- چکیده: کلامیدیوزیس پرندگان توسط کلامیدیوفیلا پسیتاسی و با بالاترین میزان بیماری در طوطی سانان و کبوتر سانان ایجاد میشود. یک طوطی خاکستری آفریقایی کنگویی 2 ساله با علایم بی اشتهایی، افسردگی، اسهال و سختی در تنفس خفیف مورد معاینه قرار گرفت و بر اساس بررسیهای خون شناسی و بیوشیمیایی، تشخیص وجود کم خونی، لوکوسیتوز، هتروفیلی، لمفوپنی و مونوسیتوز در پرنده داده شد. با توجه به یافتههای بالینی و آزمایشگاهی، بیمار به عنوان حامل گونههای کلامیدیوفیلا تشخیص داده شد. همچنین سوآب تهیه شده از شکاف کامی و کلوآک در واکنش زنجیرهای پلیمراز تشخیصی (600 جفت باز) به لحاظ کلامیدیوفیلا مثبت بود. محصول واکنش زنجیرهای پلیمراز با واکنش زنجیرهای پلیمراز بر پایه ژن ompA و با استفاده از پرایمرهای CTU/CTL (1050 جفت باز محصول واکنش زنجیرهای پلیمراز) مشخص گردید. توالی محصول واکنش زنجیرهای پلیمراز با سکانس به دست آمده از بانک ژنی مقایسه شد. درخت فیلوژنتیکی به دست آمده شباهت 100% با ژنوتیپ B به دست آمده از مطالعات قبلی را نشان میداد. پرنده بستری شده و درمان با رژیم داکسی سایکلین به مدت 45 روز و روند نمونهبرداری هفتگی به منظور تعیین حضور DNA کلامیدیوفیلا پسیتاسی در نمونه مدفوع و شکاف کامی انجام شد. این روند تا زمانی که نمونهها پس از دو هفته منفی شدند ادامه یافت. نتایج آزمایشگاهی و رادیولوژی پس از درمان در محدوده طبیعی قرار داشتند. ژنوتیپ B به طور اولیه از خانواده کبوترسانان (فاخته و کبوتر) جدا میشود و هیچ گزارشی مبنی بر بیماری بالینی طوطی خاکستری آفریقایی به این ژنوتیپ وجود ندارد. در نتیجه با توجه به دانستههای ما، این اولین گزارش از کلامیدیوزیس ژنوتیپ B در طوطی خاکستری آفریقایی کنگویی میباشد. �حرایی گروه A را تایید کردند. میتوان چنین نتیجه گرفت که تجویز تحت مزمن دوزهای تحت کشنده آمیتراز میتواند منجر به کاهش کیفیت اسپرم گردد.
- چکیده انگلیسی: Avian chlamydiosis is caused by Chlamydiophila psittaci with the highest infection rate in parrots (Psittacidae) and pigeons (Columbiformes). A two-year-old Congo African grey parrot was examined since the bird had shown clinical signs of anorexia, depression, diarrhea, and mild dyspnea and based on biochemical and hemathological analysis the bird was diagnosed as having anemia, leukocytosis, heterophilia, lymphopenia and monocytosis. With regards to clinical and paraclinical findings, the case was diagnosed to be carrying Chlamydiophila spp. In addition, choanal cleft and cloaca swabs were positive for Chlamydiophila spp. in a diagnostic polymerase chain reaction (PCR) (600 bp amplicon). Polymerase chain reaction products were typed by ompA gene-based PCR, using CTU/CTL primers (1050 bp amplicon). The PCR product sequence was compared with the sequences obtained from GenBank. The phylogenetic tree has revealed 100% identity with genotype B obtained from previous studies. The bird was hospitalized and treated with doxycycline regimen for 45 days, with a weekly sampling process to trace the presence of C. psittaci DNA in faecal and choanal swabs, this process continued to the point where the specimens turned negative after two weeks. Laboratory and radiology results were within normal limits after the treatment. Genotype B is predominantly isolated from Columbidae and there have not been any reports regarding the clinically affected African gray parrot with this genotype. Subsequently, to the best of our knowledge, this is the first report of chlamydiosis by genotype B on Congo African grey parrot.

- چکیده: کلستریدیوم پرفرینجنس، یک بیماریزای مهم بوده که در حیوانات اهلی، بیماریهای گوارشی و در انسان مسمومیت غذایی ایجاد میکند. اسپورها طی فرآیند پخت زنده مانده و نقش مهمی در شروع احتمالی بیماری ایفا میکنند. در این مطالعه RAPD-PCR و REP-PCR برای بررسی تنوع ژنتیکی 49 ایزوله کلستریدیوم پرفرینجنس تیپ A از سه منشا مختلف استفاده شدند. نتایج RAPD-PCR، بیشترین تنوع را در بین ایزولههای مرغی نشان داد در حالی که ایزولههای انسانی کمترین تنوع را نشان دادند. آنالیز گروهبندی به دست آمده از RAPD-PCR و بر اساس فاصلههای ژنتیکی، 49 ایزوله را به 5 گروه اصلی A)، B، C، D و (E تقسیم نمود. گروههای A و C از ایزولههای گوشت مرغ، گروه B از ایزولههای مدفوع انسانی، گروه D از ایزولههای گوشت چرخ کرده، گوشت مرغ و مدفوع انسان و گروه E از ایزولههای گوشت چرخ کرده تشکیل شدند. شناسایی بیشتر این سویهها به وسیله آنالیز انگشت نگاری (GTG) 5 بر اساس توالیهای تکرار شده، تمایز بیشتری میان انواع مختلف سویهها نشان نداد. به عنوان نتیجهگیری میتوان گفت که RAPD-PCR روشی مناسب به منظور یافتن منشا آلودگی در زمینه بهداشت و صنعت مواد غذایی میباشد.
- چکیده انگلیسی: Clostridium perfringens is a serious pathogen which causes enteric diseases in domestic animals and food poisoning in humans. Spores can survive cooking processes and play an important role in the possible onset of disease. In this study, RAPD-PCR and REPPCR were used to examine the genetic diversity of 49 isolates of C. perfringens type A from three different sources. The results of RAPD-PCR revealed the most genetic diversity among poultry isolates, while human isolates showed the least genetic diversity. Cluster analysis obtained from RAPD-PCR and based on the genetic distances split the 49 strains into five distinct major clusters (A, B, C, D, and E). Cluster A and C were composed of isolates from poultry meat, cluster B was composed of isolates from human stool, cluster D was composed of isolates from minced meat, poultry meat and human stool and cluster E was composed of isolates from minced meat. Further characterization of these strains by using (GTG) 5 fingerprint repetitive sequence-based PCR analysis did not show further differentiation between various types of strains. In conclusion, RAPD-PCR method seems to be very promising for contamination source tracking in the field of food hygiene.

- چکیده: وقوع آلودگی همزمان پدیدهای رایج در گونههای گیاهی است که میتواند منجر به تغییرات در آلودگی ویروسها شود. در این مطالعه، پیشرفت بیماری در آلودگیهای مخلوط و انفرادی ویروس موزائیک خیار و ویروس موزائیک زرد لوبیا توسط اندازهگیری گسترش علائم، شدت بیماری و سطح زیر منحنی پیشرفت بیماری در لوبیا و باقلا بررسی شد. آلودگی همزمان لوبیا به ویروس موزائیک خیار و ویروس موزائیک زرد لوبیا، شدت بیماری بالاتر میزبان را باعث شد، اما تفاوت معنی داری در باقلا مشاهده نشد. روش Abbottاثر هم افزایی در لوبیا بین ویروس موزائیک خیار و ویروس موزائیک زرد لوبیا نشان داد، در حالی که بر پایه پاسخهای رشدی میزبان، برهمکنش از نوع آنتاگونیست بود. در باقلا، روش Abbott و پاسخهای رشدی میزبان، برهمکنش ویروسها را از نوع افزایشی نشان داد. به طور کلی، روش Abbott روشی مناسب برای تعیین نوع برهمکنش از نوع هم افزایی در این پاتوسیستمها بود.
- چکیده انگلیسی: The occurrence of viral co-infection is a common phenomenon in cultivated and native plant species and can alter the dynamics of virus infection. In this study, disease progress was examined in single and mixed infections of Cucumber mosaic virus (CMV) and Bean yellow mosaic virus (BYMV) by measuring the rate of symptom development, disease severity and area under disease progress curve on infected bean and broad bean. Simultaneous infection of bean to CMV and BYMV caused higher disease severity; however, no significant differences in disease severity were found on broad bean. In this study, a novel statistical approach (Abbott's approach) was used to recognize virus joint action in bean and broad bean hosts. Abbott's approach indicated synergistic effect between CMV and BYMV on bean only while the interaction was antagonistic when growth responses were considered on the same host. In broad bean plants inoculated with CMV+BYMV, CMV and BYMV, the two viruses affected disease severity and growth responses in an additive manner. Taken together, Abbott's approach was an appropriate method to determine synergistic interaction in these pathosystems.

- چکیده انگلیسی: The purpose of the present study was to investigate the Theileria and Babesia infection in sheep using polymerase chain reaction (PCR) assay in Baneh, Iran. Theileria and Babesia are apicomplexan parasites that have both vertebral and invertebrate hosts. These protozoa, which are transmitted by tick vectors, are considered to be the most important causes of parasitic diseases in Iran.The detection methods of Babesia and Theileria spp. are morphological examination, serology tests, and more recently, molecular assays, such as PCR. In this study, a total of 66 blood samples were collected and analyzed using specific primers for Theileria annulata, T. ovis, T. lestoquardi, and Babesia ovis. Two PCR methods were used, namely semi-nested PCR and competitive PCR. Based on the results of the PCR assay of 66 sheep blood samples, B. ovis, T. ovis, T. lestoquardi, and T. annulata were detected in 57 (86.4%), 28 (42.4%), 0, and 16 (24%) cases, respectively. Detection of low levels of protozoan infection with high morbidity in the tested animals shows their status as a carrier that keeps the infection in the region and extends the protozoan life cycle. Another important factor is the geographical situation of Baneh as a border city since the hemoprotozoan infection is present in this region. Moreover, piroplasmida infection was found in Iraq and other neighboring provinces. Therefore, animal husbandry in Baneh is at the risk of infection with Babesia and Theileria. The collected data in this study are useful for reaching a better understanding of the epizootiology of theileriosis and babesiosis, in order to control and prevent the diseases in this region.

- چکیده انگلیسی: The present study was designed to investigate the prevalence of Eimeria spp. in Zabol, east of Iran. The samples were examined microscopically for the presence of oocysts from February 2015 to December 2016. Out of 196 cattle, 124 (63.26%) were identified to be positive for Eimeria species. Eight species of Eimeria were found as follows: E. bovis (42.54%), E. zuernii (38.67%), E. subspherica (5.52%), E. brasiliensis (4.97%), E. ellipsoidalis (4.41%), E. cylindrical (1.65%), E. pellita (1.65%), and E. wyomingensis (0.55%). Multiple infections with two or three species were identified in 56 (45.16%) and 2 (1.61%) cases, respectively. The infection rate was observed to be higher in calves (75%) as compared to adult cattle (59.02%). The prevalence of Eimeria was greater in female cattle (67.94%). The highest prevalence and mean number of oocysts were observed in winter (69%) and autumn (209.09), respectively. All the faecal samples had an oocysts per gram of feces (OPG) less than 1000. The prevalence of infection and mean OPG were directly correlated with rainfall and relative humidity and inversely correlated with temperature. Considering the high prevalence of bovine coccidiosis in Zabol, increasing awareness of farmers and veterinarians regarding the economic importance of bovine coccidiosis should be regarded as an important factor in preventing and controlling the infection.

- چکیده انگلیسی: Ticks and tick-borne diseases, including theileriosis, constitute a major constraint to livestock production. Two species, known as Theileria lestoquardi and Theileria ovis, are suspected to contribute to ovine theileriosis in Iran. However, the epidemiological aspects of ovine theileriosis are poorly understood in this country. In a survey, designed to identify Theileria species in sheep, 52 (47.27%) out of 110 blood samples were positive, based on polymerase chain reaction (PCR) results. Among 52 positive samples, 100% (52/52) were positive for T. ovis, while T. lestoquardi was not detected in any of the samples. The 18S rRNA gene sequence of T. ovis isolated from Kurdistan, Iran has been submitted to the GenBank and can be retrieved by the accession number, KP019206. The current study presents the first report of T. ovis in Iran, using molecular identification techniques. Moreover, this study evaluated the present status of Theileria infection in the west of Iran.
