- چکیده:
- چکیده انگلیسی: Background: It has been suggested that routine assessment and quantification of different lymphocyte subsets can provide clinically meaningful prognostic information in breast cancer (BC). Objective: To determine the relationship between peripheral blood lymphocyte subsets and pathological parameters and response to therapy in patients with BC. Methods: Thirty patients with operable breast cancer treated surgically with either modified radical mastectomy or breast conservative surgery, and 20 healthy controls were included. For detection of lymphocyte subsets in peripheral blood; Fluorochrome-labeled monoclonal antibodies were used and cells were analyzed by flow cytometry. Patients were treated with chemotherapy, radiotherapy and hormonal treatment, and followed up to determine relapse and recurrence-free survival (RFS).
Results: Significant differences were found in the frequencies of B, T, NK, NKT, and CD28‒T cells between patients with BC and controls. Moreover, a significant difference was found in the percentage of CD8+CD28‒ T cells between patients with different pathologic subtypes of BC and negative correlations were observed between the frequency of CD8+CD28‒T cells and memory B cells, and RFS. Also, a significant difference in the frequency of naïve B cells was found in patients with different tumor grades and a negative correlation was found between the frequencies of B cells and NKT cells. Conclusion: NK, NKT, lymphocytes, and CD28‒ T cells significantly differed between healthy controls and BC patients. Also, memory B cells were associated with good response to treatment while CD28‒ T cells were associated with shorter RFS.- انتشار مقاله: 03-07-1398

- چکیده:
- چکیده انگلیسی: Background: Lung cancer is one of the main human health threats. Survival of lung cancer patients depends on the
timely detection and diagnosis. Among the genetic irregularities that control cancer development and progression, there
are microRNAs (miRNAs). This study aimed to assess the plasma level of circulating miRNA-17 and miRNA-222 as
non-invasive markers in non-small-cell lung cancer (NSCLC) patients. Patients and methods: A total of 40 patients
with NSCLC and 20 healthy controls who were matched in terms of age and sex with the patient group were included
in this case-control study.. Estimation of miRNA-17 and miRNA-222 expression profiles in the plasma was done
using quantitative real-time PCR (qRT-PCR). The relationship between both markers and their clinicopathological
features were also determined. Receiver operating characteristic (ROC) curve analysis was done to evaluate the role of
these microRNAs in NSCLC diagnosis and follow-up. Results: MiRNA-17 and miRNA-222 levels were significantly
upregulated in NSCLC patients compared with controls (48.32±12.35 vs 1.16±0.19 and 34.53±3.1 vs 1.22±0.14)
(P=0.000). Plasma miRNA-17 level was increased, and the miRNA-222 level was decreased across different stages of
the disease; however, these differences d were not statistically significant (P=0.4, P=0.5, respectively). The miRNA-17
levels were higher in the lung cancer patients with metastasis , but miRNA-222 levels were lower patients without
metastasis. We found no statistically significant difference in this regard(P=0.4 vs P=0.3, respectively). ROC curve
analysis showed that the sensitivity and specificity of miRNA-17 were 77.78% and 87.50% , and of miRNA-222 were
50% and 88.89%. Conclusion: MiRNA-17 and miRNA-222 can be considered as non-invasive biomarkers for detection
of early lung carcinogenesis and metastasis in patients with NSCLC, hence providing a basis for the development of
novel therapeutic approaches.- انتشار مقاله: 14-07-1397

- چکیده:
- چکیده انگلیسی: Background: Resistance to chemotherapy is a major obstacle to curing acute myeloid leukaemia (AML), and
several antigens are claimed to play primary roles in this resistance. Purpose: The aim of this study was to evaluate
the roles of CD56, CD11b and Smac/DIABLO gene expression levels as prognostic markers of the clinical outcome,
response to chemotherapy and survival of AML patients. Materials and Methods: A cross-sectional observational
study was conducted on 60 naïve-AML patients who received induction therapy with mitoxantrone and cytarabine
combined with a high dose of cytarabine. The CD56,CD11b and Smac/DIABLO expression levels were assessed using
flow cytometry at diagnosis and were analysed for correlation with the possible associated risk factors, response to
chemotherapy, and median duration of disease-free survival (DFS) and overall survival (OS). Results: The overall results
revealed that AML patients who exhibited positive expression for CD56 and CD11b had short median durations of DFS
and OS.(P = 0.019, 0.006, 0.029 and 0.024, respectively). Additionally, low Smac/DIABLO expression had a negative
impact on treatment outcome in terms of CR rate (p=0.012) and reduced DFS (p=0.000) and OS(p=0.000) values.
Conclusions: CD56 and CD11b positivity and low Smac/DIABLO expression are important predictive factors for
the occurrence of chemoresistance, in addition to other risk factors, among AML patients.- انتشار مقاله: 06-02-1397
