- چکیده: مطالعه توالی زسوبات همی پلاژیک اواخر سانتونین پسین- کامپانین پسین سازند آب تلخ در برش بهادرخان (مرکز حوضه کپه داغ) ما را قادر ساخت تا براساس مجموعه روزن داران شناور و کف زی تغییرات بوم شناسی دیرینه را بررسی نماییم. سنگ شناسی برش بهادرخان متشکل از شیل آهکی، مارن آهکی، مارن و بصورت پراکنده چندین واحد سنگ آهک چاکی می باشد. وضعیت مرزهای زیرین و بالایی آن با سازندهای آبدراز و نیزارهمشیب و پیوسته است. از آنجا که روزن داران ابزار مناسبی برای بررسی های بوم شناسی دیرینه می باشند به منظور بررسی تغییرات عمق دیرینه از آنها به 3 روش استفاده شد: 1- معادلات تعیین عمق و روش Van derz van (نسبت روزن داران شناور به کف زی)(P/B، 2- مدل مورفوتایپی Leckie (1987) و 3- بررسی تغییرات پوسته روزن داران کف زی و نسبت روزن داران کف زی آگلوتینه به آهکی، همچنین با هدف بررسی میزان اکسیژن و مواد مغذی از مدل Trox نسبت روزن داران کف زی سطح زی به درون زی (Ep/In) استفاده شده است. بر اساس این شمارش های آماری 4 مرحله تغییرات عمق و شرایط محیطی را در این برش در محدوده سنی اواخرسانتونین پسین- کامپانین پسین مشاهده شد. طبق بررسیها در زمان کامپانین پیشین نسبت P/B و کف زی درون زی بالاست که این نشانه بالا بودن سطح آب و محیط یوتروفیک است که درچنین محیطی میزان اکسیژن کم و مواد مغذی بالا می باشد. سپس در اواخرکامپانین پیشین و ابتدای کامپانین میانی سطح آب پایین می آید و محیط به سمت شرایط الیگوتروفیک پیش میرود، طی کامپانین میانی و ابتدای کامپانین پسین سطح آب مجدداً افزایش می یابد و نهایتاً در زمان کامپانین پسین و با تغییر سنگ شناسی به ماسه سنگهای سازند نیزار ما شاهدکاهش در نسبت P/B و پایین آمدن سطح آب و افزایش در سطح اکسیژن وکاهش مواد مغذی می باشیم
- چکیده انگلیسی: Study of a late Late Santonian to Late Campanian hemipelagic succession from Abtalkh Formation at the Bahadorkhan section (Central Kopet-Dagh) enabled us to verify paleoecology changes based on planktonic and benthic foraminifera assemblage. Bahadorkhan section is consisted of calcareous shale, lime marl, marl, and a few dispersed chalky limestone beds. Upper and lower boundaries of Abtalkh Formation are conformable with Abderaz and Neyzar formations. Since foraminifera are proper tools for paleoecological investigations, we used them in three methods to analyze the changes of paleobathymetry: 1) Van der Zwaan’s equations of determining depth and method (the ratio of planktonic foraminifera to benthic), 2) Leckie's morphotype model, and 3) investigating the changes of benthic foraminifera and the ratio of agglutinated benthic foraminifera to the utilized calcareous benthic foraminifera. Trox model (the ratio of epifaunal to infaunal (Ep/In)) was also used with the goal of identifying oxygen level and nutrients. According to the statistical counting, four stages of changes in depth and environmental conditions in this section with age range of late Late Santonian- Late Campanian were observed. According to the analysis, in the Early Campanian, P/B ratio and infaunal benthic are high which indicates the high level of water and eutrophic environment. In such an environment, nutrient and oxygen levels are respectively high and low. Then, at end of the Early Campanian and the beginning of Middle Campanian, water level decreases, and the environment moves toward oligotrophic conditions. During Middle Campanian and the early Late Campanian, water level increases again. Finally, during Late Campanian and along with the change of lithology to Neyzar Formation sandstones, we will witness the decrease of P/B ratio, water level drop, oxygen level increase, and nutrient decrease.

- چکیده: A stratigraphic section from the Tirgan Formation was selected in Arkan village nearby Bojnourd city in order to study echinoderm fauna systematically. Many specimens of echinoderms, especially toxasteridae which is associated with brachiopods were collected and studied systematically in this paper. Microscopie slides were also prepared and studied. All slides contain benthic foraminifers and calcareous algae as well as gastropod and coral fragments. Based upon diagnostic micro and macro fossils, a Barremian-Aptian age is assigned to the Tirgan Formation which suggests Urgonien facies type indicating shallow water of open marine environment The Urgonien is named after the village with this name in the east of Tarascon, France. This facies type is characteristic of shallow-water carbonate facies which was accumulated along the Tethys northern shelf from the Barremian to the late Albian. The facies consists of hard, white-gray limestones which contains Orbitolina (foraminifers) and transitional sediments – detrital or siliceous limestones (Foury, 1968).

- چکیده انگلیسی: Background: MDM2, E-cadherin, survivin and Her2 genes are involved in the regulation of normal cell growth. However, any crucial change in their expression levels can convert a normal cell into a cancer cell. Numerous studies have identified alterations of these gene expression levels in various cancers, particularly in breast cancer. Thus, they may be used as diagnostic biomarkers. In this experiment, we aim to evaluate these gene transcripts in patients’ peripheral blood and compare the results with healthy individuals.Methods: RNA was extracted from peripheral blood cells of 52 breast cancer patients and 52 healthy volunteers. Then, cDNA was synthesized and assessed for MDM2, E-cadherin, and survivin and Her2 gene transcriptions in peripheral blood samples by quantitative real-time polymerase chain reaction.Results: There were no considerable differences in the expression of MDM2, E- cadherin, survivin and Her2 in cancer patients compared to healthy individuals. However, there were significantly correlation between E-cadherin, survivin and Her2 expression and some of the clinicopathological characteristics of patients studied. Also, survivin transcripts expression was positively correlated with Her2, E-cadherin, and MDM2 gene expressions.Conclusion: These results indicated no variations in MDM2, E-cadherin, Her2 and survivin gene expressions in patients compared to controls. We might not consider the examined biomarkers as valuable prognosis factors in primary breast cancer. However, additional research should be undertaken to assess these four genes in a larger sample size.

- چکیده انگلیسی: Statement of the Problem: One major goal of tissue engineering and regenerative medicine is to find an appropriate source of mesenchymal stem cells (MSCs) with higher differentiation ability.Purpose: In this experimental study, the osteogenic and chondrogenic differentiation ability of buccal fat pad derived MSCs (BFP-MSCs) with gingival derived cells (GDCs) were compared.Materials and Method: BFP-MSCs and GDCs were cultured enzymatically and expanded. The expanded cells were analyzed for membrane-associated markers, using flow cytometry. Then the ability of these cells to differentiate into osteocyte and chondrocyte was assessed morphologically and by mRNA expression of collagen I (COLL), BGLA and bone morphogenetic protein 2 (BMP2) using qRT-PCR.Results: Flow cytometry analysis showed that both BFP-MSCs and GDCs expressed the characteristic stem cell markers such as CD73, CD44, and CD90, whereas they did not express hematopoietic markers. Mineralized calcium deposition was observed apparently in BFP-MSCs cultured in osteogenic medium but GDCs showed fewer mineralized nodules. The mRNA expression levels of BGLA and BMP2 showed 7×105 and 733-fold more mRNA expression in BFP-MSCs treated with differentiation media compared to the control group. In chondrogenic differentiation, BFP-MSCs transformed from a spindle to a cuboidal shape while GDCs showed only a slight transformation. In addition, mRNA expression of COLL showed 282-fold higher expression in BFP-MSCs in comparison to the control group. Such significant difference in mRNA expression of BGLA, BMP2, and COLL was not observed in GDCs compared to their corresponding controls.Conclusion: Based on the present results, BFP yields a greater proportion of stem cells compared to gingiva. Therefore, this tissue can be introduced as an easily available source for the treatment of periodontal defects and other maxillofacial injuries.

- چکیده انگلیسی: In this paper, the nonlinear conjugate map is applied based on the conjugate Hasofer-Lind and Rackwitz- Fiessler (CHL-RF) method to evaluate the reliability index using the first order reliability method of the embedded nanocomposite beam, which is made of a polymer reinforced with carbon nanotubes (CNTs). The structure is simulated with the Timoshenko beam model. The Mori-Tanaka model is applied for calculating the effective material properties of the nanocomposite beam and the surrounding elastic medium is considered as spring and shear constants. The governing equations are derived based on the energy method and the Hamilton's principle. Moreover, using an analytical method, the buckling performance function of the structure is obtained. The effects of the basic random variables including the length-to-thickness ratio of the beam (L/h), the spring constant, and the shear constant of the foundation with respect to the volume fraction of CNTs are investigated based on the reliability index of the nanocomposite beam which is subjected to an axial force of 20 GPa. The results indicate that the failure probabilities of the studied nanocomposite beams are sensitive to the length-to-thickness ratio of the beam (L/h) and the spring constant of the foundation variables.

- چکیده انگلیسی: Introduction: The presence of HER-2 has been shown to be a prognostic factor in many kinds of cancers, but its role in head and neck squamous cell carcinoma (HNSCC) is not still defined. The purpose of the current study is to investigate the role of HER-2 in HNSCC and its correlation with various clinicopathologic parameters. Materials and Methods: Peripheral blood samples were obtained from 17 healthy volunteers and 69 patients with HNSCC before curative surgery. The HER-2 level was determined in each sample by sandwich ELISA. Statistical analysis was performed using an independent t-test, one-way ANOVA, and Mann-Whitney Test. Results: Mean HER-2 serum levels in patients with HNSCC compared with healthy controls was not statistically significant (3.85ng/ml vs. 3.75ng/ml; P>0.05). The mean serum level of HER-2 was higher in patients with lymph node involvement, metastasis, invasion, tumor size ≥2 cm, and stage>1, although the differences were not statistically significant (P>0.05). Discussion: Mean HER-2 serum levels in patients with tumor size T3 and higher were greater than those from patients in stage T1 and T2, a finding which may be translated into disease progression, growth and invasiveness. Conclusion: In this study the mean HER-2 serum level in patients with HNSCC was found statistically insignificant compared with the healthy control group. The role of soluble HER-2 as a tumor marker is still controversial and needs further studies to clarify its clinical relevance for early detection or screening for HNSCC.

- چکیده انگلیسی: Background: Cytokines, chemokines, and chemokine receptors regulate the proliferation and survival of tumor cells, angiogenesis, and metastasis to other organs. This network of ligands and receptors has been used in molecular targeting of cancer. Methods: We compared the mRNA expression of CXCR3, CXCL-10, CXCR4, CXCL-12, IL-4, and IL-10 in tissues of benign and malignant ovarian tumors by qRT-PCR method and evaluated serum IL-10 and CA-125 content of these patients by ELISA during one year.Results: Our result showed a trend toward a higher expression of CXCR4 in malignant ovarian tissues compared with the benign ovarian cysts (P>0.05). However, SDF-1, IP-10, IL-4, CXCR3, and IL-10 had a lower trend in mRNA expression in malignant ovarian tissues compared to the benign cyst tissues. Except for IL-4 (P=0.01) and SDF-1 (P=0.02), the data for other factors were not statistically significant. A trend toward higher concentration of IL-10 was observed in the serum of ovarian cancer patients compared to those with benign cysts; however, the difference was not significant. CA-125 concentration in the serum of ovarian cancer patients was higher than that of benign cyst patients (P=0.05). Conclusion: According to results obtained, we hypothesize that the lower expression of SDF-1 in malignant tissues may have an important role in ovarian tumor growth. However, this hypothesis requires more investigation. Higher levels of CA125 and IL-10 in the serum of patients might indicate that the combination of these biomarkers could be used for distinguishing patients with ovarian cancer from those with benign cysts.

- چکیده انگلیسی: Background: Interleukin (IL)-23 and IL-27 are two IL-12-related cytokines which their function may dramatically influence the inflammatory response to tumor development. IL-12 and IL-27 seem to have antagonistic roles with IL-23 in tumor site. In this study, IL-23 and IL-27 mRNA expressions were analyzed in peripheral blood of patients with breast cancer and healthy volunteers using quantitative real-time PCR. Methods: Peripheral blood samples were collected from 50 women with breast cancer and 50 healthy ones. The total RNA was extracted from peripheral blood after lysis with ammonium chloride and TRizol reagent and the cDNA was synthesized. The expression of IL-23 and IL-27 gene transcripts was determined with real-time polymerase chain reaction (qRT-PCR) using Syber Green PCR Master Mix. Results: It is found that IL-23 and IL-27 transcripts had significantly higher expression in peripheral blood of patients compared with the healthy controls. The ratio of IL-23 transcript expression to IL-27 was 3.4 fold lower in the studied patients compared with the normal individuals. Conclusion: It is concluded that the over expression of IL-23 and IL-27 gene transcript in peripheral blood of breast cancer patients may be an immune response against tumor development and the inflammatory response plays a critical role in tumor development via up regulating the corresponding cytokines. However, the IL-23/IL-27 ratio may play an important role in cytokine-based immunotherapy against cancer. Further research should be carried out to assess these cytokines in a larger sample size.

- چکیده انگلیسی: Background: Carcinomas of esophagus, mostly squamous cell carcinomas, occur throughout the world. There are a number of suspected genetic or environmental etiologies. Human papilloma virus (HPV) is said to be a major etiology in areas with high incidence of esophageal carcinoma, while it is hardly detectable in low incidence regions. This study was designed to evaluate the prevalence of HPV in esophageal squamous cell carcinoma (ESCC) cases diagnosed in Pathology Department, Medical School, Shiraz University of Medical Sciences.
Methods: DNA material for PCR amplification of HPV genome was extracted from formalin-fixed paraffin-embedded tissue blocks of 92 cases of ESCC, diagnosed during 20 years from 1982 to 2002. Polymerase chain reaction was performed for amplification and detection of common HPV and type specific HPV-16 and HPV-18 genomic sequences in the presence of positive control (HPV-18 and HPV positive biopsies of uterine exocervix) and additional internal controls i.e. beta-globin and cytotoxic T lymphocyte antigen 4 (CTLA4).
Result: Good amplification of positive control and internal controls was observed. However, no amplification of HPV genome was observed.
Conclusion: There is no association between HPV infection and the development of esophageal squamous cell carcinoma in the cases evaluated.- انتشار مقاله: 11-09-1392

- چکیده انگلیسی: Background: B cells can increase the expression of granzyme B in CD8+ T cells through 4-1BBL/4-1BB interaction and promote anti-tumor immunity. Objective: To investigate the expression of 4-1BBL on B cells in the breast tumor draining lymph nodes (TDLNs) and its association with disease parameters. Methods: Using Ficoll-Hypaque gradient centrifugation, mononuclear cells were isolated from axillary lymph nodes of 42 patients. Cells received 4 hours of PMA/Ionomycin stimulation, in vitro. Both unstimulated and stimulated cells were stained with anti‒CD19 and anti‒4-1BBL antibodies and subjected to flow cytometry. Results: 4-1BBL expression was detected on 2.8 ± 1.7% of unstimulated B cells, while 27.4 ± 11.9% of B cells expressed this co-stimulatory molecule following stimulation. In steady state, the percentage of 4-1BBL+ B cells was not associated with cancer characteristics. However, in patients with invasive ductal carcinoma, the percentage of 4-1BBL expressing B cells in stimulated condition had a decreasing trend in grade III, compared to grade II+I. In addition, significantly higher frequency of 4-1BBL+ B cells was seen in the TDLNs of ER+ or PR+ compared with ER‒ or PR‒ patients (p=0.021 and p=0.015, respectively). No significant associations were observed between the frequency of 4-1BBL+ B cells and the number of involved LNs, Her2 expression or disease stage. Conclusions: The frequency of 4-1BBL+ B cells significantly increased following a short time activation, and showed relative and significant associations with tumor grade and estrogen receptor status, respectively. More investigations are required to evaluate the potential of 4-1BBL+ B cells for use in immunotherapy.
