- چکیده انگلیسی: The aim of the present study was stereological evaluation of testes of azoospermic animal model using busulfan in rat. Three groups of male adult rats were used in this study. The first group was injected by single dose of busulfan (10 mg kg-1) and their testes were removed on day 35 post injection. The second group received double doses of busulfan with 21 days interval and their testes were removed on day 35 after the second injection. The testes of the third group were removed without busulfan therapy. In 10 circular transverse sections of tubules stained with hematoxylin-eosin, stereological parameters were measured. The testes were rated for its spermatogenic potential on a modified spermatogenic scale of 0 to 6. Cellular (germinal epithelium) diameter and area of the seminiferous tubules, total diameter and cross sectional area of the tubules of the seminiferous tubules in rats that received double doses of busulfan were less than the rats in single dose of busulfan and control groups (p < 0.05). Spermatogenesis index of seminiferous tubules in rats receiving two doses of busulfan was less than the rats received one dose of busulfan (p < 0.001) and the index of both treatment groups were less than the control group (p < 0.001). In conclusion, two doses of busulfan injection with 21 days interval produced an appropriate experimental model of induced azoospermia with comparable stereological indices of seminiferous tubules in rat.

- چکیده انگلیسی: Spinal cord injury (SCI) is a drastic disability that leads to spinal cord impairment. This study sought to determine the effects of bone marrow stem cells (BMSCs) on caspase-3 levels after acute SCI in mice. Forty-two mice were randomly divided into 3 groups: control (2 subcategories), subjected to no intervention; sham (3 subcategories), subjected to acute SCI; and experimental (2 subcategories), subjected to SCI and cell transplantation. In the experimental group, 2×105 BMSCs were injected intravenously 1 day after SCI. The mesenchymal property of the cells was assessed. The animals in the 3 groups were sacrificed 1, 21, and 35 days after the induction of injury and caspase-3 levels were evaluated using a caspase-3 assay kit. The obtained values were analyzed with ANOVA and Tukey tests using GraphPad and SPSS. Based on the assessments, the transplanted cells were spindle-shaped and were negative for the hematopoietic markers of CD34 and CD45 and positive for the expression of the mesenchymal marker of CD90 and osteogenic induction. The caspase-3 levels showed a significant increase in the sham and experimental groups in comparison to the control group. One day after SCI, the caspase-3 level was significantly higher in the sham group (1.157±0.117) than in the other groups (P<0.000). Twenty-one days after SCI, the caspase-3 level was significantly lower in the experimental group than in the sham group (0.4±0.095 vs. 0.793±0.076; P˂0.000). Thirty-five days following SCI, the caspase-3 level was lower in the experimental group than in the sham group (0.223±0.027 vs. 0.643±0.058; P˂0.000). We conclude that BMSC transplantation was able to downregulate the caspase-3 level after acute SCI, underscoring the role of caspase-3 as a marker for the assessment of treatment efficacy in acute SCI.

- چکیده انگلیسی: Objective(s): Adipose tissue-derived mesenchymal stem cells (AT-MSCs) with more potent immunomodulatory effects, greater proliferative potential and secretion of growth factors and cytokines in comparison with bone marrow derived MSCs are more appropriate for cell therapy. The aims of the present study were to evaluate the histomorphometric effect of AT-MSCs allotransplantation on regeneration of germinal layer cells of seminiferous tubules in busulfan-induced azoospermic hamsters.
Materials and Methods: In the present experimental case-control study, AT-MSCs were isolated from adipose tissue of two female and six male donor albino hamsters, and testes of the males were simultaneously used as negative control group. Six mature male recipient hamsters received two doses of busulfan with three weeks interval to stop endogenous spermatogenesis. Right testis of hamsters was intratubular injected with AT-MSCs via efferent duct 35 days after induction of azoospermia and was used as cell therapy group. The left testis without cell therapy was served as azoospermia group.
Results: After 35 days, testes and epididymis in all groups were removed for histological evaluation. Histomorphometric analyses of AT-MSCs-treated testes and epididymis showed that the epithelial tissue of seminiferous tubules was normally repaired in most cell-treated seminiferous tubules, and spermatozoa were present in epididymis tubes in comparison with intact testes. The untreated seminiferous tubules and epididymis tubes of azoospermia group were empty.
Conclusion: Allotransplanted AT-MSCs could successfully induce spermatogenesis in azoospermic seminiferous tubules of hamster. Therefore, AT-MSCs can be suggested as an attractive candidate in cell transplantation of azoospermia.- انتشار مقاله: 21-10-1396

- چکیده انگلیسی: Objective(s): Nowadays much effort is being invested in order to diagnose the mechanisms involved in neural differentiation. By clarifying this, making desired neural cells in vitro and applying them into diverse neurological disorders suffered from neural cell malfunctions could be a feasible choice. Thus, the present study assessed the capability of fetal brain extract (FBE) to induce rat bone marrow-derived mesenchymal stem cells (BM-MSCs) toward neural cells.
Materials and Methods: For this purpose, BM-MSCs were collected from rats and cultured and their mesenchymal properties were confirmed. After exposure of the BM-MSCs to fetal brain extract, the cells were evaluated and harvested at days 3 and 7 after treatment.
Results: The BM-MSCs that were exposed to FBE changed their appearance dramatically from spindle shape to cells with dendrite-like processes. Those neural like processes were absent in the control group. In addition, a neural specific marker, vimentin, was expressed significantly in the treatment group but not in the negative control group.
Conclusion: This study presented the FBE as a natural neural differentiation agent, which probably has required factors for making neurons. In addition, vimentin overexpression was observed in the treated group which confirms neuron-like cell differentiation of BM-MSCs after induction.- انتشار مقاله: 11-12-1395

- چکیده انگلیسی: Objective(s): Bone marrow-derived mesenchymal stem cells (BM-MSCs) potentials make them appropriate for cell therapy including ability of differentiation and release of anti-inflammatory cytokines and growth factors secreta. For treatment of azoospermia to induce proliferation and differentiation of germ cells, MSCs transplantation has been introduced. The aim of the present experimental case-control study was to histomorphometric evaluation of the germinal cells in seminiferous tubules of azoospermic rats before and after BM-MSCs allotransplantation. Materials and Methods: In the present study, BM-MSCs were isolated from six male rats and confirmed. Their testes also served as intact negative controls. The recipient rats (n=6) were received two doses of 10 mg/kg of busulfan with 21 days interval to induce azoospermia. After cessation of spermatogenesis, the rats were allotransplanted with the BM-MSCs into efferent duct of right testes. Thirty-five days later, the right cell-treated testes were compared to left azoospermic ones. Results: Histomorphometric analyses showed that the seminiferous tubules treated with BM-MSCs had normal morphology in comparison with azoospermic testes, which were without germinal layer. In most BM-MSCs-treated seminiferous tubules, spermatogenesis was observed. Conclusion: The allotransplanted BM-MSCs could induce spermatogenesis in seminiferous tubules of azoospermic rats.

- چکیده انگلیسی: Background: Chemical drugs in treatment of liver diseases are still scare and with many complications. Citrullus colocynthis known as bitter apple is on of herbals that can affect liver function. This study evaluated the effect of C. colocynthis on serum lipid profile and hepatic tissue histology in CCl4-induced liver injury rat model.
Methods: In this experimental animal study, forty-two Wistar rats were randomly divided into 6 equal groups of control received 1 mL/kg of distilled water, twice a week for 2 weeks intraperitoneally (IP); sham injected with 50% CCl4 identically; and experimental groups administered with just 20 mg/kg of C. colocynthis similarly; receipt of 20 mg/kg of C. colocynthis after induction of fatty liver disease by CCl4, identically; received 50 mg/
kg of C. colocynthis in the same manner; and receipt of 50 mg/kg of C. colocynthis after induction of fatty liver disease by CCl4, identically. The triglyceride (TG) and cholesterol levels were determined and liver tissue changes were assessed histologically before and after interventions.
Results: C. colocynthis could decrease TG, total cholesterol (TC), highdensity lipoprotein (HDL), but not low-density lipoprotein (LDL) after liver injury. C. colocynthis could significantly ameliorate the injured liver.
Conclusion: Based on the decrease in TG, TC and HDL levels and histological healing picture in hepatic tissue after administration of C. colocynthis, it can be recommended as an alternative medical therapy for liver injuries because it is inexpensive and is easily available. - چکیده انگلیسی: Background: Mango fruit (Mangifera indica) is rich of phytoestrogens, polyphenols, calcium, iron and potassium which play an important role in bone and dental growth and development. The aim of this study was to determine the effect of M. indica on embryos in pregnant mice.
Methods: In an experimental study, 24 pregnant mice were equally divided into 3 groups of those receiving aqueous extract of the mango fruit, its alcoholic extract and the control group receiving just distilled water. Animals were sacrificed at 19th day of pregnancy and embryos were removed and fixed in 10% formalin for histological studies.
Results: No abnormality and teratogenic effects were noticed in bone, cartilage, liver, spleen, kidney, digestive tract and spinal cord tissues.
Conclusion: Aqueous and hydroalcoholic extracts of the mango fruit (M. Indica) can be administered safely during pregnancy without any complications in body tissues an in bone and dental growth and

- چکیده انگلیسی: Background: Burn is a life-threatening problem which can lead to alteration in the expression of several genes. A folk medicine topical ointment made from Perovskia abrotanoides (P. abrotanoides) was extensively used in northern region of Iran for burn wound healing properties. Therefore, the current study aimed to examine the healing potency of P. abrotanoides ointment and the expression patterns of wound healing-related genes in rat experimental model of of burn.
Methods: In an experimental study, scald burn was inflicted in 28 adult male Sprague-Dawley rats by pouring 2.5 cc of boiling water into a firm rubber ring (2 cm diameter) on the dorsal surface of skin for 10 seconds. Rats were randomly divided into four groups and treated for 7 consecutive days: P. abrotanoides ointmenttreated group; a vehicle-treated group, which received the complete ointment without P. abrotanoides part; silver sulfadiazine-treated group; and the control group which received no treatment. Animals were sacrificed and a full thickness skin biopsy was taken from the burn site. Finally, the relative quantification of gene expression was performed using the vascular endothelial growth factor (VEGF) and transforming growth factor-β (TGF-β) as target genes and β2-microglobulin (β2M) as a reference gene.
Results: VEGF was up-regulated during the first week of healing in P. abrotanoides ointment-treated group; however, the local expression of TGF-β was similar between the groups.
Conclusion: P. abrotanoides ointment utilized in Iranian folk medicine can accelerate wound healing by producing VEGF.- انتشار مقاله: 27-09-1397

- چکیده انگلیسی: Background: Herbal medicine may have irreparable effects on fetal development. This study investigated the effect of Cariandrumsativum (coriander) aquous extract on mice fetal height and weight.
Methods:In an experimental study, 120 pregnant mice were divided into four equal groups. The control group was without any intervention. The experimental group received 3500, 5000 and 6500 mg/kg of aquous extract of coriander. The height and weight of the embryos were recorded in first, second and third week of pregnancy.
Results: The height and weight of embryos in experimental groups in first, second and third week was significantly lower than the control group.
Conclusion: Coriander (C. Sativum) aquous extract was shown to reduce the fetal height and weight in a dose-dependent manner. These findings can be added to the literature when coriander is going to be used during pregnancy.- انتشار مقاله: 14-12-1396
