demonstrated thatmedical students’ perceived level of professionalism is inadequateworldwide. This study aimed to investigate the medical students’perceptions of their colleagues’ professional behavior.Methods: This study is a cross-sectional study with 280 medicalstudents at Shiraz University of Medical Sciences in their fifthto seventh year of study as the sample. The study was performedduring one month in 2013, using stratified random samplingmethod. The instrument of the study was the Persian version ofthe questionnaire of the American Board of Internal Medicine(ABIM).The questionnaire includes demographic information,questions about the meaning of the professionalism, history ofmedical ethics education programs and 12 behavioral questions.The data were analyzed using student t-test and Pearson correlationtest. The significance level was set as 0.05.Results: Forty percent of respondents did not know the meaning ofprofessionalism. The mean±SD score of behavioral questions was5.91±1.2 on a scale from 0 to 10. The mean±SD score of excellencequestions was 4.94±1.7. It was 7.05±1.9 for ‘honor/integrity’, and6.07±2.1 for ‘altruism/respect’ questions. There was a significantassociation between gender and excellence score (p=0.007).Conclusion: Medical students assessed their colleagues’professional behavior as poor. They did not have properinformation about professionalism. Medical students are futuregeneral practitioners and respecting medical ethics by them is veryimportant in a perfect health system. Universities should emphasizethe importance of teaching professionalism to medical studentsand faculty members, using innovative education methods.Keywords: Professionalism; Excellence; Respect; Medical students

- چکیده انگلیسی: AbstractBackground: In recent years, laparoscopic sleeve gastrectomy (LSG) has become more acceptable for obese patients. Single-port sleeve gastrectomy (SPSG) is more popular since each abdominal incision carries the risk of bleeding, hernia, and internal organ injury as well as exponentially affecting cosmesis. This cross-sectional study aimed at comparing multi-port sleeve gastrectomy (MPSG) and SPSG in terms of their early results and complications. Methods: Out of129 obese patients candidated for LSG, 102 patients were assigned to 2 groups of SPSG and MPSG. Complications and demographic data such as body mass index (BMI), age, gender, operation time, and hospital stay were measured. All surgeries were carried out between2013 and 2015 in Shiraz, Iran. Data analysis was performed using SPSS, version 16 for Windows (SPSS Inc., Chicago, IL). The continuous and categorical variables were compared using the Student t-test and the Chi-square test or the Fisher exact test, respectively. Results: The patients’ data from both groups were similar in terms of age, intraoperative and postoperative bleeding volume, and length of hospital stay. Mean BMI was 42.8±0.7 in the SPSG group and 45.3±1.2 in the MPSG group. Duration of surgery was significantly lower in the SPSG group (P<0.001). Only 1 patient from the SPSG group and 5 patients from the MPSG group had bleeding as an early complication.Conclusion: The differences in each complication between the groups were not statistically significant. SPSG seems to be safe and is the same as MPSG in terms of major postoperative complications.Trial Registration Number: IRCT201512229936N12

- چکیده انگلیسی: Objective(s): 4-aminosalicylic acid (4-ASA) is an isomer of mesalazine that has recently been shown to be effective against inflammatory bowel disease (IBD), and more specifically, ulcerative colitis. However, the majority of orally administered 4-ASA is readily and extensively absorbed from the stomach and small intestine, so only a small amount is transported to the colon. A mutual ester and azo prodrug of 4-ASA was synthesized with polyethylene glycol (PEG) and dimethylaniline, respectively , to overcome this issue.
Materials and Methods: The 4-ASA prodrug was synthesized via a two-step process and then characterized by 1H-NMR. The stability of the prodrug was evaluated in simulated gastric fluid (pH 1.2). Furthermore, the in vitro release profiles of the drug conjugate was evaluated at pH 1.2, as well as pH 6.8 in the absence or presence of rat cecal content.
Results: The prepared prodrug was stable at pH 1.2, indicating that it could be protected from the acidic environment of the stomach. Also, the results of drug release at pH 6.8 showed that the amount of 4-ASA released was 63% within 12 hr in the absence of rat cecal content, while in the presence of rat cecal content, 97% of 4-ASA was released from the prodrug in 6 hr.

- چکیده انگلیسی: Objective(s): Gastric cancer is the third leading cause of cancer-related death worldwide. The overall survival rate of patients is poor because gastric cancers are usually diagnosed at the late stages. Therefore, further research is needed and appropriate research tools are required to develop novel therapeutic approaches.
Materials and Methods: Eight female athymic nude mice with a C57BL/6 background were used in this study. AGS cells were inoculated into the flank. The tumor volumes were calculated and growth curves were drawn. When the volume of the tumors reached 1000 mm3, the animals were humanely euthanized with CO2 gas. After harvesting, tumors were analyzed with Hematoxylin and Eosin (H&E) and immunohistochemistry (IHC). A pathologist confirmed tumor entity through H&E staining. Tumors were evaluated for expression of HER-2, P53, Ki-67, CD34, cytokeratin 8 (CK8), vimentin, estrogen receptor (ER), and progesterone receptor (PR) utilizing immunohistochemistry.
Results: The tumor take rate was 62.5%, mean doubling time was 40.984 d, and the latency period was 30.62 days. The H&E staining results showed highly malignant hyperchromatin epithelial cells. IHC assessment showed the mutation status of P53 gene. The expression score of the CK8 protein in the tumor cells was +3. Vimentin protein was not expressed and changes in mesenchymal phenotype were not observed. Ki-67 IHC indicated that the proliferation rate was >43% and angiogenesis was defined as high MVD.
Conclusion: The respective AGS xenograft model provides an opportunity to understand the pattern of tumor growth as well as to evaluate new gastric cancer therapies in in vivo studies.- انتشار مقاله: 28-01-1396

- چکیده انگلیسی: Objective(s): This study evaluates the effect of substitution of microcrystalline cellulose (MCC) with ethylcellulose (EC) on mechanical and release characteristics of theophylline pellets.
Materials and Methods: The effect of addition of EC was investigated on characteristics of pellets with varying drug content prepared by extrusion-spheronization. Also the effect of type of granulating liquid (water or Surelease) was investigated on characteristics of selected pellets. The pellets were characterized for particle size (sieve analysis), mechanical strength, morphology (microscopy), thermal (DSC) and dissolution behaviors.
Results: The exrtudability of the wet mass was reduced upon inclusion of EC so that complete replacement of MCC was not possible. Increase in EC percentage led to lower production yield and formation of pellets with larger diameter and slightly rough surfaces. Inclusion of EC also affected the mechanical properties of pellets but had negligible effect on drug release profile. The surface of selected pellets became smoother and their production yield increased upon the use of Surelease as granulating liquid. In addition the rate of drug release decreased to some extent when Surelease was used.
Conclusion: Preparation of theophylline pellets with EC alone was not possible in process of extrusion-spheronization. Partial replacement of MCC with EC changed physicomechanical properties of pellets but hardly affected drug release. Although the use of Surelease as granulation liquid slightly decreased the rate of drug release, desirable matrix pellets with sustained drug release could not be produced. - چکیده انگلیسی: Objective(s):Solid dispersion formulation is the most promising strategy to improve oral bioavailability of poorly water soluble drugs. The aim of this study was to compare the effect of polyvinylpyrrolidone K30 (PVP) and poloxamer-188 (PLX) as carrier in solid dispersion formulations of celecoxib (CLX).
Materials and Methods: Solid dispersions of CLX:PVP or CLX:PLX were prepared at different ratios (2:1, 1:1, 1:2, 1:4, 1:6) by solvent evaporation and melting methods, respectively. The characterization of samples was performed using differential scanning calorimetery (DSC), X-Ray powder diffraction (XRPD) and Fourier transform infrared spectroscopy (FT-IR). The Gordon-Taylor equation was used to estimate the Tg of solid dispersion systems and the possibility of the interaction between CLX and PVP. Also, the dissolution rate of all samples was determined.
Results: DSC and XRPD analyses confirmed the presence of amorphous state of drug in solid dispersion systems. FT-IR studies showed CLX could participate in hydrogen bonding with PVP whilst no specific interaction between CLX and PLX was observed. Both PVP and PLX enhanced the dissolution rate of drug in solid dispersion samples. The dissolution rate was dependent on the ratio of drug: carrier. Interestingly, the solid dispersion samples of PLX at 2:1 and 1:1 drug: carrier showed slower dissolution rate than pure CLX, whilst these results were not observed for PVP.
Conclusion: The effect of PVP on dissolution rate enhancement was more pronounced compared to the other carrier. Having a higher Tg and more effect on dissolution rate, PVP could be considered as a more suitable carrier compared to PLX in solid dispersion formulation of CLX.- انتشار مقاله: 30-02-1393
