- چکیده انگلیسی: Objective(s): Multiple sclerosis (MS) causes extensive damage in the hippocampus. Vitamin B12 (vit B12) and estradiol benzoate (EB) have anti-inflammatory and re-myelination properties that make them proper in improvement of cognitive impairment. This study aimed to evaluate the effects of these compounds on learning and memory disturbances.
Materials and Methods:77 adult male rats were implanted with stainless steel guide cannula bilaterally into the hippocampal area. The animals received 3 μl intrahippocampal EtB 0.01% and were randomly divided into eleven groups (7 rats/group). The groups included control, peanut oil (sham1), distilled water (sham 2), vit B12 (0.25, 0.5, 1 mg/kg), EB (25 and 50 mg/kg), vit B12 (0.25 mg/kg) plus EB (25 mg/kg), vit B12 (0.5 mg/kg) plus EB (25 mg/kg), and vit B12 (1 mg/kg) plus EB (50 mg/kg). The control group received intrahippocampal saline (as solvent). The locomotor activity and learning and memory functions were evaluated by open-field and shuttle-box tests, respectively. AKT, CREB, and BDNF levels were analyzed by Western blotting.
Results: This study has found significant deficit in passive avoidance learning, locomotor activity, as well as decrease in the levels of phosphorylated AKT, BDNF, and CREB in groups that received EtB. Vit B12 (1 mg/kg), EB (50 mg/kg), and their combination markedly improved these side effects.
Conclusion: This study demonstrated that vit B12 and estradiol benzoate, especially in combination therapy, can be helpful in treatment of memory problems and MS-induced dysfunction through activation of the hippocampal AKT, BDNF, and CREB proteins.- انتشار مقاله: 03-06-1399

- چکیده انگلیسی: Objective: Previous studies indicated anti-inflammatory effects for Portulaca oleracea in various inflammatory disorders. In this study, the effects of P. oleracea on bronchoalveolar lavage fluid (BALF) levels of total protein (TP), Phospholipase A2(PLA2) and IgE in sensitized rats were examined.
Materials and Methods: Male rats were randomly divided into six groups namely, control (group C), sensitized rats (group S), sensitized animal treated with three concentrations of the extract of P. oleracea and dexamethasone (n = 8 for each group). The levels of TP, PLA2 and IgE in BALF were measured.

- چکیده انگلیسی: Objective: Inflammation is one of the major components of asthma. Our aim was to evaluate the effects of C. sativus extract on total and differential white blood cells (WBC) count in lung lavage fluid (LLF) of ovalbumin-sensitized rats.
Materials and Methods: Forty rats were divided into five groups (n = 8 for each group) as control (C), sensitized with injection and inhalation of ovalbumin (OA) alone (S), and three groups of sensitized and treated with different doses of C. sativus extract (S+50EX, S+100EX, and S+200EX groups). Total and differential WBC counts of LLF were evaluated in control, sensitized, and treated sensitized groups.
Results: Total WBC count, neutrophil, and eosinophil percentage in LLF were increased in sensitized animals compared with the control group (p<0.001). Treatment of sensitized animals with all doses of the extract significantly reduced WBC number and the percentage of neutrophil and eosinophil compared with the sensitized animals (p<0.01–0.001).
Conclusion: According to these results, the extract of C. sativus could be effective on alleviating lung inflammatory cells specially eosinophils in lung lavage of sensitized animals which may indicate a preventive effect of this plant on lung inflammation in asthma.- انتشار مقاله: 25-02-1392

- چکیده انگلیسی: Objective: The effects of natural adjuvants were examined on total and differential WBC counts in lung lavage of sensitized guinea pigs.
Materials and Methods: In three sensitized groups of guinea pigs including: untreated sensitized animals (S), sensitized animals treated with adjuvant G2 (S+G2) and G2F (S+G2F) as well as non-sensitized group (C) (n=6 for each group), total and differential WBC counts of lung lavage were examined. Sensitization of animals was achieved by injection and inhalation of ovalbumin (OA).
Results: The results showed increased total WBC, eosinophil, neutrophil, and basophil counts, and decreased lymphocytes in lung lavage of sensitized animals compared with the control group (pConclusion: These results indicate important preventive effects of two natural adjuvants, especially G2, on lung inflammation of sensitized guinea pigs.

- چکیده انگلیسی: Objective: Asthma is a chronic inflammatory disease of the respiratory airways distinguished by edema and infiltration of inflammatory immune cells. To test our hypothesis about the anti-inflammatory effect of saffron, we examined effects of Crocus sativus (C. sativus) extract as a prophylactic anti-inflammatory agent in sensitized rats.
Materials and Methods: To induce experimental asthma, rats were sensitized with injection and inhalation of ovalbumin (OA). Forty male Wistar rats were divided into 5 groups (n=8 for each): control, sensitized (asthma), and sensitized and pretreated with three different concentrations of extract, 50, 100, and 200 mg/kg, 2 times a week (group asthma+50EX, group asthma+100EX, and group asthma+200EX). After 32 days, total white blood cells (WBC) counts, red blood cells (RBC), and platelet counts in blood were examined.

- چکیده انگلیسی: Objective: Asthma is an airway complex disease defined by reversible airway narrowing and obstruction, chronic airway inflammation, airway hyperresponsiveness, and tissue remodeling. The purpose of this study was to determine the effect of Brassica napus L. (B. napus) on airway pathologic changes in a rat model of asthma.
Materials and Methods: Twenty-four rats were divided into 4 groups: control, asthmatic, asthmatic treated with 0.5 mg/kg B. napus oil, and asthmatic treated with 0.75 mg/kg B. napus oil. To induce the experimental asthma, rats in groups 2, 3, and 4 received an i.p. injection of ovalbumin and aerosolized ovalbumin. Simultaneously, rats in groups 3 and 4 received B. napus oil daily by gavage. After 31 days, in all groups, thoracotomy was done and lung tissue samples were taken. For pathological evaluation, microscopic slides were prepared. The eosinophil numbers in the submucosal layer and thicknesses of smooth muscle layer of bronchioles were detected.
Results: Eosinophil numbers in the submucosal layer, as well as smooth muscle layer thicknesses were significantly lower in the rat group treated with 0.75 mg/kg B. napus oil as compared with asthmatic group (p<0.01, p<0.05).
Conclusion: B. napus could be useful as adjuvant therapy in rat model of asthma. This effect was probably related to its antioxidants componentsthat reduce the levels of inflammatory mediators such as leukotrienes, IL-4, IL-5, and IL-13.- انتشار مقاله: 04-06-1391
