- چکیده انگلیسی: Herbal medicines play a significant role in global healthcare systems. In this investigation, “Monzej soda” syrup was prepared by decocting a mixture containing Lavandula angustifolia, Melissa officinalis, Echium amoenum, Cordia myxa, Glycyrrhiza glabra, Ziziphus jujuba, Foeniculum vulgare, Fumaria parviflora, Adiantum capillus-veneris and Alhagi spp. Manna. along with glycerin, sodium benzoate and potacium sorbate. Physicochemical characteristics of the syrup were examined. Moreover, an HPLC technique was designed for analysis of rosmarinic acid in the syrup using C18 column, isocratic H3PO4 0.085% as the mobile phase, flow rate of 1 ml/min in λmax 330 nm. The method was validated for selectivity, linearity, precision, accuracy, LOD and LOQ. The herbal syrup was brown color with special taste and flavor. Density, pH, viscosity, dry residue, total phenolics, rosmarinic acid content were found 1.085 g/ml, 5.56, 5.35 cP, 15.22±0.43 %, 194 mg/100ml and 47.5 mg/100 ml, respectively. The syrup was stable during laboratory stability tests. The HPLC method was valid according to selectivity, linearity (72-110 µg/ml, r2: 0.9995), intra-day and inter-day precisions (RSD%˂2), accuracy (103.38-106.47%), LOD 1.6 µg/ml and LOQ 4.9 µg/ml. The syrup is a good candidate for pharmaceutical companies after pharmacological and clinical tests.

- چکیده انگلیسی: Background and objectives: People interest in traditional medicine has increased recently; however, traditional herbal medicines should be transformed into modern forms of medicine to increase patient acceptance. In this investigation, a polyherbal traditional combination “Monzej-e-balgham” has been based on according to Iranian traditional medicine (ITM) manuscripts and its quality control evaluations have been performed. Methods: The main ingredients of the formulation including Vitis venifera L., Ficus carica L., Foeniculum vulgare Mill., Glycyrrhiza glabra L., Adiantum capillus-veneris L., Rosa damascena Herrm. and Onopordum acanthium L. were crushed, mixed and extracted with distilled water by decoction method. The mixture was used to prepare formulations of the syrup. Physicochemical, microbiological properties and rheological behavior of the syrup were studied and total phenolics content of the formulation was determined. The syrup was evaluated in accelerated stability test during 6 months. Results: The product was light brown semitransparent syrup with appropriate taste and odor. There was no cap locking and precipitation. Dry residue, sedimentation, pH, viscosity, density and total phenolics were found to be 14.82%, 0.015%, 5.40, 4.6 cP, 1.076 g/mL and 127.34 mg/100 mL, respectively. Microbial evaluations of syrupwere consistent with the WHO protocol. The rheogram of the product represented the Newtonian behavior. In the accelerated stability tests, no significant changes were observed. Total phenolics content reduced to 2.50% within 6 months in 40 ºC. Conclusion: The outcome of this research was a pharmaceutical standardized formulation from the traditional “Monzej-e-balgham” which supports the idea of drug discovery based on traditional knowledge.

- چکیده انگلیسی: Jujube (Ziziphus jujuba Mill.) is an important medicinal plant in Iran. Jujube fruit contains amino acids, polysaccharides and microelements.Ripened jujube fruits were collected at fully mature stage from two research collection of jujube at Khaf and Birjand centers in south Khorasan province, Iran, early in September, 2015.The physicochemical properties and antioxidant capacity were investigated. The antioxidant activity was determined by free radicals (DPPH) scavenging method. The results were showed a significant difference between evaluated physicochemical parameters. The acidity and pHofKhafjujube were 0.72%, 4.37 and the acidity and pH of Birjand jujube were 0.37%, 4.65; respectively. Protein, fat, fruit moisture and vitamin C in Khaf jujube (3.92%, 0.72%, 0.75% and 137.59 mg/100gr fresh weight) were more than Birjandjujube(3.24%, 0.4%, 0.06% and 125.11mg/100gr fresh weight). Birjand jujube had more reducing sugars percent (35.93 %). Seed’s moisture, 100-grain weight and the antioxidant activity of Birjand jujube (0.156%, 151.03 mg, Ec50= 0.1704 mg/ml) was higher than Khaf sample (0.11, 103.06, Ec50= 0.3135 mg/ml). The viscosities of two jujube varieties werein shear thinning fluids category. The viscosity of Khaf jujube sample was higher than Birjand jujube in different durations and shear rates. The transparency of the color of the Birjand sample (18.21%) was higher than Khaf jujube (17.67%). Overall, thebased on sensory evaluation, physicochemical and consistency properties ofKhaf jujube were more suitable than Birjand jujube while the antioxidant activityofBirjand jujube was higher than Khaf sample.

- چکیده انگلیسی: Heavy metals entrance to fish body tissues and transferring to human body systems after their consuming makes numerous undesirable effects and health problems. The aim of this study was to determine some heavy metals (lead, cadmium and mercury) in fresh fishes marketed in Khorramabad City, west of Iran. In this descriptive study, five samples of 17 fish species with high consumption were purchased randomly in 2014. Measurement of mercury, lead and cadmium was performed using atomic absorption spectrometry. All measurements were performed three times for each sample. Lead mean levels in fish samples was in the range 0.736 -1.005 ppm, cadmium range was from 0.196 to 0.015 ppm and mean content of mercury was 0.431 - 0.107 ppm. At present mean concentration of lead, mercury and cadmium in supplied fishes muscle is lower than maximum recommended levels according to WHO, EC and FDA guidelines. Based on the obtained results of this study and the importance of heavy metals in foods and their impacts on human health, continuous monitoring of heavy metals levels in foods is necessary.

- چکیده انگلیسی: Production of human proteins in Pichia pastoris has significant advantages. However, there is still need for improvement in various stages of its downstream processing like clarification and purification. In fact downstream processes are usually the most critical part of production of biotech products.
This work aimed to evaluate the effect of two steps added to the downstream processes of human growth hormone (hGH) production in Pichia pastoris. Firstly the effect of clarification, with activated carbon, on capture of hGH by ion exchange chromatography (IEC) was investigated. For this purpose, a clarification process using activated carbon was used to remove process contaminants like pigments. The clarified sample was applied to the IEC column and the recovery of hGH, following IEC, was assessed using SDS-PAGE, Bradford protein assay and area under the curve (AUC). The obtained results showed that the AUC values were 2.81 and 5.61 for the with- and without-treatment samples, respectively. Protein recovery of clarified sample with activated carbon was 541 mg in comparison with 328 mg for the sample without treatment. The yield of IEC was also improved from 50.46% to 83.23% following treatment with activated carbon.
Secondly, the effect of three concentrations of ammonium sulfate in the binding buffer on resolution of hGH upon elution on hydrophobic interaction chromatography (HIC) was investigated. Biological activity was used as the main criterion for evaluation of purified hGH using HIC. The obtained results indicated that by increasing the concentration of ammonium sulfate form 1 to 3 mol/L, resolution of hGH was improved, as the purified fraction using 3 mol/L of ammonium sulfate showed a specific activity of 3.1IU/mg.
So the results of the present study demonstrated that activated carbon is a promising candidate for efficient clarification of recombinant hGH and improving the efficiency of the capture step. Therefore, it can be considered by biotech companies as a cost-effective and sustainable clarification procedure of recombinant proteins from high cell density cultures. This study also revealed that 3% ammonium sulfate has a positive effect on the separation of hGH variants with the desired biological activity.- انتشار مقاله: 10-07-1396

- چکیده انگلیسی: Radiotherapy, a highly effective way to destroy breast cancer, causes skin adverse effects. A considerable amount of studies have been conducted to find a way to alleviate or relieve dermal adverse effects of radiation. The aim of this study was to observe the clinical effect of Elaeagnus angustifolia L. cream to treat radiotherapy-induced skin destruction in breast cancer patients.
thirty two patients suffering from different stages of radiotherapy-induced skin reactions were evaluated in a double-blind randomized study; 16 patients in the treatment group received Elaeagnus angustifolia cream, while the other 16 patients, served as control group.
Two weeks after the treatment, patients in Elaeagnus angustifolia group showed significantly lower skin reaction grade compared with placebo group. The attitude of the patients of Elaeagnus angustifolia group about dryness, itching, pain, burning, blisters and sores was significantly improved after two weeks of treatment.

- چکیده انگلیسی: Breast cancer is the most common malignancy among women, both in the developed and developing countries. Women with mutations in the BRCA1 and BRCA2 genes have an increased risk of breast and ovarian cancers. Recent studies show that short-wavelength visible light disturb the secretion of melatonin and causes circadian rhythm disruption. We have previously studied the health effects of exposure to different levels of radiofrequency electromagnetic fields (RF-EMFs) such as mobile phones, mobile base stations, mobile phone jammers, laptop computers, and radars. Moreover, over the past several years, we investigated the health effects of exposure to the short wavelength visible light in the blue region emitted from digital screens. The reduction of melatonin secretion after exposure to blue light emitted from smartphone’s screen has been reported to be associated with the negative impact of smartphone use at night on sleep. We have shown that both the blue light and RF-EMFs generated by mobile phones are linked to the disruption of the circadian rhythm in people who use their phones at night. Therefore, if women with hereditary breast cancer predispositions use their smartphones, tablets and laptops at night, disrupted circadian rhythms (suppression of melatonin caused by exposure to blue light emitted from the digital screens), amplifies the risk of breast cancer. It can be concluded that women who carry mutated BRCA1 or BRCA2, or women with family history of breast cancer should avoid using their smartphones, tablets and laptops at night. Using sunglasses with amber lenses, or smartphone applications which decrease the users’ exposure to blue light before sleep, at least to some extent, can decrease the risk of circadian rhythm disruption and breast cancer.

- چکیده انگلیسی: Objective(s):Tumor-associated antigen (TAA) subunit-based vaccines constitute promising tools for anticancer immunotherapy. However, a major limitation in the development of such vaccines is the poor immunogenicity of peptides when used alone.The aim of this study was to develop an efficient vaccine delivery system and adjuvant to enhance anti-tumor activity of a synthetic HER2/neu derived peptide (P5).
Materials and Methods: P5 peptide was encapsulated with different liposomal formulations composed of DMPC:DMPG:Chol:DOPE and loaded with monophosphoryl lipid A (MPL). All formulations were characterized for their physicochemical properties. To evaluate vaccine efficacy, BALB/c mice were first immunized with free peptide or liposomal formulations, then, inoculated with a subcutaneous injection of TUBO tumor cells. Enzyme-linked immunospot, cytotoxicity and intracellular cytokine assays, as well as tumor size and animal survival analysis, were performed to evaluate the immune responses.
Results: The results demonstrated that P5 encapsulated into liposomal formulations was not able to induce CD8 and CD4 T cells to produce IFN-γ. That is why, a potent CTL response and antitumor immunity was not induced.
Conclusion: The Lip-DOPE-P5-MPL formulation in spite of using pH-sensitive lipid to direct intracellular trafficking of peptide to MHC I presentation pathway and MPL to enhance peptide adjuvanticity was interesting. The failure in inducing anti-tumor immunity may be attributed to low uptake of anionic conventional liposomes by dendritic cells (DCs) that have negative surface charge.- انتشار مقاله: 11-03-1394
